Efeitos de componentes solúveis e celulares no leite humano sobre Streptococcus mutans

Resumo: | Dental caries is a chronic, infectious and transmissible disease most common in childhood. The major microorganisms that cause caries are streptococci of the mutans group, especially Streptococcus mutans. Literature work has associated with infection of children to their caregivers and breastfeeding. On the other hand, human colostrum and milk have bioactive and soluble components that play a protective role for various microorganisms. It is possible that cellular interactions with soluble components present in the secretion exert effects during the processes of caries formation in infants. The present work verified the effects of breastfeeding on the development of caries, as well as the functional activity of phagocytes and soluble components of human colostrum for S. mutans. Males and offspring were divided into four groups - Children aged 6 to 35 months who were breastfed, children from 36 to 71 months who were breastfed, children from 6 to 36 who were being breastfed and children from 6 to 71 months who were not breastfed. Also colostrum samples were collected from 30 nursing mothers and blood samples from six males. The concentration of IgA, IgG, C3 and C4 was evaluated in the supernatant of colostrum and human serum. A pool of normal human serum and pool of supernatant was also prepared for use as the source of opsonins. In colostrum and blood cells, the release of superoxide anion, phagocytosis and microbicidal activity for S. mutans was checked. Salivation of the S. mutans colonization of mothers and their children for the different groups, the amount of IgA. Os principais microrganismos causadores da cárie são os estreptococos do grupo mutans, especialmente o Streptococcus mutans. Trabalhos na literatura têm associados à infecção de crianças a seus cuidadores e a amamentação ao peito. Por outro lado, o colostro e leite humanos, possuem componentes solúveis e celulares bioativos que desempenham um papel protetor para vários microrganismos. É possível que interações celulares com componentes solúveis presentes na secreção exerçam efeitos durante os processos de formação da cárie em lactentes. O presente trabalho verificou os efeitos do aleitamento materno sobre o desenvolvimento da cárie, bem como a atividade funcional de fagócitos e de componentes solúveis do colostro humano para S. mutans. Foram coletadas amostras salivares pareadas mães e filhos, divididos em quatro grupos- Crianças de 6 a 35 meses que foram amamentadas, crianças de 36 a 71 meses que foram amamentadas, crianças de 6 a 36 meses que estavam sendo amamentadas e crianças de 6 a 71 meses que não foram amamentadas. Também amostras de colostro foram coletadas de 30 nutrizes e amostras de sangue de 16 indivíduos do sexo masculino. Avaliou-se no sobrenadante do colostro e no soro humano a concentração de IgA, IgG, C3 e C4. Também foi preparado um “pool" de soro humano normal e "pool" do sobrenadante para utilização como fonte de opsoninas. Nas células do colostro e do sangue verificou a liberação de ânion superóxido, a fagocitose e atividade microbicida para S. mutans. Verificou na saliva a colonização da S. mutans de mães e de seus filhos para os diferentes gupos, e a concentração de IgA. Observou-se que o leite materno possui um efeito protetor frente a S.mutans e o grupo de crianças que apresentaram mais lesões de cáries foram o de mães com a maior concentração da bactéria. Houve aumento (p<0,05) na liberação de ânion superóxido e de fagocitose quando as células mononucleares (MN) do colostro e sangue foram opsonizadas com sobrenadante de colostro humano na presença de S.mutans. Quando se avaliou as células do sangue observou que aumento (p<0,05) na fagocitose e atividade microbicida independente do tipo de opsonização.
