Resumo: | Silver sulfadiazine is the drug of choice for treatment of burnings for its cicatrizing action as well as for its anti microbial action. It presents low solubility in water and it suffers almost total oxidation in 12 hours when displayed the light and humidity. Cyclodextrin has been used as an alternative to increase the solubility of drugs by the formation of inclusion complexes. This work had as objective to prepare, to characterize and to evaluate the anti microbial activity front Pseudomonas aeruginosas e Candida albicans of the inclusion complex of the Silver sulfadiazine with β-cyclodextrin. The used method of preparation was of the suspension in the molar rate 1:2 and characterized by IV and DSC. The analysis of the specters of IV demonstrated that it had reduction of the characteristic bands of the medicine suggesting that it had an interaction of the β-cyclodextrin with SDAg. The formation of the inclusion complex was confirmed by the DSC analysis, demonstrating endothermic peak in the band of temperature of 32,67°C until 42,62°C, with maximum point in 42,31°C, characterizing the temperature of fusing of the sample (Tm). The quantification of the SDAg complexed with β-cyclodextrin for High Performance/Pressure Liquid Chromatography (HPLC) varied of 11,32 until 17,66%. The results gotten for the microbiological method prove that the Silver sulfadiazine complexed with β-cyclodextrin presents activity front the Pseudomonas aeruginosa and Candida albicans, showing, this way, that the enclosed medicine a although the low concentration it was able to inhibit the growth of the tested microorganisms. |

Apresenta baixa solubilidade em água e sofre oxidação quase total em apenas 12 h quando exposta a luz e umidade. A ciclodextrina tem sido utilizada como uma alternativa para aumentar a solubilidade de fármacos através da formação de complexo de inclusão. Este trabalho teve como objetivo preparar, caracterizar e avaliar a atividade antimicrobiana frente a Pseudomonas aeruginosa e Candida albicans do complexo de inclusão da Sulfadiazina de prata com β-ciclodextrina. O método de preparação utilizado foi o da suspensão na razão molar 1:2 e caracterizado por IV e Calorimetria Exploratória Diferencial (DSC). A análise dos espectros IV demonstrou que houve diminuição das bandas características do fármaco sugerindo que houve uma interação da β-ciclodextrina com Sulfadiazina de Prata (SDAg). A formação do complexo de inclusão foi confirmada pela análise de DSC, demonstrando pico endotérmico na faixa de temperatura de 32,67 a 42,62°C, com ponto máximo em 42,31°C, caracterizando a temperatura de fusão da amostra (Tm). A quantificação da SDAg complexada com β-ciclodextrina por Cromatografia Líquida de Alta Eficiência (CLAE) variou de 11,32 a 17,66%. Os resultados obtidos pelo método microbiológico comprovam que a sulfadiazina de prata complexada com β-ciclodextrina apresentou atividade frente a bactéria Pseudomonas aeruginosa e a levedura Candida albicans, mostrando, assim, que o fármaco incluso, mesmo com baixa concentração, foi capaz de inibir o crescimento dos microrganismos testadosMade available in DSpace on 2018-06-27T18:56:00Z (GMT).
