Atividade antifúngica do óleo essencial de origanum vulgare l. (orégano) e do carvacrol, sobre cepas de cryptococcus neoformans

Resumo: Cryptococcosis is an infection caused by fungus of the genus Cryptococcus. This fungus is cosmopolitan and there are two main species, Cryptococcus neoformans and Cryptococcus gatii. The infection manifests primarily non-lung tissue and central nervous system, being highly lethal when not treated properly. Individuals with HIV or the compromised immune system are more susceptible to the development of cryptococcosis. Therapeutic agents available for the treatment of cryptococcosis are limited and toxic, besides the high incidence of physical resistance to these agents. Therefore, a search for other therapeutic alternatives for the treatment of cryptococcosis is essential. In this scenario, natural products, such as essential oils and their phytochemicals, are very promising for the development of new antifungal drugs. The objective of this study was to evaluate the antifungal activity of O. vulgare L. (oregano) and its major phytochemical, carvacrol, on Cryptococcus neoformans strains. The composition of the essential oil was analyzed by Gas Chromatography coupled to Mass Spectrometry (GC-MS). Minimum Inhibitory Concentration (MIC) and Minimum Fungicidal Concentration (CFM) were determined by broth microdilution technique. The sensitivity profile of the strains was determined by disc diffusion method, as well as the study of association of the products with antifungals. The kinetics of microbial death were also evaluated. The results of CG-MS have indicated a higher concentration of carvacrol (67.97%), p-cymene (11.67%), γ-terpinene (7.92%), thymol (7.84%) and linalool 3.44%). The MIC50, as well as MIC90, of the essential oil were 64 μg/mL. Carvacrol presented MICs of 32 μg/mL and MICs of 64 μg/mL. The CFM50 and CFM90 from the essential oil were 64 μg/mL and 256 μg/mL respectively. Carvacrol has shown CFM50 equal to 32 μg/mL and CFM90 equal to 64 μg/mL. The strains profile has shown 100% of sensitivity to amphotericin B (100 μg), 30% sensitivity to fluconazole (25 μg) and any strain has shown sensitivity to 5-fluorocytosine (10 μg). The association of essential oil and carvacrol with amphotericin B and fluconazole has not shown synergism of action. The microbial death curve has shown that the products have concentration-dependent fungicidal effect. The results of the study demonstrate that the essential oil of oregano and carvacrol have an antifungal activity, showing them as promising agents to the development of drugs capable to treat cryptococcosis.

Esse fungo é cosmopolita e tem duas espécies principais patogênicas ao homem: Cryptococcus neoformans e Cryptococcus gatii. A infecção se manifesta principalmente no tecido pulmonar e sistema nervoso central, sendo altamente letal quando não tratada adequadamente. Indivíduos portadores do HIV ou com sistema imunológico comprometido são os mais susceptíveis ao desenvolvimento da criptococose. Os agentes terapêuticos disponíveis atualmente para o tratamento da criptococose são limitados e possuem significativa toxicidade, além da alta incidência de resistência fúngica a esses agentes. Assim sendo, é imprescindível a busca por alternativas terapêuticas para o tratamento da criptococose. Nesse panorama, os produtos naturais, como óleos essenciais e seus fitoconstituintes, se mostram muito promissores para o desenvolvimento de novos medicamentos antifúngicos. Diante disto, o presente estudo teve como objetivo avaliar a atividade antifúngica do óleo essencial de O. vulgare L. (orégano) e seu fitoconstituinte majoritário, o carvacrol, sobre cepas de Cryptococcus neoformans. A composição do óleo essencial foi analisada por Cromatografia Gasosa acoplada a Espectrometria de Massas (CG-EM). A Concentração Inibitória Mínima (CIM) e Concentração Fungicida Mínima (CFM) foram determinadas pela técnica de microdiluição em caldo. O perfil de sensibilidade das cepas frente a antifúngicos licenciados, foi determinado pelo método de disco-difusão, bem como o estudo de associação dos produtos com antifúngicos. A cinética de morte microbiana também foi avaliada. Os resultados da CG-MS indicaram a presença de cinco fitoconstituintes presentes em maior quantidade: carvacrol (67,97%), p-cimeno (11,67%), ý-terpineno (7,92%), timol (7,84%) e linalol (3,44%). A CIM50, assim como a CIM90, do óleo essencial foi de 64 μg/mL. Já o carvacrol apresentou CIM50 de 32 μg/mL e CIM90 de 64 μg/mL. A CFM50 e CFM90 do óleo essencial foram 64 μg/mL e 256 μg/mL respectivamente. O carvacrol teve CFM50 de 32 μg/mL e CFM90 de 64 μg/mL. O perfil de sensibilidade das cepas mostrou que 100% das cepas foram sensíveis a anfotericina B (100 μg), 30% foram sensíveis ao fluconazol (25 μg) e nenhuma cepa apresentou sensibilidade a 5-fluorocitosina (10 μg). A associação do óleo essencial e do carvacrol com anfotericina B e fluconazol não apresentou sinergismo de ação. A curva de morte microbiana demonstrou que os produtos apresentaram efeito fungicida dependente de concentração.
