Enzimas ligninolíticas imobilizadas tem demonstrado alta eficiência na biorremediação de efluentes da indústria têxtil. Na fase I desta pesquisa, avaliou-se a descoloração do corante índigo carmim e do efluente têxtil por sete espécies de Agaricomycetes da Caatinga (Brasil). O estresse nutricional devido à limitação de nitrogênio foi realizado com três tempos de experimentação, T1 (1 dia), T2 (4 dias) e T3 (7 dias). As porcentagens de descoloração no corante e efluente foram maiores que 55% usando os fungos F1, F2, F5 e F6 por 10 dias sem esterilidade. Na fase II, os microrganismos foram cultivados nos tempos de crescimento inicial, Ci1 (10 dias) e Ci2 (25 dias), sem adição de nutrientes. Seguiu-se a indução de estresse com limitação de nitrogênio em T1 e posteriormente, com testes de descoloração do corante índigo carmim e do efluente têxtil por 28 dias. Os melhores percentuais de descoloração foram encontrados para o efluente em Ci1, e para o corante em Ci2, com F1 e F5. O F5 apresentou melhor atividade enzimática de lacase e de lignina peroxidase. O análise ecotóxico das amostras tratadas biologicamente na fase II, foram testadas frente a cistos de Artemia HIGH 5 sem adição de alimento. Os valores obtidos para KM e Vmax indicam que os extratos possuem comportamento cinético Michaelis-Menten. A lacase, lignina peroxidase e manganês peroxidase sintetizadas pelos fungos Phlebiopsis sp., Coriolopsis sp. e Phanerochaete chrysosporium, foram imobilizadas em nanopartículas de quitosana (NanoQ), apresentando porcentagens de imobilização acima de 84%. Foi calculada a porcentagem de N-acetilação usando UV-Vis. Caracterizaram-se as NanoQ (com e sem enzimas imobilizadas) por DLS, Potencial Z e MEV. A eficiência da biorremediação após o reuso demonstrou ser maior nas NanoQ com enzimas. A impressão dos dispositivos foi realizada na impressora Dimatix Materials Printer DMP-2831 (FUJIFILM Dimatix Inc.), usando trehalose no fluido de impressão para proteger a conformação estrutural das enzimas. O acetato de celulose (AC) foi o substrato flexível para a impressão. A otimização das formas de onda de impulso elétrico para a formação das gotas do fluido, foram obtidas para a impressão por demanda Drop on Demand (DoD) baseada em propriedades piezoelétricas. Os resultados mostraram que sem condições de esterilidade, o menor tempo de crescimento sem adição de nutrientes (Ci1) e o estresse por limitação de nitrogênio (T1), aumentam a biodegradação do efluente. Porém, o maior tempo de crescimento sem adição de nutrientes Ci2 favoreceu a biodegradação do corante. Todas as amostras de efluente têxtil biorremediadas na fase II, não mostraram toxicidade frente a Artemia sp. em 48 horas de experimentação. A quantidade de proteínas totais foi maior após impressão, devido ao aumento da concentração efetiva das enzimas após imobilização no biopolímero e a impressão. Os dispositivos impresos são uma inovação biotecnológica que usa impressão DoD, que permite adicionar camada sobre camada mais de um principio ativo, permitindo a liberação do material biodegradável no local albo de biorremediação.FACEPEMycotic bioremediation of effluents from industrial jeans laundries is a necessary biotechnological treatment to prevent contamination of water bodies. Immobilized ligninolytic enzymes have demonstrated high efficiency in the bioremediation of effluents from the textile industry. In phase I of this research, the discoloration of the indigo carmine dye and the textile effluent was evaluated by seven species of Agaricomycetes from Caatinga (Brazil). Nutritional stress due to nitrogen limitation was carried out with three experimentation times, T1 (1 day), T2 (4 days) and T3 (7 days). The percentage of discoloration in the dye and effluent was greater than 55% using fungi F1, F2, F5 and F6 for 10 days without sterility. In phase II, the microorganisms were grown in the initial growth times, Ci1 (10 days) and Ci2 (25 days), without adding nutrients. This was followed by stress induction with nitrogen limitation in T1 and later, with discoloration tests of the indigo carmine dye and the textile effluent for 28 days. The best percentages of discoloration were found for the effluent in Ci1, and for the dye in Ci2, with F1 and F5. F5 showed better enzyme activity of laccase and lignin peroxidase. The ecotoxic analysis of the samples biologically treated in phase II, were tested against Artemia HIGH 5 cysts without the addition of food. The values obtained for KM and Vmax indicate that the extracts have Michaelis-Menten kinetic behavior. Laccase, lignin peroxidase and manganese peroxidase synthesized by the fungi Phlebiopsis sp., Coriolopsis sp. and Phanerochaete chrysosporium, were immobilized on chitosan nanoparticles (NanoQ), with immobilization percentages above 84%. The percentage of N-acetylation was calculated using UV-Vis. NanoQ (with and without immobilized enzymes) was characterized by DLS, Potential Z and SEM. The efficiency of bioremediation after reuse proved to be higher in NanoQ with enzymes. The devices were printed on the Dimatix Materials Printer DMP-2831 (FUJIFILM Dimatix Inc.), using trehalose in the printing fluid to protect the structural conformation of the enzymes. Cellulose acetate (AC) was the flexible substrate for printing. The optimization of the electrical impulse waveforms for the formation of fluid droplets were obtained for Drop on Demand (DoD) printing based on piezoelectric properties. The results showed that without sterility conditions, the shorter growth time without the addition of nutrients (Ci1) and the stress due to nitrogen limitation (T1), increase the biodegradation of the effluent. However, the longer growth time without the addition of Ci2 nutrients favored the biodegradation of the dye. All samples of textile effluent bioremediation in phase II, did not show toxicity against Artemia sp. in 48 hours of experimentation. The amount of total proteins was higher after printing, due to the increase in the effective concentration of enzymes after immobilization in the biopolymer and printing.
