Diante disso, foi extraído um óleo essencial de folhas de P. glaziovianum (PgEO), realizada a caracterização química e avaliado o potencial hemolítico, toxicidade aguda, subaguda e genotoxicidade, bem como os efeitos antinociceptivo, anti-inflamatório aguda e crônico, antimicrobiano e cicatrizante. A caracterização química de PgEO foi realizada por Cromatografia gasosa associada a espectrometria de massas e demonstrou que o óleo era dominado por 1,8-cineole (24,29%), α-pinene (19,73%) e β-pinene (17,31%). PgEO apresentou baixa ação hemolítica (0,33% a 1,78%) para eritrócitos de camundongos. Na toxicidade oral aguda, o tratamento na concentração de 2000 mg/kg não promoveu alteração nos parâmetros fisiológicos, enquanto a concentração de 5000 mg/kg promoveu alterações hematológicas, bioquímicas e histopatológicas. A avaliação de genotoxicidade demonstrou, que a concentração de 5000 mg/kg promoveu aumento nos índices e frequência de danos de células sanguíneas nucleadas, enquanto a o tratamento de 2000 mg/kg mostrou-se seguro. No ensaio de toxicidade subaguda os animais tratados por 28 dias com PgEO (250, 500 ou 1000 mg/kg pc vo) não apresentaram alterações hematológicas, bioquímicas, urinárias, histopatológicas e temperatura retal em ambos sexos quando comparados aos controles. Já na mensuração da glicemia, os animais apresentaram uma redução significativa, além de aumento da concentração de SOD e CAT no baço e fígado de maneira dependente de dose, quando comparados aos controles. Adicionalmente, ao longo de 28 dias de tratamento as diferentes concentrações do PgEO não promoveu alterações nas atividades motora e exploratória dos camundongos. Na avaliação da atividade antinociceptiva o tratamento com PgEO (25, 50 e 100 mg/kg) reduziu as contorções abdominais induzida por ácido acético, o tempo de lambedura em ambas as fases do teste de formalina, além de promover aumento do tempo de latência em testes térmicos. Na investigação do mecanismo analgésico revelou uma ação combinada das vias opioidérgica e muscarínica. A ação anti-inflamatório (PgEO 25, 50 e 100 mg/kg), no ensaio de edema de pata o PgEO foi capaz de reduzir o edema em até 89,23%. Já no teste de peritonite o tratamento com PgEO reduziu o influxo leucócitos e neutrófilos, bem como a produção de TNF-α e IL-1β. No ensaio de granuloma o tratamento com PgEO reduziu o peso do granuloma. Na atividade antiartrítica o tratamento com PgEO (50 e 100 mg/kg) reduziu alterações osteoarticulares e edema, além dos níveis de citocinas pró-inflamatórias e o estresse oxidativo. Adicionalmente, um gel contendo PgEO (1%) apresentou atividade antimicrobiana contra bactérias gram-positivas, gram-negativas e fungos. Além disso, o gel foi eficaz na redução de feridas experimentais em camundongos com participação do sistema redox e citocinas inflamatórias. Este estudo fornece evidências que o PgEO apresenta baixa toxicidade quando administrado em alta concentração em dose única e quando administrado diariamente por até 28 dias. O tratamento com PgEO apresentou efeito no tratamento da dor e da inflamação aguda e crônica, além de apresentar atividade antimicrobiana e cicatrizante, podendo ter aplicações futuras farmacêutica.CNPqThere are no data on the therapeutic potential or safety of using Psidium glaziovianum Kiaersk, an indigenous species from Brazil that is extensively spread in the Caatinga and Atlantic Forest biomes. The hemolytic potential, acute and subacute toxicity, genotoxicity, and antinociceptive, acute and chronic anti-inflammatory, antibacterial, and healing properties of an essential oil of P. glaziovianum leaves (PgEO) were therefore assessed. Gas chromatography coupled with mass spectrometry was used to characterize the chemical composition of PgEO, and the results showed that 1,8-cineole (24.29%), α-pinene (19.73%), and β-pinene (17.31%) dominated the oil. PgEO has a negligibly hemolytic effect on mouse erythrocytes (0.33% to 1.78%). While therapy for acute oral toxicity at a dosage of 5000 mg/kg enhanced hematological, biochemical, and histopathological alterations, treatment at a dose of 2000 mg/kg had no effect on the physiological parameters. The genotoxicity assessment showed that the treatment of 2000 mg/kg was safe, however the concentration of 5000 mg/kg encouraged a rise in the rates and frequency of damage to nucleated blood cells. When compared to controls, the animals given PgEO for 28 days (250, 500, or 1000 mg/kg bw po) did not exhibit any changes in hematological, biochemical, urinary, histological, or rectal temperature. When compared to controls, the animals demonstrated a substantial decrease in blood glucose levels as well as a dose-dependent rise in the concentration of SOD and CAT in the liver and spleen. Additionally, during the course of the 28-day treatment period, none of the PgEO doses promoted modifications in the mice's motor or exploratory behaviors. Treatment with PgEO (25, 50, and 100 mg/kg) decreased abdominal writhing caused by acetic acid, licking time in both stages of the formalin test, and promoted an increase in latency time in heat tests when evaluating antinociceptive activity. A combination activity of the opioidergic and muscarinic pathways was discovered during the exploration of the analgesic mechanism. In the paw edema test, PgEO (25, 50, and 100 mg/kg) had an anti-inflammatory effect that may decrease edema by up to 89.23%. Treatment with PgEO in the peritonitis test decreased the generation of TNF-α and IL-1β as well as the inflow of neutrophils and leukocytes. PgEO therapy decreased the weight of the granuloma in the granuloma test. Treatment with PgEO (50 and 100 mg/kg) had antiarthritic effects and decreased osteoarticular alterations, edema, pro-inflammatory cytokine levels, and oxidative stress. Additionally, gram-positive and gram-negative bacteria as well as fungi were resistant to a gel containing PgEO (1%). Additionally, the gel reduced experimental wounds in mice when the redox system and inflammatory cytokines were involved. According to the results of this investigation, PgEO has minimal toxicity whether taken daily for up to 28 days and in a single dosage at a high concentration.
