Defesa: Curitiba, 23/11/2012Bibliografia: fls. 95-100Área de concentração: Insumos, medicamentos e correlatosResumo: O objetivo deste trabalho foi estudar a espécie Buddleja stachyoides Cham. & Schltdl. pertencente à família Scrophulariaceae. A partir das partes aéreas e raiz desta espécie coletadas no campus Jardim Botânico da Universidade Federal do Paraná, preparou-se extratos alcoólicos brutos e frações com diferentes polaridades, com estas amostras foram realizadas as atividades: antioxidantes, antimicrobiana, de toxicidade frente ao microcrustáceo Arthemia salina e hemolítica. Foi isolado e identificado um Fenilpropanoide denominado Verbascosídeo, a partir da fração acetato de etila das partes aéreas por RMN-1H e HPLC. O estudo de algumas atividades biológicas, com os extratos e frações, revelaram grande capacidade antioxidante da espécie pelo método da redução do complexo do fosfomolibdênio, redução do radical DPPH e teste com espécies reativas do ácido tiobarbitúrico, principalmente nas frações acetato de etila e hexano das partes aéreas e raiz, apresentando um potencial antioxidante maior do que o padrão de rutina e BHT (hidroxibutiltolueno). O composto isolado verbascosídeo presente em maior concentração na fração acetato de etila é o responsável pela atividade antioxidante em potencial. Nenhuma das frações apresentou atividade antimicrobiana após 48 h de incubação. As frações clorofórmio das partes aéreas e hexano da raiz apresentaram uma pequena atividade hemolítica, provavelmente devido à presença de saponinas. Com relação ao teste de toxicidade sobre o microcustáceo Arthemia salina, as amostras não apresentaram toxicidade. Portanto, torna-se interesse dar continuidade ao isolamento de outros compostos e realizar novas atividades biológicas e farmacológicas para a espécie Buddleja stachyoides.Abstract: The aim of this work was to study the species Buddleja stachyoides Cham. & Schltdl. belonging to the family Scrophulariaceae. From the aerial parts and roots of this species collected in the Botanical Gardens campus of the Federal University of Paraná, prepared alcoholic extracts and crude fractions with different polarities, with these samples were performed the activities: antioxidant, antimicrobial, toxicity test against the microcrustacean Arthemia salina and hemolytic. Was isolated and identified a phenylpropanoid called Verbascoside from the ethyl acetate fraction of the aerial parts by 1H-NMR and HPLC. The study of some biological activities with the extracts and fractions, showed high antioxidant capacity of the species by the method of reduction of complex phosphomolybdenum, reduction of DPPH and test with thiobarbituric acid reactive species, mainly in the fractions ethyl acetate and hexane from the aerial parts and root, an antioxidant potential higher than the standard rutin and BHT (butyl hydroxy toluene). The compound isoladed verbascoside present in higher concentration in the ethyl acetate fraction is responsible for the antioxidant activity potential. None of the fractions showed antimicrobial activity after 48 hours of incubation. The chloroform fraction of the aerial parts and hexane fraction of the roots showed little hemolytic activity, probably due to the presence of saponins. Regarding toxicity test against the microcustacean Arthemia salina, the samples showed no toxicity. Therefore, it becomes interest to continue the isolation of other compounds and hold new biological activities and pharmacological to the species Buddleja stachyoides.100f. : il.
