Defesa : Curitiba, 24/03/2017Inclui referências: p. 61-72Resumo: A reversão tumoral, processo em que uma célula tumoral perde em parte ou totalmente seu fenótipo maligno, apresenta grande potencial como nova abordagem terapêutica do câncer. A proteína TCTP esta diferencialmente expressa nas células tumorais e nas suas respectivas revertentes (que sofreram reversão tumoral), sendo altamente expressa nas células tumorais. Esta proteína caracteriza-se por ser altamente conservada, apresentar expressão preferencial em tecidos mitoticamente ativos e desempenhar várias funções em diferentes processos biológicos. Por estar relacionada com a reversão tumoral, a TCTP vem sendo considerada como um alvo terapêutico para o câncer. A sertralina, antidepressivo inibidor da recaptação de serotonina, atua sobre a proteína diminuindo seus níveis intracelulares. O importante papel de TCTP nos tumores de mama já foi descrito. Estes tumores destacam-se por ser o câncer mais comum entre as mulheres, além de ser uma doença extremamente heterogênea. O objetivo do trabalho foi melhorar o entendimento do papel biológico da TCTP em linhagens de mama, representantes de diferentes subtipos, MCF7 (hormônio dependente), PMC 42 (fenótipo de célula tronco), SKBR-3 (HER+), MDAMB- 231 e MDA-MB-436 (triplos negativos). Para tanto, foram avaliados os efeitos do tratamento com a sertralina na viabilidade, níveis intracelulares de TCTP, migração celular e clonogenicidade. Em todas as linhagens estudadas a sertralina diminui os níveis de TCTP e apresentou efeitos antitumorais. Mesmo as linhagens MDA-MB-231 e MDA-MB-436, do subtipo triplo negativo, que apresentam fenótipo agressivo e são pouco responsivas aos tratamentos disponíveis, apresentaram resultados de diminuição da migração e clonogenicidade celular. As outras linhagens (MCF7, SKBR3 e PMC 42), responderam ao tratamento com sertralina, com grande diminuição do fenótipo tumoral. Com destaque para a PMC 42, com características de célula tronco tumoral que foi bastante sensível à sertralina. Portanto, os resultados aqui obtidos evidenciam os efeitos da sertralina e da inibição da proteína TCTP sobre as diferentes linhagens de tumor de mama. Assim, sugerimos que o uso da sertralina, combinada com as estratégias terapêuticas atuais em câncer de mama, é uma possibilidade de tratamento para os pacientes. Inclusive naqueles subtipos menos responsivos clinicamente e com grande chance de resistência, como os triplo-negativos.Abstract: Tumor reversion is a process in which a tumor cell loses its malignant phenotype, partially or totally. This cellular reprogramming shows great potential as a new therapeutic approach to cancer. The TCTP protein is differentially expressed in tumor cells and their revertant counterparts (which underwent tumor reversion), being highly expressed in tumor cells. TCTP is a highly-conserved protein, presents preferential expression in mitotically active tissues and performs different functions in several biological processes. Since it is related to tumor reversion, the TCTP has been considered as a therapeutic target for cancer. Sertraline, an inhibitor of serotonin reuptake antidepressant, acts on TCTP by decreasing its intracellular levels. The important role of TCTP in breast tumors has already been described. Breast tumors stand out as the most common cancer among women, besides being an extremely heterogeneous disease. The aim of this study was to improve understanding of the biological role of TCTP in different breast cancer cell lines, representing different subtypes, MCF7 (hormone dependent), PMC-42 (stem cell phenotype), SKBR-3 (HER +), MDA- 231 and MDA-MB-436 (triple negative). The effects of treatment with sertraline on viability, intracellular levels of TCTP, cell migration and clonogenicity were evaluated. Sertraline in all studied lines decreased TCTP levels and showed antitumor effects. Even the MDA-MB-231 and MDA-MB- 436 lines from the triple negative subtype, which present an aggressive phenotype and are not responsive to the available treatments, presented results of decreased migration and cellular clonogenicity. The other lines (MCF7, SKBR3 and PMC42) responded to sertraline treatment showing a great decrease in tumoral phenotype. Especially PMC 42, showing tumor stem cell characteristics, which was very sensitive to sertraline. Therefore, the results obtained here evidence the effects of sertraline and the inhibition of the TCTP protein on the different lines of breast tumor. Thus, we suggest that the use of sertraline, with current therapeutic strategies in breast cancer, is a potential treatment for patients.
