Defesa: Curitiba, 08/05/2015Inclui referênciasÁrea de concentração : Clínica cirurgiaResumo: Introdução: O envelhecimento é um processo inevitável, complexo e que culmina na redução funcional de quase todos os órgãos e sistemas. Os rins são órgãos responsáveis pela manutenção da homeostase do organismo. Embora identifiquemos os processos fisiológicos envolvidos em seu envelhecimento, os mecanismos pelos quais estes eventos ocorrem permanecem desconhecidos. Objetivo: Avaliar a função renal e as alterações histológicas renais por meio da estereologia e morfometria em ratos submetidos ao processo natural de envelhecimento. Métodos: Estudo experimental utilizando 72 ratos Wistar, divididos em 6 grupos com 12 animais. Cada grupo foi sacrificado em uma idade diferente: 3, 6, 9, 12, 18 e 24 meses. Após a eutanásia, foi realizada nefrectomia direita, análise estereológica e morfométrica do tecido renal (volume e peso renal, densidade de volume (Vv[glom]) e densidade numérica (Nv[glom]) dos glomérulos renais e volume médio (Vol[glom]) glomerular) e também foi avaliada a função renal pela dosagem de creatinina e uréia séricas. Resultados: Houve decréscimo da função renal significativa nos ratos mais velhos. O peso renal foi menor nos animais do grupo de 3 meses (média de 1,28 ± 0,13g). O volume renal apresentou aumento progressivo durante o desenvolvimento dos ratos com os maiores valores registrados no grupo de animais com 12 meses (média de 1267,2 ± 124,0mm3) e diminuição progressiva significativa nos animais mais velhos. A Vv[glom] apresentou diminuição progressiva estatisticamente significativa entre os grupos e a Nv[glom] também diminuiu de forma significativa (média de 165,77 ± 3,13 glomérulos por mm3 nos ratos com 3 meses de idade versus média de 26,42 ± 5,07 glomérulos por mm3 no grupo de ratos com 24 meses). O volume médio dos glomérulos renais manteve-se estável nos grupos de animais mais jovens (média de 7,96 ± 0,20 ?m3). Em seguida apresentou redução significativa nos animais de até 12 meses (média de 6,45 ± 0,51 ?m3), voltando a apresentar aumento no volume médio dos glomérulos nos animais mais idosos (média de 6,88 ± 0,001 ?m3). Conclusão: A função renal, avaliada através da dosagem sérica de creatinina e ureia, mostrou ser inferior em ratos senis quando comparado a ratos jovens. A análise morfométrica e estereológica evidenciou atrofia renal, redução progressiva da densidade de volume e densidade numérica dos glomérulos renais associados ao processo de envelhecimento. Não foi encontrada hipertrofia glomerular compensatória significativa nos ratos senis. The kidneys are organs responsible for maintaining the body's homeostasis. Although identify the physiological processes involved in renal aging, the mechanisms by which these events occur remain unknown. Objective: To evaluate the renal function and the renal histological alterations through the stereology and morphometrics in rats submitted to the natural process of aging. Methods: Experimental study using 72 Wistar rats, divided in 6 groups with 12 animals. Each group was sacrificed in a different age: 3, 6, 9, 12, 18 and 24 months. After the euthanasia, it was performed right nephrectomy, stereological and morphometric analysis of the renal tissue (renal volume and weight, density of volume (Vv[glom]) and numerical density (Nv[glom]) of the renal glomeruli and average glomerular volume (Vol[glom])) and also it was evaluated the renal function for the dosage of serum creatinine and urea. Results: There was significant decrease of the renal function in the oldest rats. The renal weight was lower in the animals of the group at 3 months of age (average of 1.28 ± 0.13g). The renal volume presented gradual increase during the development of the rats with the biggest values registered in the group of animals at 12 months of age (average of 1267.2 ± 124.0mm3) and significant progressive decrease in older animals. Vv[glom] presented statistically significant gradual reduction between the groups and the Nv[glom] also decreased significantly (average of 165.77 ± 3.13 glomeruli per mm3 in 3 months old rats versus average of 26.42 ± 5.07 glomeruli per mm3 in the group at 24 months of age). The average volume of the renal glomeruli remained steady in the groups of younger animals (average of 7.96 ± 0.20 ?m3). After that, it presented significant reduction in the animals of up to 12 months of age (average of 6.45 ± 0.51 ?m3), presenting again an increase in the average volume of the glomeruli in the oldest animals (average of 6.88 ± 0.001 ?m3). Conclusion: The renal function, evaluated through the serum dosage of creatinine and urea, proved to be inferior in senile rats when compared to the young rats. The morphometric and stereological analysis evidenced renal atrophy, gradual reduction of the volume density and numerical density of the renal glomeruli associated to the aging process. It was not found significant compensatory glomerular hypertrophy in the senile rats.
