Defesa : Curitiba, 13/07/2018Inclui referências: p.74-79Resumo: Alterações nas interações do ligante do receptor ativador do fator nuclear ?B (RANKL) com seu receptor (RANK) e com seu bloqueador endogeno osteoprotegerina (OPG) estão envolvidas em varias doenças como osteoporose, artrite reumatoide e diabetes mellitus. Estudos anteriores mostraram que este sistema também esta envolvido na resposta febril induzida por lipopolissacarideo (LPS) em machos. O objetivo deste estudo foi avaliar se este sistema também esta envolvido na resposta febril em ratas e como ele se comporta frente as alterações hormonais nas femeas. Ratas falso-operadas (Sham) na fase proestro (PE) apresentaram uma temperatura corporal (Tc) mais baixa em comparação com machos, femeas na fase diestro (DE) ou ovariectomizadas (OVX) . A injeção intraperitoneal de LPS induziu uma resposta febril pronunciada em ratos machos e femeas OVX, e uma resposta significativamente menor em femeas Sham nas fases PE e DE do ciclo estral. O pre-tratamento com o antagonista de RANK, OPG, reduziu a resposta febril em machos, femeas na fase DE e OVX. Diferentemente, as femeas na fase PE não tiveram sua resposta febril alterada pela administração de OPG. A reposição de estradiol em animais OVX reduziu a febre induzida por LPS a níveis comparáveis aqueles observados em femeas em DE e PE. O tratamento com OPG tambem reduziu a resposta febril das femeas OVX a níveis similares. No entanto, a combinação de ambos os tratamentos, reposição de estradiol e tratamento com OPG, não alterou o efeito observado com qualquer tratamento individual. As femeas na fase PE apresentam uma expressao de receptores RANK no hipotalamo signifcativamente maior apos a administração de LPS enquanto que os níveis deste receptor não se alteraram significativamente em ratas em DE ou OVX, apos a administração deste pirogenio. Ainda, a reposição hormonal em ratas OVX aumentou significativamente a expressão destes receptores, resultado este similar aquele verificado nas ratas na fase PE. Esses resultados sugerem que o sistema RANK/RANKL e influenciado pelos hormonios sexuais femininos, em especial os estrogenos, desempenhando um papel importante na indução da febre apenas quando esses hormonios estão reduzidos (DE e OVX). Em femeas em PE o RANKL nao participa da resposta febril e esse fato não esta correlacionada com variações (redução) na expressão do receptor no hipotálamo. Além disso, pela primeira vez foi observado a participacao de um mediador na resposta frebril em decorrência de variações hormonais em femeas. Palavras-chave: Pirogenios Endogenos, Febre, Hormonios Sexuais.Abstract: Changes in the signaling of the Receptor Activator of Nuclear factor ?B lingant (RANKL), its receptor (RANK) and endogenous blocker osteoprotegerin (OPG) are involved in several diseases such as osteoporosis, rheumatoid arthritis and diabetes mellitus. Previous studies also showed that this system is involved in the febrile response induced by LPS in males. The aim of this study was to evaluate if this system is involved in the febrile response in rats and how this system behaves when hormonal occurs. Sham-operated female rats in proestrous (PE) showed a lower core temperature compared to males, shamoperated females in diestrous (DE) and ovariectomized (OVX) female rats. Intraperitoneal injection of LPS induced a pronounced febrile response in male and OVX female rats, and a significantly lower response in female sham-operated animals in both PE and DE phases of the hormonal cycle. Pre-treatment with the RANK antagonist, OPG, reduced the fever in males, sham-operated females in DE and OVX female rats. Differently, females in the PE phase did not have their febrile response altered by the administration of OPG. Replacement of estradiol (E2) in OVX animals reduced LPS-induced fever to levels comparable to those observed in females in DE and PE. In addition, OPG treatment also reduced the febrile response of OVX females to similar levels. However, the combination of both treatments, E2 and OPG, did not alter the effect observed with either treatment individually. Females in the PE phase exhibit significantly increased expression of RANK receptors in the hypothalamus after administration of LPS. However, in female rats in DE or OVX the levels of this receptor do not change significantly after administration of this pyrogen. Moreover, hormone replacement in OVX rats is capable of increasing this expression of receptors, a result similar to that observed in rats in the PE phase. These results suggest that the RANK / RANKL system is influenced by female hormones, especially estrogens, and plays an important role in inducing fever only when these hormones are reduced (DE and OVX). In females in PE, RANKL does not participate in the febrile response and this fact is not correlated with variations (reduction) in receptor expression in the hypothalamus. In addition, the participation of a mediator in the febrile response due to hormonal variations in females was observed for the first time.
