Resumo: Cyanobacteria are prokaryotic microorganisms, photosynthetic capable of developing blooms in eutrophic environments and producing cyanotoxins that can be neurotoxic, hepatotoxic and dermatotoxic. These can infect domestic and wild animals, except man, representing a serious risk to health. Studies show that toxic algal blooms are common in reservoirs of the Potiguar semiarid. This study aims to isolate and maintain a bank of single-species cultures of cyanobacteria Potiguar semiarid to support research in Aquatic Microbiology Laboratory - LAMAq. The samples were collected in July/2015 and March/2016, including the state of extreme drought and rain, respectively, in the Dam Engineer Armando Ribeiro Gonçalves and weir Boqueirão. The collection was carried out with the aid of a vertical trawl with 20μm mesh opening for qualitative analysis and isolation. Part of this sample was fixed in 4% formalin for subsequent identification of the species and the rest was kept in vivo for subsequent isolation. For quantitative analysis the sample was collected with bottle of Van Dorn and fixed with acetic lugol for further analysis by Utermöhl method. The evaluation of the toxicity microcystin was performed by ELISA using commercial kits. The isolation was done by seeding on a solid medium and isolating fishery in liquid medium with optical microscope. The cultures are maintained in medium ASM- 1 and ASM- 1 with double iron photoperiod of 12 hours at a temperature of 24 ± 2 ° C and light intensity of 80-1700 LUX. Eight species of cyanobacteria were identified, Cylindrspermopsis raciborskii, Geitlerinema unigranulatum, Anabaena planctonica, Anabaena circinalis, Microcystis panniformis and Microcystis aeruginosa present in Boqueirão, Coelomorom Tropicali, Planktothrix agardhii, Cylindrospermopsis raciborskii and Geitlerinema unigranulatum in Dam Engineer Armando Ribeiro Gonçalves. The species Cylindrospermopsis raciborskii and Geitlerinema unigranulatum were present in both environments. There was a predominance of Anabaena planctonica the weir Boqueirao and Planktothrix agardhii in Dam Engineer Armando Ribeiro Gonçalves. Three strains were isolated , Microcystis aeruginosa , Anabaena circinalis and planktonic Anabaena , which showed higher growth in ASM- 1 medium with bent iron concentration and are held in LAMAq. Moreover, it detected the presence of microcystin, by ELISA, the following concentrations: 3.1 μg/L for Weir Boqueirão; 3.3 μg/L for the Dam Eng Armando Ribeiro Gonçalves; 2.8 μg/L for strains of Microcystis aeruginosa ; 2.6 μg/L for strains isolated from Anabaena planctonica and 2.8 μg/L for strains isolated from Anabaena circinalis .

Essas podem contaminar animais domésticos e silvestres, além do homem, representando sérios riscos à saúde da população. Estudos revelam que florações tóxicas são frequentes em reservatórios do semiárido potiguar. O presente estudo visa isolar e manter um banco de culturas monoespecíficas de cianobactérias em amostras de açudes do semiárido potiguar para subsidiar pesquisas no Laboratório de Microbiologia Aquática – LAMAq. As amostras foram coletadas em julho/2015 e março/2016, compreendendo o estado de seca extrema e chuva, respectivamente, na Barragem Engenheiro Armando Ribeiro Gonçalves e no açude Boqueirão. A coleta foi realizada com o auxílio de uma rede de arrasto vertical com abertura de malha de 20μm para análise qualitativa e isolamento. Parte dessa amostra foi fixada em formol a 4% para posterior identificação das espécies e o restante foi mantida in vivo para posterior isolamento. Para análise quantitativa a amostra foi coletada com garrafa de Van Dorn e fixada com lugol acético para posterior análise pelo método de Utermohl. A avaliação da toxicidade por microcistina foi realizada pelo método ELISA, usando kits comerciais. O isolamento foi feito por meio de semeadura em meio sólido e isolamento por pescaria em meio líquido com auxílio de microscópio óptico. As culturas são mantidas em meio ASM-1 e ASM-1 com dobro de ferro em fotoperíodo de 12 horas a temperatura de 24±2 ºC e intensidade luminosa de 80-1700 LUX. Foram identificadas oito espécies de cianobactérias, sendo Cylindrospermopsis raciborskii, Geitlerinema unigranulatum, Anabaena planctonica, Anabaena circinalis, Microcystis panniformis e Microcystis aeruginosa presentes no Boqueirão e Coelomorom Tropicali, Planktothrix agardhii, Cylindrospermopsis raciborskii e Geitlerinema unigranulatum na Barragem Engenheiro Armando Ribeiro Gonçalves. As espécies Cylindrospermopsis raciborskii e Geitlerinema unigranulatum foram presentes nos dois ambientes. Houve predominância da Anabaena planctonica no açude Boqueirão e da Planktothrix agardhii na Barragem Engenheiro Armando Ribeiro Gonçalves. Foram isoladas três cepas, Microcystis aeruginosa, Anabaena circinalis e Anabaena planctonica, as quais apresentaram maior crescimento no meio ASM-1 com concentração dobrada de ferro e são mantidas no LAMAq. Ademais , foi detectada a presença de microcistina, pelo teste de ELISA, nas seguintes concentrações: 3,1 μg/L para o Açude Boqueirão; 3,3 μg/L para a Barragem Eng.
