Proteoma comparativo entre células-tronco mesenquimais humanas com cariótipo normal e invertido

Resumo: | Human mesenchymal stem cells (hMSCs) have been the subject of studies in the field of regenerative medicine due to its two main characteristics: self-renewal capacity and the ability to differentiate into other cell types through specific growth factors. Human umbilical cord has been an interesting source for obtaining these cells for being a disposable tissue and easily accessible. However, cytogenetic analysis made by our research group showed that one of the strands have analyzed revealed a chromosomal change constitutive. The present study had as general objective to analyze the protein profile of hMSCs expression with different karyotypes. The data from mass spectrometry were performed using Scaffold software, with it was possible to discover the amount of protein present in cells with distinct karyotypes, beyond the comparative expression of proteins between cells. In addition, were used other bioinformatic tools for discover biological processes and molecular functions related to these proteins, besides the construction of the protein -protein interaction network to discover the main proteins of different protein groups. It was possible to verify that the cells that have an inversion underwent alteration that led to a production of proteins associated to the cellular stress, in addition, there are possibilities for cells to turn into cancer cells. These analyzes corroborate the importance of carrying out studies related to the genetic instability of hMSCs, having as parameter the protein expression profile of these cells. |

These analyzes corroborate the importance of carrying out studies related to the genetic instability of hMSCs, having as parameter the protein expression profile of these cells.s Células-tronco mesenquimais humanas (CTMH) têm sido alvo de estudos na área da medicina regenerativa devido à duas características principais, sua capacidade de autorrenovação e de diferenciação em outros tipos celulares mediante fatores de crescimento específicos. O cordão umbilical humano tem sido uma fonte interessante de obtenção deste tipo celular por ser um tecido descartável e de fácil acesso, todavia, análises citogenéticas realizadas por nosso grupo de pesquisa revelaram que um dos cordões analisados possuem uma alteração cromossômica constitutiva. O presente trabalho teve como objetivo geral analisar o pe rfil proteico de expressão de CTMH com diferentes cariótipos. Os dados brutos oriundos da espectrometria de massas previamente realizada foram extraídos utilizando software Scaffold, com ele foi possível descobrir a quantidade de proteínas presentes nas células com os cariótipos distintos, além da expressão comparativa de proteínas entre as células. Além disso, foram utilizadas ferramentas de bioinformática para se descobrir os processos biológicos e as funções moleculares relacionados a essas proteínas, além da construção da rede de interação proteína-proteína para se descobrir as proteínas principais de grupos de proteínas distintos. Foi possível constatar que as células que possuem a inversão sofreram alterações que levaram a produção de proteínas relacionadas ao estresse celular, além disso, há possibilidade dessas células se transformarem em células cancerígenas.
