The desire for whiter teeth has established the tooth whitening as one of the most popular cosmetic procedure used in clinical practice. Despite its popularity, the bleaching has some side effects, and the main of them is tooth sensitivity. A clinical protocol that can reduce tooth sensitivity levels has been sought. It has been discussed whether prior oral administration of anti-inflammatory drugs is effective in reducing sensitivity during tooth whitening. Some studies have concluded that the use of these drugs reduced the prevalence of sensitivity during the bleaching procedure. There are some analgesics and antiinflammatories which allow direct application to the tooth surface as the bleaching agent, but is not known whether these drugs have permeability to tooth structure. OBJECTIVE: To evaluate the degree of transdentinal permeability of drugs (ibuprofen and codeine) applied topically on tooth enamel. MATERIALS AND METHODS: A total of 10 humans teeth were cut and placed in a permeability device developed for this research. The drugs were applied on the teeth according to the certain times and groups (G1, G2, G3, G4, G5 and G6), and the analysis was done by HPLC. RESULTS: It was not detected the presence of drugs in any of the analyzed times. CONCLUSIONS: There was no permeability of drugs to enamel and dentin.
