Resumo: Overweight in women of childbearing age and during pregnancy has consequences of fetal and adult life of offspring. This fact has been demonstrated through studies with experimental models of maternal obesity. Previous data from our group showed that maternal consumption of a high fat diet (HF) induced, at weaning, increase body mass of offspring, adiposity and hyperleptinemia. Cardiac function at 30 days of life showed lower systolic function (decreased ejection fraction and increased final systolic volume). These data demonstrate that the maternal consumption of the high fat diet promotes early changes in the cardiac function of rats regardless of sex of the offspring. This study aimed to investigate if maternal consumption of HF diet can alters cardiac structure or metabolism in male and female at weaning. For this purpose, Wistar female rats consumed control (9% lipids, C group) or HF diet (29% lipids, HF group) for 7 consecutive weeks before mating and during pregnancy and lactation. At weaning, 21 days-old, puppies were weighted and euthanized by cardiac puncture. Heart and white adipose tissues (retroperitoneal, inguinal and perigonadal) were weighted. Histological (Hematoxilin-Eosin) and biochemical (TLC and HPLC) analysis were performed using LV samples. The ventricular expression of Ir? and PI3K receptors were analysed by Western blotting (Protocol IBCCF- 114, UFRJ). HF offspring had higher body weight, adiposity and cardiac hypertrophy. Among the neutral lipids measured (monoacylglycerol, cholesterol, esterified cholesterol, triacylglycerol, fatty acids and phospholipids) the cholesterol was decreased in male HF offspring (13%), whereas the esterified cholesterol was decreased in male (52%) and female (29%) HF offspring. The triacylglycerol content was increased in both sexes of the HF offspring (52% in male and 48% in female). The quantification of the organic acids (oxaloacetic, lactic and succinic acid) did not show any alteration between C and HF offspring. There were no difference in the cardiac expression of the Ir? and PI3K receptors. These results suggest that the maternal consumption of high fat diet changes the morphology of LV cardiomyocytes and the cardiac metabolism in rats regardless of the sex of the offspring, which can explain the decreased systolic function observed in juvenile rats

Dados anteriores do nosso grupo demonstraram que o consumo materno de dieta hiperlip?dica (DH) induziu, ao desmame, aumento de massa corporal da prole, adiposidade e hiperleptinemia. A fun??o card?aca aos 30 dias de vida demonstrou preju?zo na fun??o sist?lica (diminui??o da fra??o de eje??o e aumento do volume sist?lico final). Estes resultados demonstram que o consumo materno de dieta hiperlip?dica promove altera??es precoces na fun??o card?aca de ratos independentemente do sexo do filhote. Este estudo teve como objetivo verificar se o consumo materno de dieta hiperlip?dica pode alterar a morfologia e o metabolismo dos cardiomi?citos de animais machos e f?meas da prole ao desmame. Para isso, ratas Wistar consumiram dieta controle (9% lip?deos, grupo C) ou hiperlip?dica (29% lip?deos, grupo DH) durante 7 semanas antes do acasalamento, e durante a gesta??o e lacta??o. Ao desmame, 21 dias de vida, as proles foram pesadas e eutanasiadas por pun??o card?aca. O cora??o e os tecidos adiposos branco (retroperitoneal, inguinal e perigonadal) foram pesados. An?lises histol?gicas (cortes corados com HE) e bioqu?micas (TLC e HPLC) foram realizadas utilizando amostras do ventr?culo esquerdo (VE) e a express?o card?aca dos receptores Ir? e PI3K foi avaliada atrav?s da t?cnica de Western Blotting (Protocolo IBCCF- 114, UFRJ). A prole DH apresentou maior peso corporal e hipertrofia card?aca. Dentre os lip?deos neutros mensurados (monoacilglicerol, colesterol, colesterol esterificado, triacilglicerol, ?cidos graxos e fosfolip?deos) o colesterol apresentou-se diminu?do nos machos (13%) da prole DH, enquanto o colesterol esterificado estava diminu?do nos machos (52%) e f?meas (29%) da prole DH. O conte?do de triacilglicerol estava aumentado em ambos os sexos da prole DH (52% nos machos e 48% nas f?meas). A quantifica??o de ?cidos org?nicos (?cido oxaloac?tico, l?tico e succ?nico) n?o demonstrou nenhuma varia??o entre as prole C e DH. N?o houve altera??o na express?o card?aca dos recptores Ir? e PI3K. description Overweight in women of childbearing age and during pregnancy has consequences of fetal and adult life of offspring. This fact has been demonstrated through studies with experimental models of maternal obesity. Previous data from our group showed that maternal consumption of a high fat diet (HF) induced, at weaning, increase body mass of offspring, adiposity and hyperleptinemia. Cardiac function at 30 days of life showed lower systolic function (decreased ejection fraction and increased final systolic volume). These data demonstrate that the maternal consumption of the high fat diet promotes early changes in the cardiac function of rats regardless of sex of the offspring. This study aimed to investigate if maternal consumption of HF diet can alters cardiac structure or metabolism in male and female at weaning. For this purpose, Wistar female rats consumed control (9% lipids, C group) or HF diet (29% lipids, HF group) for 7 consecutive weeks before mating and during pregnancy and lactation. At weaning, 21 days-old, puppies were weighted and euthanized by cardiac puncture. Heart and white adipose tissues (retroperitoneal, inguinal and perigonadal) were weighted. Histological (Hematoxilin-Eosin) and biochemical (TLC and HPLC) analysis were performed using LV samples. The ventricular expression of Ir? and PI3K receptors were analysed by Western blotting (Protocol IBCCF- 114, UFRJ). HF offspring had higher body weight, adiposity and cardiac hypertrophy. Among the neutral lipids measured (monoacylglycerol, cholesterol, esterified cholesterol, triacylglycerol, fatty acids and phospholipids) the cholesterol was decreased in male HF offspring (13%), whereas the esterified cholesterol was decreased in male (52%) and female (29%) HF offspring. The triacylglycerol content was increased in both sexes of the HF offspring (52% in male and 48% in female). The quantification of the organic acids (oxaloacetic, lactic and succinic acid) did not show any alteration between C and HF offspring. There were no difference in the cardiac expression of the Ir? and PI3K receptors. These results suggest that the maternal consumption of high fat diet changes the morphology of LV cardiomyocytes and the cardiac metabolism in rats regardless of the sex of the offspring, which can explain the decreased systolic function observed in juvenile rats
