Desenvolvimento de protocolo para coleta de amostras dos n?cleos da base do c?rebro de bovinos e equinos, para exame histopatol?gico

Resumo: Production animals affected by diseases that present with nervous symptoms should be necropsied and fragments of the nervous system collected for histopathological examination. With regard to brain collection, it is important to standardize procedures and at the same time simplify protocols. The present work is justified by the importance of standardizing brain cleavages in the neuropathology routine, maximizing the representation of subcortical structures, with economy of histological processing. The objective was to describe brain cleavages for two sets of nervous tissue fragments obtained in different sectional planes, standardizing the location of the constant nuclei of the striatum, obtained through the transverse section of the telencephalon at the optic chiasm level; and another set, components of the limbic system, obtained through the cut in the dorsal plane of the piriform lobe.Twenty- nine equine brains and 11 bovine brains from a convenience sample from a laboratory collection were used. The brains were weighed with formalin and measured in terms of length and total width, height of the right and left hemispheres at the level of the piriform lobe, and length of the right and left hemispheres. After the measurements, cleavages were performed and the fragments were processed according to the histological routine. The slides were illustrative for the microscopic identification of the proposed structures. The average weight of the brains, whose formaldehyde concentration was 20%, was 400.0 ? 85.0 grams for cattle and 380.0 ? 69.0 grams for horses. For cattle, the mean values obtained from the measurements were: total brain length (12.00 cm), right hemisphere length (9.60 cm), left hemisphere length (9.50 cm), right hemisphere height ( 6.20 cm), left hemisphere height (6.20 cm) and total brain width (9.10 cm). For horses, the mean values were: total brain length (12.00 cm), right hemisphere length (9.60 cm), left hemisphere length (9.60 cm), right hemisphere height (6.00 cm ), left hemisphere height (5.90 cm) and total brain width (8.52 cm). The cross section at the optic chiasm level made it possible to frame the structures of the striatum in all samples in the vertical position of the cassette, with the exception of the cloister in the fragments obtained from larger brains, in which the dorsal orientation accommodated the cloister. For the cerebral amygdala, hippocampus and thalamus, the dorsal cut incident on the caudal part of the lateral rhinal sulcus made it possible to locate these structures in all samples. Submacroscopic images of the blades and the correlations with the cleaved pieces helped to confirm the identifications of the cores considered in this work. It is concluded that the cuts adopted in this work were adequate for the cleavage aiming at the location of the proposed nuclei, regardless of the weight and metric variables of the brain considered in this study.

No que se refere ? coleta de enc?falo, ? importante a padroniza??o de procedimentos e ao mesmo tempo a simplifica??o de protocolos. O presente trabalho justifica- se na import?ncia em padronizar clivagens do c?rebro na rotina em neuropatologia, maximizando a representatividade de estruturas subcorticais, com economia de processamento histol?gico. O objetivo foi descrever clivagens do c?rebro para dois conjuntos de fragmentos de tecido nervoso obtidos em diferentes planos de sec??o, padronizando a localiza??o dos n?cleos constantes do corpo estriado, obtido atrav?s do corte transversal do telenc?falo ao n?vel do quiasma ?ptico; e outro conjunto, componentes do sistema l?mbico, obtido atrav?s do corte no plano dorsal do lobo piriforme. Foram utilizados 29 enc?falos de equinos e 11 enc?falos de bovinos constantes de amostra de conveni?ncia de acervo laboratorial. Os enc?falos foram pesados formolizados e mensurados em seu comprimento e largura total, altura dos hemisf?rios direito e esquerdo ao n?vel do lobo piriforme e comprimento dos hemisf?rios direito e esquerdo. Ap?s as mensura??es, foram realizadas as clivagens e os fragmentos foram processados de acordo com a rotina histol?gica. As l?minas foram ilustrativas para a identifica??o microsc?pica das estruturas propostas. O peso m?dio dos enc?falos, cujo a concentra??o do formol era a 20%, foi de 400,0 ? 85,0 gramas para bovinos e 380,0 ? 69,0 gramas para equinos. Para os bovinos, os valores m?dios obtidos das mensura??es foram: comprimento total do enc?falo (12,00 cm), comprimento do hemisf?rio direito (9,60 cm), comprimento do hemisf?rio esquerdo (9,50 cm), altura do hemisf?rio direito (6,20 cm), altura do hemisf?rio esquerdo (6,20 cm) e largura total do c?rebro (9,10 cm). Para os equinos os valores m?dios foram: comprimento total do enc?falo (12,00 cm), comprimento do hemisf?rio direito (9,60 cm), comprimento do hemisf?rio esquerdo (9,60 cm), altura do hemisf?rio direito (6,00 cm), altura do hemisf?rio esquerdo (5,90 cm) e largura total do c?rebro (8,52 cm). O corte transversal ao n?vel do quiasma ?ptico possibilitou o enquadramento das estruturas do corpo estriado em todas as amostras na posi??o vertical do cassete, ? exce??o do claustro nos fragmentos obtidos de c?rebros de maior dimens?o, em que a orienta??o dorsal acomodou o claustro. Para a am?gdala cerebral, hipocampo e t?lamo, o corte dorsal incidindo a parte caudal do sulco rinal lateral possibilitou a localiza??o das referidas estruturas em todas as amostras. Imagens submacrosc?picas das l?minas e as correla??es com as pe?as clivadas auxiliaram na confirma??o das identifica??es dos n?cleos considerados neste trabalho. description Production animals affected by diseases that present with nervous symptoms should be necropsied and fragments of the nervous system collected for histopathological examination. With regard to brain collection, it is important to standardize procedures and at the same time simplify protocols. The present work is justified by the importance of standardizing brain cleavages in the neuropathology routine, maximizing the representation of subcortical structures, with economy of histological processing. The objective was to describe brain cleavages for two sets of nervous tissue fragments obtained in different sectional planes, standardizing the location of the constant nuclei of the striatum, obtained through the transverse section of the telencephalon at the optic chiasm level; and another set, components of the limbic system, obtained through the cut in the dorsal plane of the piriform lobe.Twenty- nine equine brains and 11 bovine brains from a convenience sample from a laboratory collection were used. The brains were weighed with formalin and measured in terms of length and total width, height of the right and left hemispheres at the level of the piriform lobe, and length of the right and left hemispheres. After the measurements, cleavages were performed and the fragments were processed according to the histological routine. The slides were illustrative for the microscopic identification of the proposed structures. The average weight of the brains, whose formaldehyde concentration was 20%, was 400.0 ? 85.0 grams for cattle and 380.0 ? 69.0 grams for horses. For cattle, the mean values obtained from the measurements were: total brain length (12.00 cm), right hemisphere length (9.60 cm), left hemisphere length (9.50 cm), right hemisphere height ( 6.20 cm), left hemisphere height (6.20 cm) and total brain width (9.10 cm). For horses, the mean values were: total brain length (12.00 cm), right hemisphere length (9.60 cm), left hemisphere length (9.60 cm), right hemisphere height (6.00 cm ), left hemisphere height (5.90 cm) and total brain width (8.52 cm). The cross section at the optic chiasm level made it possible to frame the structures of the striatum in all samples in the vertical position of the cassette, with the exception of the cloister in the fragments obtained from larger brains, in which the dorsal orientation accommodated the cloister. For the cerebral amygdala, hippocampus and thalamus, the dorsal cut incident on the caudal part of the lateral rhinal sulcus made it possible to locate these structures in all samples. Submacroscopic images of the blades and the correlations with the cleaved pieces helped to confirm the identifications of the cores considered in this work. It is concluded that the cuts adopted in this work were adequate for the cleavage aiming at the location of the proposed nuclei, regardless of the weight and metric variables of the brain considered in this study.
