Diversidade de fungos micorr?zicos arbusculares e de bact?rias fixadoras de nitrog?nio simbi?ticas em ?reas de minera??o de bauxita com diferentes idades de revegeta??o

Resumo: To the adequate management and use of different resources of a certain ecosystem, knowledge of the ecological processes is of fundamental importance. In this context, biodiversity, nutrients cycling and energy flux are shown to have important regulatory roles. Recent data showed that there is a relationship between the plants diversity and biomass productivity and that in addition to soil fertility, temperature, latitude, precipitation and herbivory, the interaction among species from the different functional groups and the microbial diversity participates in the determination of vegetal diversity. Currently, natural ecosystems are known to have interconnected components located above and below-ground and regulatory effects are known to be established between the populations of each component. The aims of this study was to improve protocols of DNA extraction and to evaluate the diversity of arbuscular mycorrhizal fungi and nitrogen-fixation bacteria in areas with different ages of revegetation after bauxite mining, as well as to relate the diversity of these microorganisms with the plant diversity. It was observed that addition of the PVP, PVPP and activated charcoal improved both quality and yield of DNA extracts. Also, high rhizobial diversity was observed in the different areas, and this was highest in the degraded areas that were revegetated longer ago. Glomus macrocarpum was the most abundant arbuscular mycorrhizal fungi species among the 13 species found. The results showed that the revegetation strategies adopted in these areas are enabling the establishment of a community of arbuscular mycorrhizal fungi in the soil. However, the richness level and the spore numbers continue to be low, mainly in the degraded areas revegetated after the year 2000. No correlation was observed with respect to the richness values and Shannon-Wiener index between plants community and arbuscular mycorrhizal fungi. The relationship between above and below-ground diversity was dependent of plant community composition.

Dentro desse contexto, a biodiversidade, a ciclagem de nutrientes e o fluxo de energia apresentam pap?is regulat?rios importantes. Resultados recentes mostram que h? rela??o entre diversidade de plantas e produtividade de biomassa nos ecossistemas. Al?m de fertilidade do solo, temperatura, latitude, precipita??o, herbivoria, intera??es competitivas, separa??o espacial e temporal da utiliza??o de recursos, a intera??o entre esp?cies pertencentes a diferentes grupos funcionais e a diversidade de microrganismos participam na determina??o da diversidade vegetal. Atualmente, ecossistemas naturais s?o conhecidos por possu?rem componentes interligados localizados acima e abaixo do solo, sendo que efeitos regulat?rios s?o estabelecidos entre as popula??es de cada componente. Neste trabalho buscou-se aprimorar protocolos de extra??o de DNA e avaliar a diversidade de fungos micorr?zicos arbusculares e de bact?rias simbi?ticas fixadoras de nitrog?nio em ?reas com diferentes idades de revegeta??o ap?s a explora??o de bauxita, bem como, relacionar a diversidade destes microrganismos com a diversidade de plantas. Foi observado que a adi??o de polivinilpolipirrolidona, polivinilpirrolidona e de carv?o ativado proporcionou obten??o de produtos de extra??o de DNA de qualidade suficiente para realiza??o de rea??es de amplifica??o de DNA. Observou-se, tamb?m, elevada diversidade de bact?rias simbi?ticas fixadoras de nitrog?nio nas diferentes ?reas, e esta foi mais elevada em ?reas revegetadas a mais tempo. Glomus macrocarpum foi a esp?cie de fungo micorr?zico arbuscular predominante entre as 13 esp?cies encontradas. Os resultados mostram que as pr?ticas de revegeta??o adotadas nestas ?reas est?o permitindo o estabelecimento de comunidade de fungos micorr?zicos arbusculares no solo. Entretanto, o n?vel de riqueza e o n?mero de esporos ainda est?o baixos, principalmente nas ?reas revegetadas ap?s o ano de 2000. N?o foi observada correla??o para os valores do ?ndice de Shannon entre as comunidades de plantas e de fungos micorr?zicos arbusculares.
