Resumo: A study carried out of chemical and spectroscopic characterization in humic acids extracted from soils (AH1-Red-Yellow Latosols (Oxisols); AH2-Haplic Chernosol and AH3- Haplic Organosols (Histosols)) (Experiment 1) to verify the difference between these humic acids. Subsequently an experiment was performed (Experiment 2), to evaluate the parameters of growth of transformed roots of sweet basil (Ocimum basilicum L.) treated with the humic acids extracted from different soils and with sodium nitroprusside (SNP) as a donor of nitric oxide (NO) at doses of 0, 1, 2, 3, 4 and 5 mM Carbon L-1 and 0, 0.1, 10, 50, 100 and 200 ?M SNP L-1 medium, respectively, with assessments conducted in the assembly, 5 and 15 days of experiment. It was found that the dose of carbon 3mM L-1 for the humic acids and the dose of 100 ?M SNP L-1 (nitric oxide) were the doses provided the best responses in growth parameters. With the results of this study were mounted two more tests, the first (Experiment 3), to evaluate the nitrogen metabolism, the responses of proton pumps, catalase activity and NO histolocalization and reactive oxygen species (ROS) in vivo in roots transformed basil, collected at 5 and 15 days after installation of the experiment. The second trial (Experiment 4) was to evaluate the NO and ROS histolocalization in vivo, and the activity of important enzymes involved in the growth and nutrient uptake, and this essay collected six hours after treatment. The roots were treated with two concentrations of nitrate, a low dosage of N-NO3- (0.5 mM) and high (5.0 mM) were added in which the bioactive substances (AH3-3mM Carbon L-1 and 100 ?M SNP L-1). In all parameters evaluated root growth, the treatments with humic acid and nitric oxide showed higher growth compared to control. Moreover, the evaluation of the parameters of nitrogen metabolism in the two tested doses of N-NO3- bioactive substances provided with increased enzyme activities and nutrient content, compared to controls, especially at the lower dose of N-NO3 -. It is a consensus among scientists that the bioactivity of humic acid is mainly related to the presence of molecules similar to auxin and possibly other hormones. The generation of nitric oxide in plants is related to root growth and this growth can be provided by endogenous production of auxin by application of synthetic auxin analogs or application of humic acids. Thus the signaling of nitric oxide may be related to the bioactivity of humic acids. These effects were evidenced by the increase of root growth, uptake N-NO3 - of -activation of H+-ATPase plasma membrane in transformed roots of basil. Similarly, we observed increased activity of catalase, an enzyme related to root growth, by providing the dismutation of hydrogen peroxide produced in the processes of cell differentiation and expansion as well as to detect ROS in vivo, increased in treatments with bioactive substances. With the results in this thesis can give the use of humic acids may serve as a biotechnological tool in the production of biofertilizers fertilizers, for growing plants in soils of low fertility and also as components in the production of inoculants of beneficial microorganisms.

De acordo com o espectro de IV TF, n?o houve diferen?a significativa entre os ?cidos h?micos extra?dos dos tr?s solos estudados. O ?ndice de condensa??o e de h?drofobicidade que express?o o grau de humifica??o dos solos demonstraram que os ?cidos h?micos provenientes dos solos estudados s?o bastante est?veis e resistentes a degrada??o oxidativa. Posteriormente foi realizado um experimento (Experimento 2), com o objetivo de avaliar os par?metros de crescimentos de ra?zes transformadas de manjeric?o (Ocimum basilicum L.) tratadas com os ?cidos h?micos extra?dos dos diferentes solos estudados e tamb?m com nitroprussiato de s?dio (SNP), como doador de ?xido n?trico (NO), nas doses de 0, 1, 2, 3, 4 e 5 mM de Carbono L-1 e 0, 0,1, 10, 50, 100 e 200 ?M de SNP L-1 de meio de cultura, respectivamente, com avalia??es realizadas na montagem, 5 e 15 dias de experimento. Verificou-se que a dose de 3mM de Carbono L-1 para os ?cidos h?micos e a dose de 100 ?M de SNP L-1 (?xido n?trico) foram as doses que proporcionaram as melhores respostas nos par?metros de crescimento. Com os resultados obtidos nesse experimento foram montados mais dois ensaios, o primeiro (Experimento 3), com o objetivo de avaliar o metabolismo do nitrog?nio, as respostas das bombas de pr?tons, atividade da enzima catalase e a histolocaliza??o de NO e de esp?cies reativas de oxig?nio (ROS) in vivo nas ra?zes transformadas de manjeric?o, coletadas aos 5 e 15 dias ap?s a montagem do experimento. O segundo ensaio (Experimento 4) teve o objetivo de avaliar a histolocaliza??o de NO e de ROS in vivo, e a atividade de enzimas importantes envolvidas no crescimento e absor??o de nutrientes, sendo este ensaio coletado ap?s seis horas de tratamento. As ra?zes foram tratadas com duas concentra??es de nitrato, uma dose baixa de N-NO3 - (0,5mM) e outra alta (5,0mM), nas quais foram adicionadas as subst?ncias bioativas (3mM de Carbono L-1 de AH3 e 100 ?M de SNP L-1). Para todos os par?metros de crescimento radicular avaliados, os tratamentos com ?cidos h?micos e ?xido n?trico apresentaram maiores sobre o crescimento radicular, em rela??o ao controle. Al?m disso, conforme a avalia??o dos par?metros do metabolismo do nitrog?nio nas duas doses testadas de N-NO3-, as subst?ncias bioativas proporcionaram aumento de atividades enzim?ticas e conte?do de nutrientes, em rela??o aos controles, principalmente na dose mais baixa de N-NO3 -. J? ? consenso na comunidade cient?fica que a bioatividade dos ?cidos h?micos est? relacionada principalmente com a presen?a de mol?culas an?logas a auxina e possivelmente com outros fitorm?nios. A gera??o de ?xido n?trico em plantas est? relacionada ao crescimento de ra?zes, e este crescimento pode ser proporcionado pela produ??o end?gena de auxina, pela aplica??o de an?logos sint?ticos aux?nicos ou aplica??o de ?cidos h?micos. Dessa forma a sinaliza??o de ?xido n?trico pode estar relacionada ? bioatividade dos ?cidos h?micos. Estes efeitos foram evidenciados, pelo aumento do crescimento radicular, absor??o de N-NO3 - ativa??o de da H+-ATPase de membrana plasm?tica em ra?zes transformadas de manjeric?o. Do mesmo modo, foi observado o aumento da atividade da catalase, enzima relacionada ao crescimento de raiz, por proporcionar a dismuta??o do per?xido de hidrog?nio, produzidos nos processos de diferencia??o e expans?o celular. Do mesmo modo, foi observado aumento na detec??o de ROS in vivo, com os tratamentos com as substancias bioativas.
