Summary: | Zearalenone (ZEA) is a mycotoxin that causes a lot of problems in the reproductive tract, being related to the hyperestrogenism in gilts. Preventive measures to prevent fungal growth are not always effective, the use of anti- mycotoxin additives (AMA), which are added to food products, capable of adsorbing, or neutralizing biotransform mycotoxins, preventing the development of mycotoxicosis are required. The cell wall of Saccharomyces cerevisiae has become the focus of numerous studies and is used as AMA, because it has high binding capacity, and adsorption of mycotoxins. The objective of this study was to analyze the effects of zearalenone on serum biochemical parameters and histopathological and morphometric parameters of the reproductive tract of pre-pubertal male and female piglets experimentally poisoned with different doses of this mycotoxin and to evaluate the effectiveness of the anti-mycotoxin additive made of yeast cell wall in the prevention of mycotoxicosis. The trial period had a total duration of 21 days. Eight pre-pubertal male piglets were used, whose diets presented four levels of ZEA (0.0, 0.25, 0.6 and 2.0 mg kg -1 or ppm). Histomorphometric analyzes of the testis length and width were performed. Histological measurements were also performed on the testicles and histopathological evaluations of the right and left testicles, right and left (head, body and tail) epididymis, right and left deferent duct, prostate, bulbourethral gland and seminal vesicle, and biochemistry serum analysis. In addition, 48 prepubertal female piglets were used, whose diets presented two levels of inclusion of anti-mycotoxin additive (AMA) (0.0 and 0.2%) and three levels of ZEA (0.25; 0.6 and 2.0 mg kg -1 or ppm). And still histomorphometric analyzes of the proximal vagina, distal vagina, proximal uterus, medial uterus, distal uterus, right and left ovary, and histopathological findings of the liver, kidneys and organs previously mentioned, and analysis of biochemistry serum. The inclusion of ZEA in gilts diet interfered with the normal levels of serum biochemistry, but also caused dose-dependent histological changes in the two portions of the vagina, in the three portions of uterus, liver and kidney. Dose-dependent alterations were also observed in the ovaries; there was an increase in the number of atresic, and gilts that consumed 2 ppm of ZEA, there was little follicular development and abundant connective tissue stroma. Regarding histomorphometry, the dose-dependent hyperestrogenic effect was pronounced in the proximal vagina, the same did not occur in the three portions of uterus. ZEA did not interfere in the histomorphometry of non-atretic ovarian follicles. There were no significant changes in the histomorphometry and morphometry of the testes of the prepubertal male piglets. In the histopathological evaluation of the reproductive organs and accessory glands of the male piglets there was mainly hyperemia of the vessels and inflammatory infiltrate in the animals that consumed ZEA in their different concentrations. The addition of the AAM in diets of female piglets experimentally intoxicated with zearalenone has been shown to be an alternative to reduce the toxic effects of this mycotoxin. Increasing inclusions of ZEA in the diet of prepubertal male piglets did not alter the morphometry and histomorphometry of the testis. More work in this theme is needed to clearly define the ZEA role on the reproductive organs histology of male pigs. |

Medidas preventivas que impe?am o crescimento f?ngico nem sempre s?o eficientes, sendo necess?rio o uso de aditivos anti-micotoxinas (AAM), que s?o produtos adicionados ? ra??o, capazes de adsorver, biotransformar ou neutralizar as micotoxinas, impedindo o desenvolvimento da micotoxicose. A parede celular de Saccharomyces cerevisiae virou foco de in?meros estudos, sendo utilizada como AAM, pois possui alta capacidade de liga??o, e de adsor??o ?s micotoxinas. Tendo em vista tais considera??es, o objetivo deste trabalho foi analisar os efeitos da zearalenona sobre par?metros bioqu?micos s?ricos, e par?metros histopatol?gicos e morfom?tricos do trato reprodutivo de leitoas e leit?es pr?-p?beres intoxicadas experimentalmente com diferentes doses desta micotoxina e assim, avaliar a efic?cia do aditivo anti-micotoxina ? base de parede celular de leveduras na preven??o da micotoxicose. O per?odo experimental foi de 21 dias. Foram utilizados oito leit?es pr?-p?beres, cujas dietas apresentaram quatro n?veis de inclus?o de ZEA (0,0; 0,25; 0,6 e 2,0 mg Kg -1 ou ppm). Foram realizadas an?lises morfom?tricas de comprimento e largura dos test?culos. E ainda, foram realizadas mensura??es histol?gicas nos test?culos, e avalia??es histopatol?gicas do: test?culo direito e esquerdo, epid?dimo direito e esquerdo (cabe?a, corpo e cauda), ducto deferente direito e esquerdo, pr?stata, gl?ndula bulbouretral e ves?cula seminal, e an?lise da bioqu?mica s?rica. Foram tamb?m utilizadas 48 leitoas pr?-p?beres, cujas dietas apresentaram dois n?veis de inclus?o de aditivo anti-micotoxina (AAM) (0,0 e 0,2%) e tr?s n?veis de inclus?o de ZEA (0,25; 0,6 e 2,0 mg Kg -1 ou ppm), sendo realizadas an?lises histomorfom?tricas da vagina proximal e distal, ?tero proximal, m?dio e distal, ov?rio direito e esquerdo e an?lises histopatol?gicas do f?gado, rins e dos ?rg?os citados anteriormente, al?m de an?lise da bioqu?mica s?rica. A inclus?o de ZEA na dieta das leitoas interferiu nos ?ndices normais da bioqu?mica s?rica, como tamb?m provocou altera??es histol?gicas dose dependentes nas duas por??es da vagina e nas tr?s por??es uterinas, f?gado e rins. Nos ov?rios tamb?m foram observadas altera??es dose dependentes, ocorreu aumento no n?mero de fol?culos atr?sicos, sendo que em leitoas que consumiram 2 ppm de ZEA foi observado escasso desenvolvimento folicular e abundante estroma de tecido conjuntivo. Com rela??o a histomorfometria, o efeito hiperestrog?nico dose dependente foi pronunciado na vagina proximal, o mesmo n?o ocorreu nas tr?s por??es uterinas. A ZEA n?o interferiu na histomorfometria dos fol?culos ovarianos n?o atr?sicos. N?o houveram altera??es significativas na histomorfometria e na morfometria dos test?culos dos leit?es pr?-p?beres. J? na avalia??o histopatol?gica dos ?rg?os reprodutores e gl?ndulas acess?rias dos leit?es houve principalmente hiperemia dos vasos e infiltrado inflamat?rio nos animais que consumiram ZEA nas suas diferentes concentra??es. A adi??o do AAM em quest?o nas dietas das leitoas contendo ZEA demonstrou ser uma alternativa para redu??o dos efeitos t?xicos desta micotoxina. As inclus?es crescentes de ZEA na dieta de leit?es pr?-p?beres n?o alteraram a morfom?tria e histomorfom?tria dos test?culos. No entanto, causa altera??es inespec?ficas na estrutura histol?gica de ?rg?os do sistema reprodutor e gl?ndulas acess?rias. Zearalenone (ZEA) is a mycotoxin that causes a lot of problems in the reproductive tract, being related to the hyperestrogenism in gilts. Preventive measures to prevent fungal growth are not always effective, the use of anti- mycotoxin additives (AMA), which are added to food products, capable of adsorbing, or neutralizing biotransform mycotoxins, preventing the development of mycotoxicosis are required. The cell wall of Saccharomyces cerevisiae has become the focus of numerous studies and is used as AMA, because it has high binding capacity, and adsorption of mycotoxins. The objective of this study was to analyze the effects of zearalenone on serum biochemical parameters and histopathological and morphometric parameters of the reproductive tract of pre-pubertal male and female piglets experimentally poisoned with different doses of this mycotoxin and to evaluate the effectiveness of the anti-mycotoxin additive made of yeast cell wall in the prevention of mycotoxicosis. The trial period had a total duration of 21 days. Eight pre-pubertal male piglets were used, whose diets presented four levels of ZEA (0.0, 0.25, 0.6 and 2.0 mg kg -1 or ppm). Histomorphometric analyzes of the testis length and width were performed. Histological measurements were also performed on the testicles and histopathological evaluations of the right and left testicles, right and left (head, body and tail) epididymis, right and left deferent duct, prostate, bulbourethral gland and seminal vesicle, and biochemistry serum analysis. In addition, 48 prepubertal female piglets were used, whose diets presented two levels of inclusion of anti-mycotoxin additive (AMA) (0.0 and 0.2%) and three levels of ZEA (0.25; 0.6 and 2.0 mg kg -1 or ppm). And still histomorphometric analyzes of the proximal vagina, distal vagina, proximal uterus, medial uterus, distal uterus, right and left ovary, and histopathological findings of the liver, kidneys and organs previously mentioned, and analysis of biochemistry serum. The inclusion of ZEA in gilts diet interfered with the normal levels of serum biochemistry, but also caused dose-dependent histological changes in the two portions of the vagina, in the three portions of uterus, liver and kidney. Dose-dependent alterations were also observed in the ovaries; there was an increase in the number of atresic, and gilts that consumed 2 ppm of ZEA, there was little follicular development and abundant connective tissue stroma. Regarding histomorphometry, the dose-dependent hyperestrogenic effect was pronounced in the proximal vagina, the same did not occur in the three portions of uterus. ZEA did not interfere in the histomorphometry of non-atretic ovarian follicles. There were no significant changes in the histomorphometry and morphometry of the testes of the prepubertal male piglets. In the histopathological evaluation of the reproductive organs and accessory glands of the male piglets there was mainly hyperemia of the vessels and inflammatory infiltrate in the animals that consumed ZEA in their different concentrations. The addition of the AAM in diets of female piglets experimentally intoxicated with zearalenone has been shown to be an alternative to reduce the toxic effects of this mycotoxin. Increasing inclusions of ZEA in the diet of prepubertal male piglets did not alter the morphometry and histomorphometry of the testis. More work in this theme is needed to clearly define the ZEA role on the reproductive organs histology of male pigs. |
