Resumo: | Parkinson's disease (PD) is a chronic and progressive neurodegenerative disease,characterized by progressive loss of dopaminergic neurons, mainly in the substantia nigraparscompacta (SNpc). Studies employing animal models have sought to mimic the progressivenature of behavioral and pathophysiology features of PD, each one with advantagesanddisadvantages. Reserpine (RES) model, which mimics the progressive nature of PD, isbasedon repeated administration of 10 to 15 injections with a low dose (0.1 mg.kg-1) inrats.However, the interruption of the RES administration is accompanied by a recoveryoftheobserved changes. The present study aimed to verify whether the prolonged treatment withRES allows to mimic the progressive and irreversible nature of PD. Thirty-two maleWistarrats were used, divided into 4 groups: Control (CTR); Reserpine (RES); Control andsubsequent abstinence period (CTR.A); RES and subsequent abstinence period (RES.A). Allanimals received 45 subcutaneous injection of RES (0.1 mg.kg -1) or vehicle, one every48h.On day 90, animals from CTL and RES groups were killed by transcardiac perfusionandthebrains were submitted to immunohistochemical analysis of tyrosine hydroxylase(TH).Animals in groups CTR.A and RES.A remained in the experimental protocol for another30days, but without receiving injections. Throughout the experiment the animals were subjectedto catalepsy tests and evaluation of oral movements to assess motor activity. Animals treatedwith RES showed progressive motor alteration in the catalepsy test fromday36andprogressive alterations, from day 20, in the evaluation of oral movements, inthethreeparameters observed: 1- oral tremor, 2- tongue protrusion and 3-chewing. In conclusion, the results found showed that, after 45 injectionsofRES, the animals showed reversible motor changes, however they induced an irreversibledamage in nigrostriatal pathway during the evaluation period. |

Múltiplos modelos animaisprocuram mimetizar as alterações comportamentais e fisiopatológicas da DP, cada umcomvantagens e desvantagens. O modelo da Reserpina (RES), que mimetiza a naturezaprogressiva da DP, é baseado na administração repetida de 10 a 15 injeções comumabaixadose (0,1 mg.kg -1) em ratos. Entretanto, a interrupção da administração deRESéacompanhada de uma recuperação das alterações observadas. O presente estudotevecomoobjetivo verificar se o tratamento prolongado com RES permite mimetizar a naturezaprogressiva e irreversível da DP. Foram utilizados 32 ratos Wistar machos divididosem4grupos: Controle (CTR); Reserpina (RES); Controle e posterior período de abstinência(CTR.A); RES e posterior período de abstinência (RES.A). Todos os animais receberam45injeções s.c. de RES (0,1 mg.kg -1) ou veículo, uma a cada 48h. No dia 90, os animaisdosgrupos CTR e RES foram mortos por perfusão transcardíaca e os encéfalos submetidosaimunohistoquímica para Tirosina Hidroxilase (TH), enquanto os animais dos grupos CTR.AeRES.A permaneceram no protocolo experimental por mais 30 dias, porémsemreceberinjeções. Ao longo do experimento os animais foram submetidos aos testes de catalepsiaeavaliação de movimentos orais para avaliação da atividade motora. Os animais tratadoscomRES apresentaram alteração motora progressiva no teste de catalepsia a partir dodia36ealterações progressivas, a partir do dia 20, na avaliação dos movimentos orais, nostrêsparâmetros observados: 1- tremor oral, 2- protrusão de língua e 3-mastigação. Animaisdogrupo RES.A apresentaram reversibilidade das alterações motoras (catalepsia e movimentosorais) 30 dias após a interrupção do tratamento. A avaliação da imunorreatividade paraTHmostrou que o tratamento com RES foi capaz de induzir redução do número de neurôniosnaSNpc e Área Tegmentar Ventral (VTA), além de uma redução na densidade óptica relativanoEstriado Dorsal (ED). O grupo RES.A apresentou recuperação da imunorreatividadeparaTHapenas em VTA.
