Resumo: | Fungi, because they present an enormous biodiversity, give rise to a great variety of these secondary metabolites, that have diverse functionalities. Its biotransformation study has been very useful in the production of new compounds, due to the increase of productivity of the desired substances, overcoming problems encountered in chemical analysis and the possibility of elucidation of the biosynthetic pathway. Thus, substances with broad therapeutic power, such as those isolated from Zingiber officinale, are being widely used in biocatalytic studies. Based on this, this work evaluated the biotransformation potential of two endophytic fungi isolated from the Humirianthera ampla plant against gingerols isolated from Zingiber officinale. The two fungal strains were submitted to molecular identification and cultured in Petri dishes in semisolid potato Agar Dextrose (BDA) for 7 days. For the biotransformation experiments, small pieces (˜ 6 mm) of each fungus were placed in the liquid culture media Dextrose Potato (BD), Meat Extract Medium (MEP) and Saboraud (SBD) for 7 days in growth. After the fungal growth, it was added the enriched fraction of gingerols (organic substrate), leaving under agitation for another 7 days. Experiments varying the growth days (7 °, 8 °, 9 °, 10 ° and 14 ° days) with the enriched fraction were also performed. At the end of the biotransformations, the extracts were obtained by liquid-liquid partition with ethyl acetate and analyzed by chromatographic methods. All procedures were performed in triplicate and with control experiments. The fungi were identified in Scytalidium lignicola and Scytalidium sp. For both fungi, the biotransformation was only observed in the medium of MEP growth and from the different chromatographic methods the appearance of a more apolar substance was observed in relation to the gingerols tested in all the studied days. This work has an unprecedented character since there are no papers in the literature on the isolation and identification of the endophytic fungi of the H. ampla nor the use of them in processes of biotransformation. |

O seu estudo em processos de biotransformação tem sido muito útil na produção de novos compostos, devido ao aumento da produtividade das substâncias desejadas, superação dos problemas encontrados em análises químicas e a possibilidade da elucidação de vias biossintéticas. Assim, substâncias com amplo poder terapêutico, como as isoladas de Zingiber officinale, estão sendo bastante utilizados em estudos biocatalíticos. Baseado nisso, este trabalho avaliou o potencial de biotransformação de dois fungos endofíticos isolados da planta Humirianthera ampla frente à gingeróis isolados de Zingiber officinale. As duas cepas fúngicas foram submetidas a identificação molecular e cultivadas em placas de Petri em meio semissólido Batata Dextrose Ágar (BDA) durante 7 dias. Para os experimentos de biotransformação, pequenos pedaços (˜ 6 mm) de cada fungo foram colocados nos meios de cultura líquidos Batata dextrose (BD), Meio de Extrato de carne (MEP) e Saboraud (SBD) por 7 dias em crescimento. Após o crescimento fúngico, adicionou a fração enriquecida dos gingeróis (substrato orgânico), deixando sob agitação por mais 7 dias. Experimentos variando os dias de crescimento (7°, 8°, 9°, 10° e 14° dias) com a fração enriquecida também foram realizados. Ao final das biotransformações, os extratos foram obtidos por partição líquido-líquido com acetato de etila e analisados por métodos cromatográficos. Todos os procedimentos foram realizados em triplicata e com experimentos controles. Os fungos foram identificados em Scytalidium lignicola e Scytalidium sp. Para ambos os fungos, a biotransformação foi observada apenas no meio de crescimento MEP e a partir dos diferentes métodos cromatográficos observou-se o aparecimento de uma substância mais apolar em relação aos gingeróis testados em todos os dias estudados.
