Resumo: Endophytic fungi are potential producers of bioactive substances that perform various biological functions. This work was an investigation of the biological activities of the organic extracts of endophytic fungi Humirianthera ampla. n the identification the genetic materials were extracted, amplified, sequenced and analyzed by a phylogenetic study. UFCM 0533 fungus was grown in different culture media (potato dextrose - BD, sabouraud dextrose - SBD, meat extract and peptone - MEP and rice) for 22 days. After this period were extracted by different methods of extraction: maceration, ultrasound and ultrasound combined with maceration in order to determine what provided greater biological yield. The chemical profile of the extracts was evaluated by analytical thin layer chromatography (ADCC). In the biological activities the extracts of two fungi were used, and extracts of UFCM 0550 were obtained in previous studies by our research group. The extracts were tested for antibacterial activity against Escherichia coli and Staphylococcus aureus and cytotoxic activity against four tumor cell lines - HCT-116 (Colorretal), SF295 (Glioblastoma), PC3 (Prostate) and HL 60 (leukemic). In the phylogenetic analysis, isolates UFCM 0533 and UFCM 0550 were respectively designated as Hawksworthiomyces sp. and Arthrographis sp. The analyzes revealed significant differences between the extracts and that the best condition in terms of higher biological yield was the extracts from the BD medium and extracted by maceration. The ADCC of the extracts of Hawksworthiomyces sp. revealed a change in the production of compounds in extracts of different growth media and extraction methods. In the antibacterial test of the extracts of both fungi, only rice extracts from the Hawksworthiomyces sp fungus inhibited Staphylococcus aureus. In the cytotoxicity test of Hawksworthiomyces sp fungi extracts the most promising was obtained from growth in BD and extracted by maceration that inhibited two tumor cell lines (leukemic and colorectal).

Esse trabalho realizou uma investigação de atividades biológicas dos extratos orgânicos obtidos do cultivo de fungos endofíticos isolados de Humirianthera ampla. Na identificação os materiais genéticos foram extraídos, amplificados, sequenciados e analisados por um estudo filogenético. O fungo UFCM 0533 foi cultivado em diferentes meios de cultura (batata-dextrose - BD, sabouraud dextrose – SBD, extrato de carne e peptona – MEP e arroz) durante 22 dias. Após esse período foram extraídos por diferentes métodos de extração: maceração, ultrassom e ultrassom combinado com maceração a fim de determinar o que fornecia maior rendimento biológico. O perfil químico dos extratos foi avaliado por cromatografia em camada delgada analítica (CCDA). Nas atividades biológicas utilizou-se os extratos de dois fungos, sendo que os extratos do UFCM 0550 foram obtidos em estudos anteriores pelo nosso grupo de pesquisa. Os extratos foram testados quanto a atividade antibacteriana contra Escherichia coli e Staphylococcus aureus e atividade citotóxica contra quatro linhagens de células tumorais - HCT-116 (Colorretal), SF295 (Glioblastoma), PC3 (Próstata) e HL 60 (leucêmica). Na análise filogenética, os isolados UFCM 0533 e UFCM 0550 foram apontados respectivamente como Hawksworthiomyces sp. e Arthrographis sp. As análises revelaram diferenças significativas entre os extratos e que a melhor condição em termos de maior rendimento biológico, foi a dos extratos oriundos do meio BD e extraídos por maceração. A CCDA dos extratos de Hawksworthiomyces sp. revelou mudança na produção de compostos nos extratos dos diferentes meios de crescimento e métodos de extração. No teste antibacteriano dos extratos de ambos os fungos, apenas os extratos do cultivo em arroz do fungo Hawksworthiomyces sp inibiram Staphylococcus aureus.
