Influência dos hormônios ovarianos sobre estruturas ocitocinérgicas pré-autonômicas envolvidas no controle da pressão arterial em ratas

Resumo: | The prevalence of cardiovascular diseases including hypertension increases dramatically in women after menopause, however the mechanisms involved remain incompletely understood. Oxytocinergic (OTergic) neurons are largely present within the paraventricular nucleus of the hypothalamus (PVN). Several studies have shown that OTergic drive from PVN to brainstem increases baroreflex sensitivity and improves autonomic control of the circulation. Since preautonomic PVN neurons express different types of estrogen receptors, we hypothesize that ovarian hormone deprivation causes baroreflex impairment, autonomic imbalance and hypertension by negatively impacting OTergic drive and oxytocin levels in pre-autonomic neurons. Here, we assessed oxytocin gene and protein expression (qPCR and immunohistochemistry) within PVN subnuclei in sham-operated and ovariectomized Wistar rats. Conscious hemodynamic recordings were used to assess resting blood pressure and heart rate and the autonomic modulation of heart and vessels was estimated by power spectral analysis. We observed that the ovarian hormone deprivation in ovariectomized rats decreased baroreflex sensitivity, increased sympathetic and reduced vagal outflows to the heart and augmented the resting blood pressure. Of note, ovariectomized rats had reduced PVN oxytocin mRNA and protein expression in all pre-autonomic PVN subnuclei. Furthermore, reduced PVN oxytocin protein levels were positively correlated with decreased baroreflex sensitivity and negatively correlated with increased LF/HF ratio. These findings suggest that reduced oxytocin expression in preautonomic neurons of the PVN contributes to the baroreflex dysfunction and autonomic deregulation observed with ovarian hormone deprivation. |

Entretanto os mecanismos envolvidos na gênese desses processos patológicos não estão completamente elucidados. Neurônios ocitocinérgicos (OTérgicos) estão presentes em grande quantidade no núcleo paraventricular do hipotálamo (PVN). Estudos prévios mostraram que o aumento de ação OTérgica para o tronco encefálico pode melhorar a sensibilidade do barorreflexo, bem como, o controle autonômico da circulação. Considerando que neurônios pré-autonômicos do PVN expressam diferentes tipos de receptores para estrogênio, hipotetizamos que a privação de hormônios ovarianos causaria diminuição da eficiência do barorreflexo, disautonomia e hipertensão por impactar negativamente as projeções OTérgicas e a expressão de ocitocina nesse local. Para testar tal hipótese, mensuramos gene e expressão proteica de ocitocina (qPCR e imunohistoquímica) em subnúcleos do PVN de ratas Wistar ovariectomizadas ou sham. Registros hemodinâmicos em animais acordados foram utilizados para mensuração da pressão arterial, frequência cardíaca, sensibilidade espontânea do barorreflexo. A modulação autonômica cardíaca e a vascular foram estimadas por meio da potência de análise espectral. Observamos que a privação de hormônios ovarianos promoveu redução da sensibilidade do barorreflexo, aumento da modulação simpática concomitante a diminuição da modulação vagal cardíaca e elevação da pressão arterial média em repouso. Ademais, ratas ovariectomizadas tiveram menor expressão de RNAm e expressão de ocitocina em todos os núcleos pré-autonômicos do PVN. Além disso, reduzido nível proteico de ocitocina correlaciona-se positivamente com a diminuída sensibilidade do barorreflexo e negativamente com a razão LF/HF.
