Resumo: | During intrauterine life, the fetus needs adequate conditions for its correct development. If some insult or different stimulus occurs, physiological and metabolic physiological and metabolic adaptations that may have negative repercussions in the adult life of this individual. This concept is known as fetal programming. It is known that the fetus depends thyroid hormones for its correct growth. These exert essential essential physiological actions in the functioning of biological systems, with emphasis on the development of the immune system. Based on studies that have shown that experimental gestational hypothyroidism (EGH) alters the immune response and that adverse conditions during intrauterine development are important predictors of susceptibility to the development susceptibility to the development of allergic diseases such as asthma, the objective of this to investigate the influence of HGE on the inflammatory response of offspring subjected to allergic asthma. HGE was induced in pregnant Swiss mice by the administration of Methimazole 0.02% in the drinking water, from the 9th gestational day until the day of delivery. To verify the influence of HGE on lung inflammation, offspring from hypothyroid (PMH) and (PME) mothers were subjected to an animal model of allergic asthma induced by egg albumin-induced allergic asthma (OAI). The data obtained were expressed as mean ± standard error of the mean (S.E.P.M.). Analyses were performed using two-way ANOVA, followed by Bonferroni's post-test. Bonferroni post-test. Se ocorrer algum insulto ou estímulo diferente, surgem adaptações fisiológicas e metabólicas que podem repercutir de maneira negativa na vida adulta deste indivíduo. Esse conceito é conhecido como programação fetal. Sabe-se que o feto depende dos hormônios tireoideanos maternos para o seu correto crescimento. Esses exercem ações fisiológicas essenciais no funcionamento dos sistemas biológicos, com destaque para o desenvolvimento do sistema imunológico. Baseado em trabalhos que mostraram que o hipotireoidismo gestacional experimental (HGE) altera a resposta imunológica e que as condições adversas durante o desenvolvimento intrauterino são importantes programadoras da susceptibilidade ao desenvolvimento de doenças alérgicas, como a asma, o objetivo desta pesquisa foi investigar a influência do HGE na resposta inflamatória da prole submetida a asma alérgica. O HGE foi induzido em camundongos Swiss prenhas pela administração de Metimazol 0,02% na água de beber, do 9° dia gestacional até o dia do parto. Para verificar a influência do HGE na inflamação pulmonar, proles de mães hipotireoideanas (PMH) e eutireoideanas (PME) foram submetidas a um modelo animal de asma alérgica induzida por ovoalbumina (AAIO). Os dados obtidos foram expressos em média ± erro padrão da média (E.P.M.). As análises foram feitas utilizando a ANOVA two-way, seguida do pós-teste de Bonferroni. O nível crítico fixado foi de 5%. A PMH-OVA apresentou menor número total de células no lavado broncoalveolar (LBA) (p < 0,001); houve, também, menor atividade da mieloperoxidase (MPO) no pulmão (p < 0,001) e menor conteúdo de peroxidase eosinofílica no LBA (p < 0,05), comparados à PME-OVA.
