Resumo: The species Aloe vera (L.) Burm.F. (Xanthorroeaceae), popularly known as slug, has been used since antiquity for various purposes. From its most used part, leaf gel, was discovered a protein of molecular mass 14 kDa that has a potent antifungal action against Candida albicans. Considering that the therapeutic options available to treat fungal infections present limitations, it is sought for substances that, in addition to inhibiting microbial growth, combat their virulence factors, aiding in the treatment of infections. Thus, the objective of the present work was to obtain protein fractions of A. vera, to characterize them and to evaluate the antifungal action against Candida albicans, as well as to evaluate the effect of inhibition of biofilm formation. The process of obtaining A. vera gel proteins was performed using three-phase liquid-liquid partitioning and dialysis techniques. In the first step of the process, a three-phase liquid-liquid partition was performed using t-butanol, tris-hydroxymethylaminomethane buffer (TRIS, 50 mM, pH 8.0) and ammonium sulfate solution (1.6 M). The fraction obtained after this step was dissolved in TRIS buffer and dialyzed for a period of 24 h at 4 ° C under constant stirring. The two fractions obtained after dialysis were lyophilized and quantified by the Bradford method. The antifungal activity assays were conducted by the 96-well plate microdilution method, and then the biofilm formation evaluation was performed with the same plate. It was not possible to detect / quantify the proteins present in the fractions, however by an NMR analysis the presence ofsugars was verified. These fractions at the following concentrations of 250; 125; 31.2 and 7.8 µg / mL, presented antifungal activity with respect to the inhibition of C.albicans biofilm formation, inhibiting about 70% of it. Thus it is expected that these results may contribute to the discovery of new prototypes to antifungal drugs.

Da sua parte mais utilizada, o gel das folhas, foi descoberta uma proteína de massa molecular 14 kDa que possui uma potente ação antifúngica frente à Candida albicans. Tendo em vista que as opções terapêuticas disponíveis para tratar infecções fúngicas apresentam limitações busca-se por substâncias que, além de inibir o crescimento microbiano, combatam seus fatores de virulência, auxiliando no tratamento das infecções. Assim, o objetivo do presente trabalho foi obter frações proteicas de A. vera, caracterizá-las e avaliar a ação antifúngica frente à Candida albicans, bem como avaliar o efeito de inibição de da formação de biofilme. O processo de obtenção das proteínas do gel de A. vera foi realizado utilizando técnicas de partição líquido-líquido trifásica e diálise. Na primeira etapa do processo, foi realizada uma partição líquido-líquido trifásica, utilizando tbutanol, tampão tris-hidroximetilaminometano (TRIS, 50 mM, pH 8,0) e solução de sulfato de amônio (1,6 M). A fração obtida, após esta etapa, foi dissolvida em tampão TRIS e dialisada, por um período de 24 h a 4°C, sob agitação constante. As duas frações obtidas após a diálise foram liofilizadas e quantificadas pelo método de Bradford. Os ensaios de atividade antifúngica foram conduzidos pelo método da microdiluição em placa de 96 poços e, posteriormente, com a mesma placa realizouse a avaliação da formação de biofilme. Não foi possível detectar/quantificar as proteínas presentes nas frações, entretanto por meio de uma análise por RMN verificou-se a presença de açúcares. Estas frações nas concentrações de 250; 125;31,2 e 7,8 ?g/ mL, apresentaram atividade antifúngica no que concerne à inibição da formação de biofilme de C. albicans, inibindo cerca de 70% do mesmo.
