Resumo: | Nopalea cochenillifera (L.) Salm-Dyck occurs from Mexico to Panama and is acclimatized in several countries. It is a shrubby plant that reaches 3-4 m tall, has a cylindrical body and items called cladodes. This species is known by names such as "sweet palm" and "little palm". It is used as human food and in traditional medicine as anti-inflammatory, analgesic, hypoglycemic, lipid-lowering, antimicrobial and diuretic. In cladodes of N. cochenillifera are presents flavonoids, saponins, tannins, anthraquinones and sitosterol. Given the importance of its uses, it is essential to the analysis of mineral constituents, their antioxidant profile and their toxicological effects to ensure safe operation. Thus, this study aimed to determine the occurrence of mineral nutrients in the cladodes by atomic absorption spectrometry; analysis of oxidative parameters like amount of total free radicals, lipid peroxidation and amount of protein carbonyls; genotoxicological assessments of the extract in the human leukocytes culture, by cell viability and proliferation, micronucleus test and DNA damage, also extract cytotoxicity culture of CHO and NCTC cells. It was observed that the cladodes of Nopalea cochenillifera have a significant calcium content (6271.0 mg/100g), potassium (3849.0 mg/100g), magnesium (2998.0 mg/100g), zinc (14.7mg/100g), iron (7.29 mg/100g) and a low sodium content (5mg/100g). N. cochenillifera can contribute to the Recommended Daily Intake considering the amounts needed for a healthy adult. This extract acted as antioxidant agent, reducing and reversing the effects caused by hydrogen peroxide. In peripheral blood cells was observed that the samples showed no mutagenic effect, didn't cause DNA damage or decrease in cell viability, not being genotoxic. Nopalea cochenillifera showed low cytotoxicity on NCTC and CHO cells. The data obtained cooperate with the studies in the field and can be used as inputs for the preparation of pharmaceutical preparations with this substance |

The data obtained cooperate with the studies in the field and can be used as inputs for the preparation of pharmaceutical preparations with this substanceNopalea cochenillifera (L.) Salm-Dyck ocorre do México ao Panamá e encontra-se aclimatada em diversos países. É uma planta arbustiva que atinge 3 a 4 m de altura, possui tronco cilíndrico e artículos denominados cladódios. Esta espécie é conhecida por denominações como palma-doce e palma miúda. É utilizada na alimentação humana e na medicina tradicional como anti-inflamatória, analgésica, hipoglicemiante, hipolipidemiante, antimicrobiana e diurética. Nos cladódios de N. cochenillifera estão presentes, flavonoides, saponinas, taninos, antraquinonas e sitosterol. Diante da importância dos seus usos, torna-se fundamental a análise deconstituintes minerais, do seu perfil antioxidante e seusefeitos toxicológicos visando garantir a segurança no uso. Sendo assim, este trabalho teve como objetivo determinar a ocorrência de nutrientes minerais nos cladódios através da espectrometria de absorção atômica; análises de parâmetros oxidativos como quantidade de radicais livres totais, peroxidação lipídica e quantidade de proteínas carboniladas; avaliações genotoxicológica do extrato frente à cultura leucócitos humanos, através da viabilidade e proliferação celular, teste de micronúcleo e dano ao DNA. Assim como,citotoxicidade do extrato frente a cultura de CHO e NCTC. Observou-se que os cladódios de Nopalea cochenillifera apresentam teor significativo de cálcio (6271,0 mg/100g), potássio (3849,0 mg/100g), magnésio (2998,0 mg/100g), zinco (14,7mg/100g), ferro (7,29 mg/100g) e um baixo conteúdo de sódio (5mg/100g). N. cochenillifera pode contribuir para os aportes de Ingestão Diária Recomendada considerando os valores necessários para um adulto saudável. O extrato de Nopalea cochenillifera agiu como agente antioxidante reduzindo e revertendo os efeitos causados pelo peróxido de hidrogênio.Em células de sangue periférico foi possível observar que as amostras não apresentaram efeito mutagênico, não causaram dano ao DNA e não causaram diminuição da viabilidade celular, não sendo genotóxica. Nopalea cochenillifera apresentou baixa citotoxicidade em células NCTC e CHO. Os dados obtidos contribuem para os estudos na área e podem ser utilizados como subsídios para elaboração de preparações farmacêuticas a base dessa substância.Coordenação de Aperfeiçoamento de Pessoal de Nível Superior - CAPESporUniversidade Federal de Santa MariaCentro de Ciências da SaúdePrograma de Pós-Graduação em Ciências FarmacêuticasUFSMBrasilFarmacologiaAttribution-NonCommercial-NoDerivatives 4.0 Internationalhttp://creativecommons.org/licenses/by-nc-nd/4.0/info:eu-repo/semantics/openAccessNopalea cochenilliferaMineraisPerfil oxidativoGenotoxicidadeMineralsOxidative profileGenotoxicityCNPQ::CIENCIAS BIOLOGICAS::FARMACOLOGIATeor de minerais, perfil oxidativo e toxicidade de Nopalea cochenillifera (L.) salm-dick (cactaceae)info:eu-repo/semantics/publishedVersioninfo:eu-repo/semantics/doctoralThesisManfron, Melânia Palermohttp://lattes.cnpq.br/6737713729653264Fogaça, Aline de Oliveirahttp://lattes.cnpq.br/4441961081202251Carvalho, Leandro Machado dehttp://lattes.cnpq.br/6652387343920028Souto, Ricardo Bizognehttp://lattes.cnpq.br/2609140331593396Dalmora, Sergio Luizhttp://lattes.cnpq.br/4505166045049607http://lattes.cnpq.br/4029402462196629Necchi, Raquel Medina Martins2010000000006000e590c6d-726c-423c-8af6-55070bd29d4a1e4600ed-a56e-47f8-ad46-a998a002f2d97557b840-0452-4882-940e-2f5fefe4f6c6f9cee7fd-315c-4026-aedb-7f1cf62ed49fcf76b7c6-598e-40f0-9a07-08760c74cbd3c4a5b1b0-42e8-4585-bd7d-b0972d95d7d8reponame:Manancial - Repositório Digital da UFSMinstname:Universidade Federal de Santa Maria (UFSM)instacron:UFSMORIGINALTES_PPGCF_2016_NECCHI_RAQUEL.pdfTES_PPGCF_2016_NECCHI_RAQUEL.pdfTese de Doutoradoapplication/pdf1765493http://repositorio.ufsm.br/bitstream/1/18050/1/TES_PPGCF_2016_NECCHI_RAQUEL.pdf52b697ce840bde83af81008607b227b3MD51CC-LICENSElicense_rdflicense_rdfapplication/rdf+xml;
