Resumo: Peltophorum dubium (Sprengel) Taubert is a native species occurring from the Northeast region to the Southern region of the Brazil. Its rusticity and rapid growth in addition to the durability of its wood give it great environmental and economic importance. Their seeds have dormancy and are classified as orthodox, conferring specific physical, sanitary and physiological characteristics. In our Research Group several studies have already been developed with this species, however, some components of the in vitro rooting medium have not yet been evaluated. Thus, the objective of the present study was to optimize the micropropagation process of Peltophorum dubium, in particular the in vitro rhizogenesis stage of the shoots. The quality of the seeds that gave explants to in vitro rhizogenesis experiments was analyzed for moisture, weight of one thousand seeds, biometry, sanity and germination. In the in vitro rhizogenesis, nutritive medium components such as pH, agar and sucrose were evaluated at different levels and concentrations, combined with the presence or absence of vermiculite. In addition, different nutritive medium, in the absence of sucrose, were analyzed, associated to two growing periods. The degree of moisture and the weight of one thousand seeds confirmed its classification as orthodox and small seeds. The dimensions evaluated, as well as the small amplitude of variation, confirmed the homogeneity of the lot. The seeds have associated to 10 fungi genera and presented 86% germination. In the absence of sucrose, higher averages of root formation and length were observed (57.14 % and 2.79 cm respectively). Regardless of the concentration of agar, the presence of vermiculite favored the formation of roots (17.46 versus 3.2%) and regardless of sucrose concentrations, vermiculite impaired the quality of the root system (0.46 versus 2.41 cm and 9.51 versus 1.59% for length and secondary roots respectively). The results observed for calogenesis indicated that the presence of vermiculite in the culture medium is controversial, varying according to the composition of the nutritive medium, with respect to agar and sucrose. After 60 days of in vitro culture the highest mean (59.83%) of rhizogenesis was observed for the species. The physical characteristics of the seed lot are within the standard registered in the literature. The pH levels and agar concentrations do not influence in vitro rhizogenesis. The absence of sucrose promotes greater root formation, in addition to greater length. The presence of vermiculite increases the rooting, however it negatively affects the quality of the root system. Addition of the substrate affects the callogenesis, varying according to the composition of the nutritive medium used. The nutritive medium does not influence the in vitro rhizogenesis, but the period of growt is relevant, presenting at 60 days a greater formation and length of roots as well as of secondary roots.

Sua rusticidade e rápido crescimento, além da durabilidade de sua madeira, atribuem-lhe grande importância ambiental e econômica. Suas sementes possuem dormência e são classificadas como ortodoxas, conferindo-lhe características físicas, sanitárias e fisiológicas específicas. Em nosso Grupo de Pesquisa já foram desenvolvidos vários estudos com essa espécie, entretanto, não foram ainda avaliados alguns componentes do meio de enraizamento. Assim, o objetivo do presente estudo foi otimizar o processo de micropropagação de Peltophorum dubium, em particular da etapa de rizogênese in vitro das brotações. A qualidade das sementes que doaram explantes para os experimentos de rizogênese in vitro foi analisada quanto a umidade, peso de mil sementes, biometria, sanidade e germinação. Na rizogênese in vitro foram avaliados componentes do meio nutritivo, como pH, ágar e sacarose, em diferentes níveis e concentrações, combinados à presença ou ausência de vermiculita. Além disso, foram analisados diferentes meios nutritivos, na ausência de sacarose, associados a dois períodos de cultivo. O grau de umidade e o peso de mil sementes confirmaram sua classificação como ortodoxa e portadora de sementes pequenas. As dimensões avaliadas, bem como a pequena amplitude de variação atestaram a homogeneidade do lote. As sementes associaram-se a 10 gêneros fúngicos e apresentaram 86 % de germinação. Na ausência de sacarose foram observadas médias superiores de formação e comprimento de raízes (57,14 % e 2,79 cm respectivamente). Independentemente da concentração de ágar, a presença de vermiculita favoreceu a formação de raízes (17,46% versus 3,2%), e, independentemente das concentrações de sacarose, a vermiculita prejudicou a qualidade do sistema radicular (0,46 versus 2,41 cm e 9,51 versus 1,59 % para comprimento e raízes secundárias respectivamente). Os resultados observados para a calogênese indicaram que a presença de vermiculita no meio de cultivo é controversa, variando de acordo com a composição do meio nutritivo, no que diz respeito ao ágar e sacarose. Aos 60 dias de cultivo foi obtida a maior média (59,83 %) de rizogênese in vitro já observada para a espécie. As características físicas do lote se encontram dentro do padrão registrado na literatura. Os níveis de pH e as concentrações de ágar não influenciam a rizogênese in vitro. A ausência de sacarose promove a maior formação de raízes, além de maior comprimento. A presença de vermiculita aumenta o enraizamento, entretanto afeta negativamente a qualidade do sistema radicular. A adição do substrato afeta a calogênese, variando de acordo com a composição do meio nutritivo utilizado. O meio nutritivo não influencia a rizogênese in vitro, mas o período de cultivo é relevante, apresentando, aos 60 dias, uma maior formação e comprimento de raízes, bem como de raízes secundárias.Coordenação de Aperfeiçoamento de Pessoal de Nível Superior - CAPESporUniversidade Federal de Santa MariaCentro de Ciências RuraisPrograma de Pós-Graduação em Engenharia FlorestalUFSMBrasilRecursos Florestais e Engenharia FlorestalAttribution-NonCommercial-NoDerivatives 4.0 Internationalhttp://creativecommons.org/licenses/by-nc-nd/4.0/info:eu-repo/semantics/openAccessCanafístulaQualidade físicaSanitária e fisiológicaSacaroseVermiculitaPeríodo de cultivoForestry speciesPhysicalSanitary and physiological seed qualitySucroseVermiculiteCulture periodCNPQ::CIENCIAS AGRARIAS::RECURSOS FLORESTAIS E ENGENHARIA FLORESTALQualidade de sementes e rizogênese in vitro em Peltophorum dubium (Sprengel) TaubertQuality of seeds and in vitro rhizogenesis of shoot Peltophorum dubium (Sprengel) Taubertinfo:eu-repo/semantics/publishedVersioninfo:eu-repo/semantics/masterThesisReiniger, Lia Rejane Silveirahttp://lattes.cnpq.br/5739294882585391Paim, Aline Ferreirahttp://lattes.cnpq.br/5813893425276768Curti, Aline Ritterhttp://lattes.cnpq.br/4019523438822052http://lattes.cnpq.br/4821939486921369Gomes, Clarissa Stahl500200000003600809860fd-9f1f-4d0e-81fb-36d7e9781d1cc20c39da-da63-44af-80a8-9a40bdf5f5addf2a3c09-adf3-4326-ba66-7c8905cd6aac212a6f8d-1d8c-4ee1-af28-29fe2385f7d0reponame:Manancial - Repositório Digital da UFSMinstname:Universidade Federal de Santa Maria (UFSM)instacron:UFSMORIGINALDIS_PPGEF_2017_GOMES_CLARISSA.pdfDIS_PPGEF_2017_GOMES_CLARISSA.pdfDissertação de Mestradoapplication/pdf1360231http://repositorio.ufsm.br/bitstream/1/13284/1/DIS_PPGEF_2017_GOMES_CLARISSA.pdfac263a57406a7f3d9c3f17e3f57668a4MD51CC-LICENSElicense_rdflicense_rdfapplication/rdf+xml;
