Resumo: | The weed control is accomplished primarily through chemical herbicides. Currently, the use phytotoxins produced by microorganisms are presented as sustainable alternatives. The objective of this study was to produce a herbicide by solid state fermentation (SSF) using the fungus Diaporthe sp. The phytotoxic potential of the fermentation broth was assessed in control of target (Cucumis sativus). The experiments were divided into two steps: i) the fermentation condition for bioherbicide production was assessed. In this step the response was the percentage of damage in leaves of target; ii) evaluation of different formulations containing palm oil, Tween® 80 and Span® 80, in order to increase phytotoxicity. In step 1, the maximum herbicidal activity (1.23% of the leaves had lesions) was obtained at 25°C, moisture content of 50 wt%, supplemented with 10 wt% of corn steep liquor and soybean bran and inoculum density of 15 wt%. In step 2, the formulation containing 8.2 wt% of palm oil, 8.2 wt% of Tween® 80 and Span® 80, resulting in an HLB of 12.8 showed the highest phytotoxicity on the leaves. At this condition, dry weight of target was reduced about 36% in comparison with control. Diaporthe sp. has the potential to produce molecules with herbicidal activity and the use of adjuvants enhances its efficiency in weed control. |

Atualmente, tem-se buscado métodos mais sustentáveis no manejo destas plantas e as fitotoxinas produzidas por micro-organismos apresentam-se como alternativas. O objetivo deste trabalho foi a produção de herbicida por fermentação em estado sólido (FES) a partir do fungo Diaporthe sp. O potencial fitotóxico do caldo fermentado foi avaliado no controle da planta teste Cucumis sativus - pepino. Os experimentos foram divididos em duas etapas. Na etapa 1, foi realizada FES utilizando 10 gramas de bagaço de cana de açúcar, suplementados com água de maceração de milho e farelo de soja e inoculados com a biomassa fúngica macerada, de acordo com o planejamento de experimentos, e mantidos em câmara de acondicionamento por 7 dias. Após, foi realizada a extração dos compostos e bioensaios em casa de vegetação para a seleção da condição com maior porcentagem de dano causado em folhas. Na segunda etapa, utilizou-se a melhor condição da primeira etapa para obter moléculas e avaliado diferentes formulações contendo óleo de palma, Span® 80 e Tween® 80, com o objetivo de aumentar a fitotoxicidade. Na etapa 1, a condição de FES que produziu maior dano às folhas de Cucumis sativus foi 25°C de temperatura, 50% de teor de umidade do substrato, suplementação com água de maceração de milho e farelo de soja de 10% da massa do substrato e 15% de densidade de inóculo. Neste tratamento 1,23% das folhas apresentaram lesões. Na etapa 2, os tratamentos 8 (8,2% m/m de óleo de palma, 8,2% m/m de Span® 80 e Tween® 80 e EHL 12,8) e 14 (5,5% m/m de óleo de palma, 5,5% m/m de Span® 80 e Tween® 80 e EHL 15) apresentaram maior efeito na supressão de crescimento, em torno de 36% e maior fitotoxicidade nas folhas. O tratamento 14 teve uma redução de 36,8% na massa seca em comparação com a testemunha.
