Atividade das enzimas ntpdase, 5´-nucleotidase e adenosina deaminase em plaquetas de ratos infectados por Trypanosoma evansi

Resumo: | Nucleotide- and nucleoside-degrading enzymes are present in the surface of platelets, blood cells involved in clotting disturbances of Trypanosoma evansi-infected animals. Thus, this study was aimed at evaluating the activity of the enzymes NTPDase, 5 - nucleotidase and adenosine deaminase in platelets of rats experimentally infected by T. evansi. Animals were divided into four groups, according to the degree of parasitemia. Samples were collected at days 3 (group A: at the beginning of parasitemia), 5 (group B: high parasitemia) and 15 (group C: chronic infection). Group D (control group) was composed of non-infected animals. Blood samples with citrate as the anticoagulant were collected and used for platelet separation and enzymatic assays. NTPDase, 5 - nucleotidase and adenosine deaminase (ADA) activities were decreased (p<0.05) in platelets from rats of groups A and B, when compared to the control group. In group C, only NTPDase and 5 -nucleoside activities were decreased (p<0.001), observed by ADP and AMP hydrolysis. The correlation between platelet count and nucleotide and nucleoside hydrolysis was positive and statistically significant (p<0.05) in groups A and B. Platelet aggregation of all infected groups was decreased in comparison to the control group (p<0.05). Based upon the results, it is concluded that the alterations observed in the activity of the enzymes NTPDase, 5 -nucleotidase and adenosine deaminase in platelets of T. evansi-infected animals might be related to thrombocytopenia. |

Deste modo, o objetivo deste estudo foi avaliar a atividade das enzimas NTPDase, 5’- nucleotidase e adenosina deaminase em plaquetas de ratos infectados experimentalmente pelo protozoário T. evansi. Os animais foram divididos em quatro grupos de acordo com o grau de parasitemia. As amostras foram coletadas nos dias 3 (grupo A: início da infecção), 5 ( grupo B: alta parasitemia) e 15 (grupo C: infecção crônica). O grupo D (grupo controle) foi formado por ratos não infectados. Amostras de sangue armazenadas em citrato de sódio foram utilizadas para a separação de plaquetas, permitindo a realização dos ensaios enzimáticos e os testes de agregação plaquetária. Nos grupos A, B e C ocorreram alterações significativas das plaquetas em relação ao controle. Os grupos A e B apresentaram uma diminuição significativa (p<0,05) na atividade das enzimas NTPDase, 5’- nucleotidase e adenosina deaminase (ADA) se comparados ao grupo controle. No grupo C somente as enzimas NTPDase e 5’-nucleotidase apresentaram um aumento significativo (p< 0, 001), observada pela hidrólise de ADP e AMP. A correlação entre a contagem de plaquetas e hidrólise de nucleotídeos/nucleosídeos foi positiva e significativa (p<0,05) nos grupos A e B e a agregação plaquetária no grupo A, B e C apresentou uma redução significativa (p<0,05) se comparado ao grupo controle. Com os resultados obtidos, conclui-se que a atividade das enzimas NTPDase, 5'- nucleotidase e adenosina deaminase em plaquetas de ratos infectados com T. evansi apresentaram alterações, como a diminuição da atividade das enzimas no início da infecção, infecção aguda verificadas através da hidrolise de ATP, ADP e AMP e o aumento da atividade enzimática na infecção crônica na utilização do subtrato ADP e AMP.
