Resumo: | Objectives: Evaluate whether or not D-alpha-Tocopherol has a protective effect on renal morphology and function in hypertensive rats. Methods: Wistar Kyoto rats (control) or SHR (hypertensive), male, 2 month old, were distributed in 4 experimental groups: C – control treated with mineral oil; CT – control treated with D-alpha-Tocopherol (CT); H – hypertensive treated with mineral oil; and HT – hypertensive treated with D-alpha-Tocopherol. CT e HT rats received D-alpha- Tocopherol (36mg/kg) in every other day, for 60 days. The arterial blood pressure was measured each 15 days. Kidney morphology and function were studied when the animals completed 4 months old. Results: In HT rats the blood pressure was smaller than in H rats but was increased in comparison to C rats. H rats presented lower values of GFR and RPF than C. Hypertensive rats treated with D-alpha- Tocopherol showed values of GFR and RPF similar to the control values. The urinary flow, the excreted amount of sodium and the excreted amount of potassium in H rats was smaller than in C rats but in HT these parameters were similar to C. In rats H the renal vascular resistance was increased in comparison to rats C. Hypertensive rats treated with D-alpha-Tocopherol showed values of renal vascular resistance similar to the control values. Morphological analysis showed that hypertensive rats presented glomerular hypertrophy and enlarged interlobular artery’s wall. Conclusions: The D-alpha-Tocopherol treatment prevented glomerular and vascular hypertrophy. It also caused a decrease in arterial blood pressure and increased the excreted amount of sodium and potassium. The D-alpha-Tocopherol treatment improved renal and vascular function. These results suggest that D-alpha-Tocopherol has a beneficial effect on renal morphology and function of hypertensive rats. |

These results suggest that D-alpha-Tocopherol has a beneficial effect on renal morphology and function of hypertensive rats.Objetivos: Estudar a morfologia e a função renal de ratos hipertensos tratados com D-alfa-Tocoferol visando avaliar um possível efeito protetor do D-alfa-Tocoferol sobre as alterações da função renal causadas pela hipertensão arterial. Métodos: Ratos Wistar Kyoto (controle) e SHR (hipertensos) machos, com 2 meses de idade (200g), foram divididos em 4 grupos experimentais: controle tratado com óleo mineral (C), controle com D-alfa-Tocoferol (CT), hipertenso com óleo mineral (H), hipertensos com D-alfa-Tocoferol (HT). Os grupos CT e HT receberam D-alfa- Tocoferol (36mg/kg) em dias alternados por 60 dias. As pressões arteriais sistólicas foram aferidas quinzenalmente. A avaliação da função renal e a análise morfológica foram realizadas aos 4 meses de idade. Resultados: Observamos queda significativa da pressão arterial em HT em relação ao grupo H. Os animais do grupo H apresentaram valores de ritmo de filtração glomerular (RFG) menores que o grupo C. Em HT houve aumento do RFG em relação ao H. O fluxo urinário (V) no grupo H foi significativamente menor que os de C. Em HT os valores de V foram semelhantes aos de C. A fração de excreção e a carga excretada de Na+ em H foram significativamente menores que em C. Em HT houve aumento de excreção de Na+ em relação ao grupo H. Os animais do grupo H apresentaram valores de carga excretada e fração de excreção de K+ significativamente menores que em C. Já em HT, os valores de excreção de K+ foram semelhantes aos do grupo C. Em H observamos aumento da ECMAI e da RVR em relação ao grupo C, enquanto que nos animais hipertensos tratados com D-alfa-Tocoferol não observamos estas alterações. Quanto à área glomerular, nos animais hipertensos, o tratamento com Dalfa- Tocoferol causou redução neste parâmetro. Observamos valores de proteinúria significativamente maiores nos animais hipertensos que no grupo controle e o tratamento com D-alfa-Tocoferol não alterou a excreção de proteínas no grupo HT. Conclusões: O tratamento com D-alfa-Tocoferol preveniu a hipertrofia glomerular e vascular; melhorou a função renal e a função vascular; normalizou a excreção renal de eletrólitos e água e reduziu a pressão arterial.
