Nanopartícula pH responsiva para administração oral de antiparasitários no tratamento de peixes ornamentais

Resumo: Studies show that the global trade in ornamental fish has a high economic impact, as it involves buying and selling millions of specimens a year, generating thousands of jobs and revenues. Meanwhile, it increases the risk of cross-border spread of infectious diseases through the spread of live animals, with devastating consequences. The present study aimed to develop a strategy of innovative aspects in the treatment of endoparasites in ornamental fish, through the production, characterization and application of nanoparticles with specific molecular characteristics of biological interaction. These chitosan-N-arginine and alginate composite nanoparticles were developed as a carrier system for anthelmintic ivermectin and praziquantel drugs and with muco-adhesive and pH responsive characteristics as a function of the application medium. For the development and study, physicochemical techniques of characterization were applied and application studies of the composites containing drugs were performed by oral administration in adult ornamental fish Corydoras schwartzi, parasitized with nematodes and digenetic trematodes, for evaluation of endoparasitic treatment. Studies have shown that the particles have pH-dependent surface charge and colloidal structure characteristics. With in vivo experiments it was observed that the particles remained in the fish's gastrointestinal system for periods longer than eight hours after administration, favoring the attenuated release of the drugs and the efficacy of the treatments, which achieved a reduction of up to 95% of the fish intestinal parasites. Thus, it was evidenced that the particles were effective in transporting and releasing drugs to the target region, enabling an innovative and effective treatment. The studies also open perspectives for the use of fish as an in vivo biological model to evaluate the applicability of bioactive carrier systems.

Entretanto, aumenta o risco de propagação transfronteiriça de doenças infecciosas através da dispersão de animais vivos, com consequências devastadoras. O presente estudo buscou desenvolver uma estratégia de aspectos inovadores no tratamento de endoparasitas em peixes ornamentais, por intermédio da produção, caracterização e aplicação de nanopartículas com características moleculares específicas de interação biológica. Estas nanopartículas compósitas de quitosana-N-arginina e alginato foram desenvolvidas como sistema carreador de fármacos anti-helmínticos, ivermectina e praziquantel e com características muco-adesivas e de resposta em função do pH do meio de aplicação. Para o seu desenvolvimento e estudo foram empregadas técnicas físico-químicas de caracterização e os estudos aplicativos dos compósitos contendo medicamentos foram procedidos via administração oral em peixes ornamentais adultos Corydoras schwartzi, parasitados com nematódeos e trematódeos digenéticos, para avaliação do tratamento endoparasitário. Os estudos evidenciaram que as partículas apresentam características de carga de superfície e de estrutura coloidal dependentes do pH. Com os experimentos in vivo observou-se também que as partículas permaneceram no sistema gastrointestinal dos peixes por períodos superiores a oito horas após a administração, favorecendo a liberação atenuada dos fármacos e a eficácia de tratamentos que alcançaram a redução de até cerca de 95% dos parasitas intestinais encontrados. Dessa forma, evidenciou-se que as partículas foram eficazes no carreamento e liberação dos fármacos até a região alvo, possibilitando o estabelecimento de um tratamento inovador e efetivo.
