Resumo: Lolium multiflorum (Lm) has been an important source for sensitizing individuals in the southern region of Brazil but despite its importance, there is no information in the literature that characterizes its allergens, making it difficult for specific diagnosis and treatment of patients. The aim was to characterize novel immunodominant antigens derived from Lm total extract (S1) and a fraction isolated (S2) in this study with potential application for allergy diagnosis by evaluating the frequency of IgE and IgG4 reactivity. An ion-exchange chromatography protocol was used to isolate -expansins from the S1 and the eluted aliquot was named S2. After that, both extracts were submitted to SDS-PAGE to obtain the electrophoretic profile. For evaluating the reactivity of specific antibodies, 46 atopic (AT) patients and 16 non–atopic (NAT) individuals were selected. The levels of specific IgE and IgG4 antibodies against S1 or S2 were determined by ELISA and Immunoblotting. The electrophoretic profile of S1 total extract showed more intensity with 12, 32, and 58 kDa bands while the 32 kDa was the main component in the S2 fraction. Specific IgE antibody levels measured by ELISA were significantly greater in AT compared to NAT subjects, using either S1 extract or S2 fraction, showing a similar degree of IgE reactivity in all individuals when using S1 or S2 in the coating phase. Specific IgG4 antibody levels measured by ELISA were significantly greater in AT compared to NAT subjects, using either S1 extract or S2 fraction, showing a similar degree of IgG4 reactivity in all individuals when using S1 or S2 in the coating phase. Thus, this study demonstrated that S2 is as effective as S1 allergen extract for diagnosing atopic individuals, particularly, the 32 kDa band was the most immunodominant component and it may be a major allergen in triggering the allergic response induced by Lm. The frequency of serum reactivity from AT patients against S1 and S2 extracts determined that the 32 kDa band was the component with the highest reactivity in both extracts, recognized by 100% of the serum pools of AT patients. Thus, this study demonstrated that the S2 fraction is as effective as the S1 allergenic extract for the diagnosis of atopic individuals, especially the 32 kDa band that was the most immunodominant component and could therefore be the main allergen in triggering the allergic response induced by Lm. In addition, IgG4 results are also promising and further studies are needed to be performed to better understand whether S2 may be useful in specific immunotherapy.

Apesar de sua relevância, não há muitas informações importantes na literatura que caracterizem seus alérgenos, bem como a resposta imune dos pacientes frente a essa fonte alergênica, o que dificulta o diagnóstico e tratamento específico dos pacientes. O objetivo nesse estudo foi avaliar os antígenos imunodominantes do extrato total de Lm (S1) e da fração isolada (S2) com potencial aplicação para o diagnóstico da alergia, por meio da frequência da reatividade dos anticorpos IgE e IgG4. Para tanto, foi feita a extração de proteínas do pólen de Lm com PBS, centrifugada e dialisada e denominada sobrenadante 1 (S1), e então foi utilizado um protocolo de cromatografia de troca iônica para isolar -expansinas de S1, fração essa nomeada de sobrenadante 2 (S2). Após isso, tanto S1 quanto S2 foram submetidas a SDS-PAGE para obter o perfil eletroforético. Para a avaliação da reatividade de anticorpos específicos, 46 pacientes atópicos (AT) e 16 não-atópicos (NAT) foram selecionados por meio do teste cutâneo de puntura. Os níveis de anticorpos IgE e IgG4 específicos a S1 e S2 foram determinados por ELISA e Immunoblotting. O perfil eletroforético de S1 revelou maior intensidade nas bandas de 12, 32 e 58 kDa, enquanto a de 32 kDa foi o principal componente na fração S2. Os níveis de anticorpos específicos da classe IgE mensurados por ELISA frente a S1 foram significativamente maiores em AT quando comparados a indivíduos NAT, da mesma maneira, os níveis de anticorpos específicos da classe IgE mensurados por ELISA frente a S2 foram significativamente maiores em pacientes AT quando comparados a NAT, mostrando um grau semelhante de reatividade de IgE ao extrato S1 e fração S2. Os níveis de anticorpos da subclasse IgG4 também foram significativamente maiores, em soros de indivíduos AT quando comparados a soros de indivíduos NAT frente ao extrato S1, da mesma forma os níveis de anticorpos da subclasse IgG4 foram significativamente maiores em soros de indivíduos AT quando comparados aos soros de indivíduos NAT frente a fração S2, o que revela similitude nas reatividades ao extrato S1 e a fração S2. A reatividade dos anticorpos das classes IgE e IgG4 dos soros de pacientes AT frente a S1 e S2 demonstrou que a banda de 32 kDa foi o componente com maior reatividade em ambos os extratos, reconhecidos por 100% dos pools de soros. Assim, este estudo demonstrou que a fração S2 é tão efetiva quanto o extrato alergênico S1 para o diagnóstico de indivíduos atópicos, com destaque para a banda de 32 kDa que foi o componente mais imunodominante, podendo assim ser o principal alérgeno no desencadeamento da resposta alérgica induzida por Lm.
