Avaliação histomofométrica de concentrado sanguíneo fibrina rica em plaquetas (PRF) associado à hidroxiapatita sintética: Estudo piloto

Resumo: | Platelet-Rich Fibrin (PRF) is an autologous fibrin matrix that acts as a reservoir for cells and growth factors, which is relevant in tissue regenerative therapy. The biofunctionalization of hydroxyapatite (HA) with PRF offers the advantage of transforming the originally osteoconductive synthetic material into a histogenic material. However, few studies have evaluated the ratio relationship between HA and PRF. The present study aimed to evaluate the pattern of distribution of HA particles associated with different PRF protocols, as well as the PRF/HA ratio. Four individuals participated in the study, undergoing 3 venipunctures at different time points. Approximately 48ml of blood were collected in 6 plastic tubes without additives during each venipuncture, separated into three groups according to the centrifugation protocol: 700g/12min (L-PRF), 350g/15min (GM350), and progressive centrifugation from 60g to 700g/15min (PROPRF). The portion above the red blood cells in tubes of the same protocol was aspirated and dispensed into glass containers (5ml) containing 0.25g of HA. After polymerization, the clots were compressed in the PRF-box, and the membranes were analyzed by micro-computed tomography (micro-CT) for the density of HA particles. Subsequently, the membranes were embedded in methacrylate and analyzed under a light microscope (LM) for histomorphometric analysis. There was no significant difference in density (P = 0.579) and ratio (P = 0.132) of HA among the groups. Considering the methodology used and the evaluated protocols, the pattern of distribution of HA particles in the biofunctionalization with PRF was similar for the different protocols. Additional studies are needed to examine the degree of binding and contact in the surface area of hydroxyapatite. |

A biofuncionalização de hidroxiapatita (HA) com PRF, oferece a vantagem de transformar o material sintético, originalmente osteocondutor, em um material histogênico. Entretanto, poucos estudos têm avaliado a relação de proporção entre HA e PRF. O presente estudo teve como objetivo avaliar o padrão de distribuição de partículas de HA associadas a diferentes protocolos de PRF, assim como a relação de proporção PRF/HA. Participaram do estudo 4 indivíduos, que foram submetidos a 3 venopunções em momentos diferentes. Em cada venopunção foram coletados cerca de 48ml de sangue em 6 tubos de plástico sem aditivos, separados em três grupos, de acordo com o protocolo de centrifugação: 700g/12min (L-PRF), 350g/15min (GM350) e centrifugação progressiva de 60g a 700g/15min total (PROPRF). A porção acima das hemácias, nos tubos de mesmo protocolo, foi aspirada e dispensada em recipientes de vidro (5ml) contendo 0,25g de HA. Após a polimerização, os coágulos foram prensados no PRF-box e as membranas foram analisadas por microtomografia computadorizada (micro-CT) quanto à densidade das partículas de HA. Em seguida, as membranas foram incluídas em metacrilato e analisadas ao microscópio de luz (ML) para análise histomorfométrica. Não houve diferença significativa na densidade (P = 0,579) e relação de proporção (P = 0,132) de HA, entre os grupos. Considerando a metodologia utilizada e os protocolos avaliados, o padrão de distribuição de partículas de HA na biofuncionalização com PRF foi semelhante para os diferentes protocolos.
