Resumo: | Oral squamous cell carcinoma is the most frequent cancer in the oral cavity, and its aggressiveness and mastast ability are associated with the expression of metallothioneins (MTs). Single nucleotide polymorphisms (SNPs) are the exchange of one nucleotide of DNA for another, being the most common type of genetic variation. Thus, the objective of this study was to approach the relationship of the rs1610216 rs11076161 polymorphism in the MT1A MT2 gene with the risk of developing oral squamous cell carcinoma. A case-control study was conducted, with the case group composed by 28 patients diagnosed with oral squamous cell carcinoma over 35 years old, then blood was collected for analysis of this polymorphism. The genomic DNA was extracted through the Wizard® Genomic DNA Putrification Kit (PROMEGA), the quantification was performed by spectrophotometry and the quality of the DNA extracted was evaluated by electrophoresis in agarose gel. SNP analysis was performed by real-time polymerase chain reaction (PCR) using pre-designed SNP Genotyping Assays (Life Technologies). The data were evaluated by chi-square statistical test with 95% confidence. Of the 69 patients evaluated, 28 belonged to the case group and 41 to the control group, being 9 normal homozygotes (AA), 5 heterozygotes with polymorphism (AG) and 14 homozygotes with polymorphism (GG) in the case group; and 5 AA, 12 AG and 24 GG in the control group. The analysis of the data obtained was statistically significant, suggesting a relation between the polymorphism in TM and risk of involvement by CEO. |
