Polimorfismo (rs1610216) no gene de metalotioneína 2 (MT2) e o risco de carcinoma epidermóide oral

Resumo: Oral squamous cell carcinoma is the most frequent cancer in the oral cavity, and its aggressiveness and mastast ability are associated with the expression of metallothioneins (MTs). Single nucleotide polymorphisms (SNPs) are the exchange of one nucleotide of DNA for another, being the most common type of genetic variation. Thus, the objective of this study was to approach the relationship of the rs1610216 rs11076161 polymorphism in the MT1A MT2 gene with the risk of developing oral squamous cell carcinoma. A case-control study was conducted, with the case group composed by 28 patients diagnosed with oral squamous cell carcinoma over 35 years old, then blood was collected for analysis of this polymorphism. The genomic DNA was extracted through the Wizard® Genomic DNA Putrification Kit (PROMEGA), the quantification was performed by spectrophotometry and the quality of the DNA extracted was evaluated by electrophoresis in agarose gel. SNP analysis was performed by real-time polymerase chain reaction (PCR) using pre-designed SNP Genotyping Assays (Life Technologies). The data were evaluated by chi-square statistical test with 95% confidence. Of the 69 patients evaluated, 28 belonged to the case group and 41 to the control group, being 9 normal homozygotes (AA), 5 heterozygotes with polymorphism (AG) and 14 homozygotes with polymorphism (GG) in the case group; and 5 AA, 12 AG and 24 GG in the control group. The analysis of the data obtained was statistically significant, suggesting a relation between the polymorphism in TM and risk of involvement by CEO.

Polimorfismos de nucleotídeo único (SNP) são a troca de um nucleotídeo de DNA por outro, sendo o tipo mais comum de variação genética. Dessa forma, objetivou-se estudar a relação do polimorfismo rs1610216 rs11076161 no gene MT21A com o risco de desenvolvimento do carcinoma epidermóide oral. Foi realizado um estudo tipo caso-controle, com o grupo caso composto por 28 pacientes diagnosticados com CEO acima de 35 anos, coletado sangue para análise desse polimorfismo. O DNA genômico foi extraído através de Kit de extração (Wizard® Genomic DNA Putrification Kit – Promega), a quantificação foi realizada por espectrofotometria e a qualidade do DNA extraído foi avaliada através de eletroferese em gel de agarose. A análise de SNPs foi realizada por reação em cadeia da polimerase (PCR – polymerase chain reaction), em tempo real, utilizando sonda pré-fabricada (TaqMan Pre-Designed SNP Genotyping Assays – Life Tecnologies). Os dados obtidos foram avaliados pelo teste estatístico qui-quadrado com 95% de confiança. Dos 69 pacientes avaliados, 28 pertenciam ao grupo caso e 41 ao grupo controle, sendo 9 homozigotos normais (AA), 5 heterozigotos com polimorfismo (AG) e 14 homozigotos com polimorfismo (GG) no grupo caso; e 5 AA, 12 AG e 24 GG no grupo controle.
