Resumo: | Prostate cancer (PCa) is the second most common type of cancer in men. Metastatic PCa is considered a more advanced type of tumor with a worse prognosis, therefore, it is important to diagnose the disease at an early stage, as well as choose the best treatment option for the patient. Aptamers are oligonucleotides that allow the search for biomarkers in target cells and can also provide information related to diagnosis, therapy and disease mechanisms. The objective of the study was to evaluate, using the resazurin method, whether there was a change in cell proliferation and viability of prostate lines when incubated with aptamers A6 and A11. The highest concentration of aptamer A6 used showed cytotoxicity in the tumor lineage and was not cytotoxic to the normal prostate lineage. The A11 aptamer did not influence cell viability and proliferation characteristics in both prostate lineages. These data suggest that aptamer A6 can be used as a therapy in patients with PCa, while A11 can be used as a possible diagnostic tool. Other analyzes and tests are needed to validate this information. |

Dessa forma, é importante realizar o diagnóstico da doença em um estágio precoce, assim como escolher a melhor opção de tratamento para o paciente. Aptâmeros são oligonucleotídeos que permitem buscar biomarcadores nas células alvo e fornecer informações relacionadas ao diagnóstico, terapia e mecanismos da doença. O objetivo do estudo foi avaliar, pelo método da resazurina, se houve alteração da proliferação e viabilidade celular de linhagens da próstata quando incubadas com os aptâmeros A6 e A11. A maior concentração utilizada do aptâmero A6 apresentou citotoxicidade na linhagem tumoral e não foi citotóxico para a linhagem normal da próstata. O aptâmero A11 não alterou as características de viabilidade e proliferação celular em ambas as linhagens da próstata. Esses dados sugerem que o aptâmero A6 pode ser utilizado como terapia em pacientes com CaP, enquanto que o A11 pode ser usado como possível ferramenta diagnóstica.
