Atividade antioxidante, antimicrobiana e caracterização de compostos bioativos da casca interna de Kielmeyera coriacea Mart. & Zucc

Resumo: | This work aims to provide information on the chemical constitution and evaluation of antimicrobial activity of ethanolic extract and fractions from the inner bark of Kielmeyera coriacea Mart & Zucc, a typical Cerrado tree and used as a medicinal plant. The study evaluated the antimicrobial and antifungal potential of ethanolic extract (EE) and partitions. Antibacterial activities study were analyzed using aerobic and anaerobic buccal bacteria by Microdiluition Method in broth, with the results expressed in minimum inhibitory concentrations (MIC). In this analysis, the n-hexane (HP) partition was the most active, presenting MIC values between 1.56 and 6.25 μg mL-1, and for the Streptococcus mitis and Streptococcus sanguinis bacteria values were better than those found for amphotericin (CIM 3.688 μg mL-1), the drug used as positive control. These results awakened interest for the isolation of compounds. The tests with fractions of n-hexane partition showed a decrease of this activity with MIC values between 12.5 and 1500μg mL-1. These results may be relates to compounds that were acting synergistically and separated during the fractionation on column. In evaluating the antifungal potential against the yeasts of the genus Candida, both the extract and partitions inhibited the growth of all species tested, with minimum inhibitory concentrations (MIC) varying from 23.43 and 1500 μg mL-1, being the partitions dichloromethane and n-butanol that presented the most promising values, at concentrations between 23.43 and 46.87 μg mL-1. The antioxidant activity evaluated by the DPPH free radical method and by the electrochemical method. The Effective Concentration (EC50) values were better for the ethyl acetate (AP) and n-butanol (BP) partitions (between 2.0 and 1.6 μg mL-1, respectively), and lower than the BHT control (4.8 μg mL-1). The antioxidant activity confirmed in the electrochemical method. These results are in accordance with the highest levels of phenolic compounds present in these partitions. Phytochemical screening also revealed the presence of phenolic compounds (tannins, xanthones, coumarins, flavonoids and saponins), as well as terpenes and terpenoids. In the n-hexanic partition identified by Gas Chromatography coupled to Mass Spectrometry and by Nuclear Magnetic Resonance the steroids β-sitosterol, stigmasterol, campesterol and sitost-4-en-3-one, the triterpenes α-amirin and lupeol and the fatty acid hexadecanoic. The presence of these compounds in the n-hexane partition may explain the strong antibacterial activity, since these compounds act in the inhibition against different microorganisms, including the buccal ones. The analysis of ethanolic extract and more polar partitions polarity by High Performance Liquid Chromatography coupled to Mass Spectrometry with Electrospray Ionization in negative and positive modes have been identified protocatecuic acid, flavonoid epi-catechin and proanthocyanidins A, B, C type and its oligomers. In the partition n-butanol was identified a dimer of epi-catechin-epi-gallocatechin and gallocatechin-3-O-glycoside; on partition ethyl acetate and methanol-water residue has been identified the dimer epi-afzelequin-epi-catechin. The identification of these phenolic compounds in polar partitions can explain the results of antioxidant and antifungal activity. This study indicates that the inner bark of Kielmeyera coriacea Mart & Zucc is rich in bioactive compounds, exhibiting biological activity against oral bacteria and fungi. Biological activities.UFU - Universidade Federal de UberlândiaDissertação (Mestrado)Este trabalho tem como objetivo contribuir com informações sobre a constituição química e a avaliação da atividade antimicrobiana do extrato etanólico e frações da casca interna de Kielmeyera coriacea Mart & Zucc, uma árvore típica do Cerrado e muito utilizada como planta medicinal. O estudo avaliou o potencial antibacteriano e antifúngico do extrato etanólico e partições. No estudo antibacteriano foram analisadas bactérias bucais aeróbias e anaeróbias pelo método da microdiluição em caldo, sendo os resultados expressos em concentrações inibitórias mínimas (CIM). Nesta análise, a partição n-hexano foi a mais ativa, apresentando valores de CIM entre 1,56 e 6,25 μg mL-1 sendo que, para as bactérias Streptococcus mitis e Streptococcus sanguinis os valores foram melhores que os encontrados para a anfotericina (CIM 3,688 μg mL-1), fármaco usado como controle positivo. Estes resultados despertaram interesse para o isolamento de compostos. Os ensaios realizados com as frações da partição hexânica mostraram uma diminuição dessa atividade com valores de CIM entre 12,5-1500 μg mL-1. Estes resultados podem estar relacionados com compostos que estavam agindo de forma sinérgica e foram separados durante o fracionamento em coluna. Na avaliação do potencial antifúngico frente às leveduras do gênero Cândida, tanto o extrato quanto as partições inibiram o crescimento de todas as espécies testadas, com concentrações inibitórias mínimas (CIM) variando entre 23,43 e 1500 μg mL-1, sendo que as partições diclorometano e n-butanol apresentaram os valores mais promissores, com concentrações entre 23,43 e 46,87 μg mL-1. A atividade antioxidante foi avaliada pelo método do radical livre DPPH e pelo método eletroquímico. Os valores de concentração efetiva (CE50) foram melhores para as partições acetato de etila e n-butanol (entre 2,0 e 1,6 μg mL-1, respectivamente), e menores que o controle BHT (4,8 μg mL-1). O potencial antioxidante foi confirmado no método eletroquímico. Estes resultados estão em conformidade com os maiores teores de compostos fenólicos presentes nestas partições polares. A prospecção fitoquímica também revelou a presença de compostos fenólicos (taninos, xantonas, cumarinas, flavonoides e saponinas), além de terpenos e terpenoides. Na partição hexânica foram identificados por Cromatografia Gasosa acoplada à Espectrometria de Massas e por Ressonância Magnética Nuclear os esteroides β-sitosterol, estigmasterol, campesterol e sitost-4-en-3-ona, os triterpenos α-amirina e lupeol e o ácido graxo hexadecanoico. A presença desses compostos na partição hexânica está relacionada à forte atividade antibacteriana, já que esses compostos atuam na inibição contra diferentes microrganismos, incluindo os orais. Na análise do extrato etanólico e das partições de maior polaridade por Cromatografia Líquida de Alta Eficiência acoplada à Espectrometria de Massas com Ionização por Electrospray nos modos negativo e positivo foram identificados o ácido protocatecóico, o flavonoide epi-catequina, e algumas proantocianidinas do tipo A, B, C e seus oligômeros. Na partição n-butanol foi identificado um dímero de epi-catequina-epi-galocatequina e a epi-catequina-3-O-glicosídeo; na partição acetato de etila e no resíduo metanol-água foi identificado o dímero epi-afzelequina-epi-catequina. A identificação destes compostos fenólicos nas partições polares pode explicar os resultados de atividade antioxidante e antifúngica. Este estudo indica que a casca interna da Kielmeyera coriacea Mart & Zucc é rica em compostos bioativos, exibindo atividade biológica contra bactérias orais e fungos.Palavras-chaves: Kielmeyera coriacea Mart & Zucc.
