Uma de suas aplica??es ? na rea??o de transesterifica??o para produ??o de biodiesel. O objetivo foi coletar, isolar fungos filamentosos produtores de lipase e analisar as caracter?sticas morfol?gicas macrosc?picas dos fungos filamentosos isolados e do banco de fungos do laborat?rio, assim como, padronizar o cultivo do microrganismo selecionado para uma maior atividade lipol?tica, seguindo caracteriza??o bioqu?mica da lipase sol?vel produzida em meio submerso e caracteriza??o f?sico-qu?mica dos ?leos vegetais. Sendo assim foi poss?vel isolar doze fungos filamentosos de distintas amostras coletadas em Diamantina-MG, cuja maioria apresentou colora??o branca, aspecto seco, topografia plana e pigmenta??o. Analisou-se o efeito da temperatura no crescimento dos doze fungos isolados e dos oito fungos filamentosos do banco de fungo do laborat?rio em meio de cultura s?lido contendo aveia e Batata-Dextrose-?gar (BDA), onde obteve-se as maiores taxas de crescimento em ambos os meios de cultura a 30?C. Pela triagem de produ??o lipol?tica o fungo do banco 3.2TA pertencente ao filo Mucoromycota e obteve o maior halo de crescimento, sendo selecionado para a fermenta??o submersa. Analisaram-se cinco meios de cultura submersos para a produ??o de lipase e a melhor atividade foi obtida no meio Adams ap?s 72 horas de cultivo, seguindo a otimiza??o, observou-se que a maior atividade na fonte de nitrog?nio foi em ureia, peptona e extrato de levedura. Para a fontes de sais a maior atividade foi em sais CP e Khanna com pH inicial de 4,0. A fonte de carbono foi analisada e a melhor atividade foi obtida em ?leo de oliva. Foi realizada a caracteriza??o bioqu?mica da enzima sobre os efeitos do pH e temperatura onde apresentou melhor atividade nas faixas de pHs 7-14 a altas temperaturas. A termoestabilidade da enzima foi a 55oC nos tempos de 60 e 90 min. A estabilidade ao pH foi a um pH de 5,5, nos tempos de incuba??o de e 60 min. A estabilidade a solventes foi obtida em metanol e etanol. A maior hidr?lise enzim?tica ocorreu em azeite de oliva, seguido do ?leo de algod?o, soja e milho. Foi realizada a caracteriza??o f?sico qu?mica de diferentes ?leos vegetais, o maior ?ndice de acidez foi no ?leo de dend? e de oliva. O maior valor para ?ndice de peroxido foi em ?leo de abacate, oliva, pequi e os menores em ?leo de trigo. Para o ?ndice de saponifica??o os maiores valores foram no ?leo de abacate, trigo, canola. Para o material insaponific?vel os maiores valores foram nos ?leos de soja, algod?o, girassol, e o menor valor no ?leo de dend?. One of its applications is in the transesterification reaction for biodiesel production. The objective was to collect, isolate lipase-producing filamentous fungi and analyze the macroscopic morphological characteristics of the isolated filamentous fungi and the laboratory fungal bank, as well as standardize the cultivation of the selected microorganism for greater lipolytic activity, following biochemical characterization of the soluble lipase. produced in submerged medium and physicochemical characterization of vegetable oils. Thus, it was possible to isolate twelve filamentous fungi from different samples collected in Diamantina-MG, most of which presented white coloration, dry appearance, flat topography and pigmentation. The effect of temperature on the growth of the twelve isolated fungi and of the eight filamentous fungi from the fungus bank in the laboratory was analyzed in a solid culture medium containing oat and Potato-Dextrose-Agar (PDA), where the highest growth rates were obtained. in both culture media at 30?C. By screening for lipolytic production, the fungus from bank 3.2TA belonging to the phylum Mucoromycota obtained the highest growth halo, being selected for submerged fermentation. Five submerged culture media were analyzed for the production of lipase and the best activity was obtained in Adams medium after 72 hours of cultivation, following the optimization, it was observed that the highest activity in the nitrogen source was in urea, peptone and extract of yeast. For the salt sources, the highest activity was in CP and Khanna salts with an initial pH of 4.0. The carbon source was analyzed and the best activity was obtained in olive oil. The biochemical characterization of the enzyme was carried out on the effects of pH and temperature where it showed better activity in the ranges of pH 7-14 at high temperatures. The enzyme thermostability was at 55oC for 60 and 90 min. The pH stability was at a pH of 5.5, at incubation times of and 60 min. Solvent stability was achieved in methanol and ethanol. The highest enzymatic hydrolysis occurred in olive oil, followed by cotton, soybean and corn oil. The physicochemical characterization of different vegetable oils was carried out, the highest acidity index was in palm and olive oil. The highest value for peroxide index was in avocado, olive, pequi oil and the lowest in wheat oil. For the saponification index the highest values were in avocado, wheat and canola oil. For the unsaponifiable material, the highest values were in soybean, cotton, sunflower oils, and the lowest value in palm oil.
