Resumo: | Bone loss is a problem, both in medicine and in veterinary medicine, which has led to researchers in the search for biomaterials to promote bone regeneration. This study aimed to evaluate the biocompatibility, osseointegration and biodegradation of the composites of hydroxyapatite (HA), polycaprolactone (PCL) and alendronate (ALO) and the polymer polycaprolactone (PCL) in circular bone defects of 5 mm diameter made in the right olecranon of rabbits. The study included 36 New Zealand rabbits that were randomly divided into four groups of equal numbers. The composites were manufactured with different proportions, composite one was composed by 49.5% hydroxyapatite, 49.5% polycaprolactone, 1% alendronate ( PCL + HA + ALO), and composite two was composed by 50% hydroxyapatite, 50% polycaprolactone (PCL + HA) and polymer polycaprolactone (PCL group). Each group received a composition above and one group received no treatment and was used as control. The evaluation was made by clinical examination on days 2, 4 and 8 after surgery and Iameness, pain, infection and dehiscence parameters were evaluated. To assess edema the circumference of the member was measured on days 1, 2, 4 and 8 post-operatively. Radiographs were taken before, immediately after surgery and the day of euthanasia. The dates 8, 45 and 90 postoperative were evaluated, with three animals per group for each date. In the last three dates olecranon was collected for histologic and morphometric analysis. There was no difference in the Iameness, pain, infection and dehiscence parameters between groups. In clinical analysis, differences were observed between the variable edema of the Iimb circumference values on day 1 and 8 in all groups. The X- Rays shown similar radiopaticy 8 days after surgery at the in the PCL+HA+ALO e PCL+HA and a smaller one at the groups PCL e .45 after surgery the groups PCL+HA+ALO e PCL+HA Showed a high percent of similar radiopacity 90 days after surgery the radiopaticy showed was 100% for PCL+HA+ALO, PCL+HA e PCL groups and 83.3% for control group. Histologically, the control group showed edges of the defect easily identifiable beginning of trabecular bone formation, this organized bone, followed by a large number of trabecular bone, the group PCL, after eight days, trabecular bone formation in the vicinity of the defect, since the groups PCL + + ALO HA and PCL + HA formation was observed both in the center and the periphery of the defect. The biomaterials used has direct contact with the tissue proving biocompatibility and also it might occour a bone formation as biomaterials have been degraded, since it was within the bone defect. |

O presente trabalho teve como objetivo avaliar a biocompatibilidade, a osseointegração e a biodegradação de compósitos de hidroxiapatita (HA), policaprolactona (PCL) e alendronato (ALO) e o polímero policaprolactona (PCL) em defeitos ósseos circulares de 5 mm de diâmetro realizados no olecrano direito de coelhos. Foram utilizados 36 coelhos da raça Nova Zelândia, distribuídos aleatoriamente em quatro grupos, de igual número. Os compósitos foram fabricados com diferentes proporções, sendo que o compósito um foi composto por 49,5% de hidroxiapatita, 49,5% de policaprolactona e 1% de alendronato (grupo PCL+HA+ALO) e o compósito dois foi de 50% de hidroxiapatita e 50% de policaprolactona (grupo PCL+HA). Um grupo recebeu o compósito um, outro recebeu o compósito dois, um terceiro grupo foi tratado apenas com o polímero de policaprolactona (grupo PCL) e um grupo não recebeu tratamento, sendo utilizado como controle. A avaliação foi feita por meio de analise clínica nos dias 2, 4 e 8 pós-cirurgia onde foram avaliados os parâmetros claudicação, sensibilidade dolorosa, deiscência e infecção. Para avaliar edema, foi medida a circunferência do membro nos dias 1, 2, 4 e 8 pós-operatórios. Foram feitas radiografias antes, imediatamente após a cirurgia e no dia da coleta de material para exame histopatológico e histomorfométrico, nas datas 8, 45 e 90 pós-operatórias, com três animais por grupo para cada data. Não houve diferença para os parâmetros claudicação, dor, infeção e deiscência entre os grupos. Na analise clinica foi observada diferença na variavel edema entre o dia 1 e 8 em todos os grupos. Radiograficamente, no lugar do defeito foi observado aos 8 dias nos grupos PCL+HA+ALO e PCL+HA pontos de radiopacidade semelhante a do osso adjacente e nos grupos PCL e controle radiopacidade menor a do osso adjacente. Aos 45 dias os grupos PCL+HA+ALO e PCL+HA apresentaram melhor resultado por terem maior porcentagem de quadrantes com pontos de radiopacidade semelhante a do osso adjacente e bordas de difícil delimitação em comparação aos grupos PCL e controle. Aos 90 dias, a radiopacidade foi observada em 100% dos quadrantes para os grupos PCL+HA+ALO, PCL+HA e PCL e só em 83.3% para o grupo controle. Histologicamente, observou-se que os animais do grupo controle apresentaram as bordas do defeito facilmente identificãveis, com início de formação de osso trabecular partindo dele, osso esse organizado, seguido por um grande número de trabéculas ósseas, o grupo PCL, aos oito dias, formação de osso trabecular na periferia do defeito, jã nos grupos PCL+HA+ALO e PCL+HA a formação foi observada tanto no centro quanto na periferia do defeito. Os biomateriais que compuseram os grupos PCL+HA+ALO e PCL+HA demostraram contato direto com o tecido ósseo aos 45 e 90 dias. Concluiu-se que os compósitos PCL+HA+ALO, PCL+HA e o polímero PCL são biocompatíveis. Acredita-se que ocorreu neoformação óssea à medida que os biomateriais foram degradados, uma vez que houve crescimento ósseo dentro do defeito, onde anteriormente era preenchido por biomaterial.
