Resumo: Sacha kiruma (Plukenetia volubilis L.) is an oleaginous native plant of the Peruvian Amazon. The oil extracted from its seeds is very rich in polyunsaturated fatty acids, directly related to the prevention of cardiovascular diseases, among them, the atherosclerosis. For transportation, lipids form complexes with proteins, the apoproteins. The apoprotein E (Apo E) is mainly synthesized in the liver and intestine and is related to the cellular uptake of triglyceride-rich chylomicrons and VLDL remmants. The production of knockout animals for the apoprotein E gene presents as main characteristic the rapid and spontaneous development of atherosclerosis lesions in the arteries, similar to those found in humans. The small intestine is the site of absorption of dietary components, including lipids, whiles the liver is the site of modification and/or storage of compounds. The objectives were extract sacha kiruma seeds´s oil by chemical method (hexane) and determine the acid profile. Also evaluate the histological structure of the duodenum and the liver of Apo E-/- mice and wild BlackC57/6 after the treatment with this oil. The evaluation of the oil extracted in this study indicates the percentage of polyunsaturated fatty acids similar to that found in the literature. There were no histological changes due the treatment in the duodenum, with regard to the surface absorption, morphology of the crypt and the thickness of muscle layers and mucosa. With respect to the height of the absorptive epithelium, the control animals have high average compared with animals treated with the oil. The frequency of enterocytes, lymphocytes, goblet cells and Paneth cells did not change due to the proposal treatment. In relation to the goblet cell AB+PAS+ was observed more frequently in the groups of BlackC57/6 mice treated with the sacha kiruma´s oil compared to control groups. In the liver were not found differences due to the treatment in the frequency of liver components or the ratio cytoplasm/ nucleus of the liver cells. The solvent extraction method was efficient in extracting oil from sacha kiruma´s seeds. The oil showed similar lipid composition from the described in the literature. The test showed no evidence that the diet with sacha kiruma oil may have altered the histological structure of the duodenum and liver of mice Apo E -/- or wild BlackC57/6.

O óleo extraído de suas sementes é muito rico em ácidos graxos poliinsaturados, diretamente relacionados à prevenção de doenças cardiovasculares, dentre elas, a aterosclerose. Para seu transporte, os lipídeos formam complexos com proteínas, as apoproteínas. A apoproteína E (Apo-E) é sintetizada principalmente no fígado e no intestino e se relaciona com a absorção celular de quilomícrons ricos em triglicerídeos remanescentes e VLDL. A produção de animais knockout para o gene da apoproteína E apresenta como principal característica o rápido e espontâneo aparecimento de lesões ateroscleróticas nas artérias, similares àquelas encontradas em humanos. O intestino delgado é o sítio de absorção de componentes da dieta, dentre eles os lipídeos, enquanto o fígado apresenta-se como o sítio de modificação e/ou armazenamento de compostos. Os objetivos foram extrair o óleo de sementes de sacha kiruma por meio de método químico (hexano), determinar o perfil de ácidos graxos e avaliar os efeitos desse óleo na estrutura do duodeno e fígado de camundongos Apo-E -/- e selvagens BlackC56/7. Os resultados da avaliação do óleo extraído para este estudo indica percentual de ácidos graxos similar ao citado na literatura. Entre os tratamentos não foram encontradas alterações no duodeno com relação à superfície de absorção, morfometria de cripta e espessura das camadas musculares e da mucosa. Com relação à altura do epitélio absortivo, os animais knockout do grupo controle apresentaram maior média se comparados com os grupos knockout tratados com o óleo. A freqüência de enterócitos, linfócitos, células caliciformes e células de Paneth não sofreram alterações devido ao tratamento proposto. Em relação às células caliciformes AB+PAS+, foi observada maior freqüência nos grupos selvagens tratados com óleo de sacha kiruma em relação aos controles. No fígado também não foram encontradas diferenças devido ao tratamento na frequência dos componentes hepáticos, nem na relação de diâmetro do citoplasma/ diâmetro do núcleo dos hepatócitos. Com isso, concluiu-se que o método de extração com hexano mostrou ser eficiente na extração de óleo das sementes de sacha kiruma. Este óleo apresentou composição lipídica similar daquela descrita na literatura, mantendo suas propriedades terapêuticas.
