Uso da dimetil-formamida associada ou não ao glicerol na criopreservação de sêmen caprino

Resumo: The objetive of this study was to evaluate the dimethyl formamide and its association effects on the goat semen cryopreservation, through the semen physical analyses and complementary tests, carried out during the cryopreservation process. Alpine and Saanen bucks from the Federal University of Viçosa Dairy Goat Experimental Section- DZO were used. The semen samplings were frozen with the yolk-skimmed milk diluent, associated with different cryoprotectors and concentration: glycerol 7%; (T1); glycerol 3.5% with dimethyl formamide 3.5% (T2) 5% dimethyl formamide5% (T3). The semen were centrifuged, diluted and bottled in a 0.5 mL straws. These straws were cooled at 5.0°C for more than 1:20h. Then they were submitted to liquid nitrogen vapor for 15 minutes and immersed in N2 liquid. The thawing was made in water bath at 37°C for 50 seconds. The parameters evaluated in vitro were progressive motility and sperm vigour, acrossome integrity, plasmatic membrane integrity (HOST) and acrossome reaction. A 30.0% initial motility loss was observed when the samples were submitted to the cryopreservation procedures. There was a 19% plasmatic membrane integrity loss, evaluated through the hipoosmotic swelling test (HOST). The acrossome lesions increased 3% during slow TTR (P<0.05). No differences were observed in the motility aspects among the sperm from the treatments, motility aspects at 0.00h; 0.05h; 1.00h of TTR and vigor in all the times given (P<0.05). At 2.00h of TTR a difference between the treatments (1) and (2) was observed (P<0.05), with the T2 semen motility superior to the others (P<0.05). The plasmatic membrane integrity values, acrossome integrity and acrossome reaction after the thawing did not differ between the semen from both treatments (P>0.05). These results show that the dimethyl formamide can be an alternative in the goat semen cryopreservation, when used alone or associated with glycerol.

Foram utilizados reprodutores das raças Alpina e Saanen pertencentes ao Setor de Caprinocultura- DZO-UFV. As partidas de sêmen foram congeladas com o diluente leite desnatado-gema, associado a diferentes crioprotetores e concentrações: 7% de glicerol (T1); 3,5% de glicerol e 3,5% de dimetil-formamida (T2) e 5% de dimetilformamida (T3). O sêmen foi centrifugado, diluído e envasado em palhetas de 0,5 mL. Estas palhetas foram resfriadas a 5,0°C, permanecendo nesta temperatura por mais 1:20h. Em seguida, foram submetidas ao vapor de nitrogênio líquido durante 15 minutos e imersas no N2 líquido. O descongelamento foi realizado em banho-maria, a 37°C por 50 segundos. Os parâmetros avaliados in vitro consistiram de motilidade progressiva e vigor espermático, integridade acrossômica, integridade da membrana plasmática (HO) e reação acrossômica. Foi observada uma perda de 30,0% da motilidade inicial, quando as amostras foram submetidas aos procedimentos de criopreservação. A perda de integridade da membrana plasmática, avaliada pelo teste hiposmótico (HO) foi de 19%. As lesões de acrossoma aumentaram em 3% durante o TTR lento (P<0,05). Não foi observada diferença nos aspectos de motilidade, entre os espermatozóides dos tratamentos, nos aspectos de motilidade, nos tempos de 0:00h, 0:05h, e 1:00h do TTR e vigor em todos os tempos (P>0,05). No tempo de 2:00h do TTR foi observada diferença entre os tratamentos (1) e (2) (P<0,05), sendo a motilidade do sêmen do T2 superior às demais. Os valores de integridade de membrana plasmática, integridade acrossômica e reação acrossômica pósdescongelamento não diferiram entre os sêmem dos tratamentos (P>0,05).
