Resumo: | In this work, separation of glycomacropeptide (GMP) was studied using liquid-liquid extraction with aqueous two-phase systems (ATPS) composed by poly (ethylene glycol) (PEG) and an inorganic salt. This technique is an advisable purification process applied to large scale since it provides a selective partition with high yields. The partition behavior of GMP in ATPS was investigated studying the influence of the type of salt (potassium phosphate, sodium citrate, lithium sulfate or sodium sulfate), polymer molar mass (PEG1500, PEG2000, and PEG4000), phase concentrations (tie line length), sodium chloride addition, pH value, and temperature in the system in order to fit ratio of system to the separation. The partition coefficient was defined as the ratio of GMP concentration in the upper phase (PEG) and in the bottom phase (salt). It was observed that the protein partitioned almost in the top PEG-rich phase, where the prevalent non- protein contaminants are PEG and a little amount of the salt, which can easily be removed from the target protein by means of size exclusion chromatography technique. Among the systems analysed and due to citrates characteristics as a biodegravel and nontoxic compound, the extraction conditions which provided the best results were: PEG1500 with concentration of PEG of 15.00 mass % and sodium citrate with concentration of 18.91 mass %; at pH 8.0 and temperature of 318.15 K. In this system were found values of partition coefficient above 30 and approximately 94.66 % of GMP was recovered in the top phase, what demonstrates a good separation. In addition, thermodynamic parameters (ΔH◦, ΔS◦, ΔG◦) as a function of temperature, were calculated for the system PEG1500-sodium citrate at different PEG concentrations and the results imply thermodynamic differences between partitioning of GMP in this system. This study clearly showed that ATPS can be successfully used for recovery and initial purification of GMP. |

This study clearly showed that ATPS can be successfully used for recovery and initial purification of GMP.Neste trabalho, estudou-se a aplicação da extração líquido-líquido para a separação do glicomacropeptídeo (GMP), utilizando sistemas aquosos bifásicos (SAB) compostos por polietileno glicol (PEG) e um sal inorgânico. Esta técnica pode ser usada para purificação de biocompostos em larga escala pela possibilidade de partição seletiva com altos rendimentos. O comportamento da partição do GMP em SAB foi verificado estudando a influência do tipo de sal (fosfato de potássio, citrato de sódio, sulfato de lítio e sulfato de sódio), massa molar do polímero (PEG1500, PEG2000 e PEG4000), adição de cloreto de sódio, valor de pH, concentração das fases e temperatura do sistema, a fim de se obter o melhor sistema para a separação da biomolécula. Nestes ensaios foi obtido o coeficiente de partição do GMP entre a fase aquosa superior (PEG) e a fase aquosa inferior (sal). Observou-se que a proteína migrou predominantemente para a fase superior, rica em PEG, onde os contaminantes não protéicos presentes na fase são o PEG e uma pequena quantidade de sal, que podem ser removidos da proteína alvo por meio de uma técnica cromatográfica, como a cromatografia de exclusão molecular (CEM). Entre os sistemas avaliados e em razão das características nãotóxicas e biodegradáveis do citrato, o sistema de extração que proporcionou as melhores condições foi o constituído por PEG1500 na concentração de 15 % em massa e citrato de sódio na concentração de 18,91 % em massa, em pH 8,0 e na temperatura de 318,15 K. Foram encontrados valores de coeficientes de partição para esse sistema acima de 30, sendo que aproximadamente 94,66 % do GMP foi recuperado na fase superior, o que indica uma boa separação. Em adição, parâmetros termodinâmicos (ΔH◦, ΔS◦, ΔG◦) em função da temperatura do sistema, foram calculados para o sistema formado por PEG1500 e citrato de sódio em diferentes concentrações de PEG. Os resultados exibiram diferenças termodinâmicas para a partição do GMP nos diversos sistemas avaliados.
