Resumo: The genus Brycon is the main representative of the subfamily Bryconinae and its species occur in basins to the West and East of the Andes. In Brazil, Bryconinae species are distributed in the main drainages; because they are affected by anthropogenic impacts, most of these species are threatened. This study is a hypothesis test of whether Bryconinae of the eastern Brazilian coast are a phylogeographic unit, using standard cytogenetic techniques (Giemsa, argyrophylic-nucleolar organizer region -Ag-NORsdetection, and C-banding) and mitochondrial molecular markers (cytochrome oxidase subunit I COI- and mitochondrial ribosomal 16S) on Brycon devillei, Brycon ferox, Brycon insignis, Brycon opalinus e Brycon vermelha. All these species were 2n=50, as all other Bryconinae. However, their karyotypic formulae were species-specific: B. devillei (26m+22sm+2st), B. ferox (28m+18sm+4st), B. insignis (22m+20sm+8st), B. opalinus (24m+20sm+6st) e B. vermelha (24m+20sm+6st). All species present in coastal basins of eastern Brazil showed NORs on the first pair of subtelocentric chromosomes and the first pair of chromosomes of the karyotype showed non-equilocal heterochromatin, indicating that all these species, with the monotypic genus Henochilus form a group reciprocally monophyletic in relation to the continental species. The 16S allowed to recover phylogenetic relationships among the oldest species Bryconinae, and COI gene was used to reconstruct the phylogeny of the species of eastern Brazil. The pattern obtained for both genes corroborate the hypothesis of monophyly of species Bryconinae of coastal basins of eastern Brazil. This condition probably was caused by the long history of isolation of the eastern coastal basins in relation to continental basins.

No Brasil, os briconíneos estão distribuídos nos principais sistemas hidrográficos, e por serem sensíveis às alterações negativas no ambiente de correntes das ações antrópicas, a maioria das espécies dessa subfamília estão ameaçadas de extinção. O objetivo deste trabalho foi testar a hipótese de existência de uma possível unidade filogeográfica dos Bryconinae que ocorrem nas bacias costeiras do leste brasileiro, uma região caracterizada pelo alto grau de endemismo, utilizando dados citogenéticos (coloração convencional, regiões organizadoras de nucléolos NORs e banda C) e moleculares mitocondriais (citocromo oxidase I e DNA ribossomal 16S). As espécies de Brycon devillei, Brycon ferox, Brycon insignis, Brycon opalinus e Brycon vermelha apresentaram número diploide 2n=50, semelhante às outras espécies de Bryconinae. Porém, suas fórmulas cariotípicas foram características de cada espécie: B. devillei (26m+22sm+2st), B. ferox (28m+18sm+4st), B. insignis (22m+20sm+8st), B. opalinus (24m+20sm+6st) e B. vermelha (24m+20sm+6st). Todas as espécies presentes nas bacias costeiras do leste do Brasil apresentaram NORs no primeiro par de cromossomos subtelocêntricos e o primeiro par de cromossomos do cariótipo apresentou padrão não equilocal de heterocromatina, indicando que todas essas espécies, junto ao gênero monotípico Henochilus, formam um grupo reciprocamente monofilético em relação às espécies continentais. O gene 16S permitiu recuperar as relações filogenéticas mais antigas entre as espécies de Bryconinae, e o gene COI foi utilizado na reconstrução da filogenia das espécies do leste brasileiro. O padrão obtido por ambos os genes corroboraram com a hipótese de monofiletismo das espécies de Bryconinae das bacias costeiras do leste do Brasil.
