Caracterização e avaliação do potencial antioxidante de mangas (Mangifera indica L.) cultivadas no Estado de Minas Gerais

Summary: Mango culture is an important economic activity both at national and international level. Mango agro-industry is also a growing sector in domestic and foreign markets, and producers expectation is of expanding the consuming market. Diffusion of information concerning mango nutritional and functional values, therefore, can add value to the fruit and byproducts, resulting in increased consumption and market competitiveness. Mango pulp has potential for becoming an excellent dietary antioxidant source, having in its composition molecules with biological antioxidant activities such as carotenoids, ascorbic acid and phenolic compounds. Also, mango agro-industrial residues, peel and seed, have the same phenolic compounds making them antioxidant sources for a range of uses. However, there is lack of works in the literature on the antioxidant potential of pulp and residues from mangos cultivated in Brazil. This work investigated the antioxidant potential of pulp of four commercial mango cultivars cultivated in Minas Gerais and the agro-industrial residues from cultivar Ubá. The study was carried out through the quantification of antioxidant compounds (ascorbic acid, total carotenoids, β-carotene, total phenolics), in vitro antioxidant activity tests, and biological assays using Wistar rats. Ripe fruits, characterized by pulp firmness, total soluble solids and color, were used for the assays. It was found that the levels of ascorbic acid in pulp were 15.69; 9.79; 10.54 and 77.71 mL per 100 g of pulp for varieties Haden, Tommy Atkins, Palmer and Ubá, respectively. There was no significant difference between varieties Haden and Tommy Atkins for ascorbic acid content, while the other varieties were statistically different for this parameter. Total carotenoids varied from 1.91 to 2.63 µg per 100 g, with no significant difference among varieties Tommy Atkins, Palmer and Ubá; variety Haden gave total carotenoids content significantly lower than the others. The β-carotene contents were significantly different among the four varieties, with values of 888; 608.39; 661.27 and 2,221 µg per 100 g for varieties Haden, Tommy Atkins, Palmer and Ubá, respectively. Total phenolics differed among the four varieties with values of 62.1; 48.4; 128.2 and 208.7 mg of gallic acid equivalent per 100 g of pulp for varieties Haden, Tommy Atkins, Palmer and Ubá, respectively. Zinc contents ranged from 0.249 to 0.33 mg per 100 g of pulp, with no significant differences among the four varieties. There were no significant differences among iron contents, ranging from 0.121 to 0.298 mg per 100 g of pulp. Copper contents ranged from 0.081 to 0.097 µg per 100 g of pulp, with no significant differences between Palmer and Ubá (0.083 and 0.081 µg per 100 g of pulp, respectively); Haden gave higher level (0.097 µg per 100 g of pulp) and Tommy Atkins gave significant lower level (0.077 µg per 100 g). All the four varieties showed antioxidant potential in the chemical tests, but there was significant difference among varieties, with variety Ubá being superior. There was difference regarding the contents of quercetin, mangiferin and kaempherol. Mangiferin was detected in pulp of varieties Ubá, Haden and Tommy Atkins; quercetin was only detected in varieties Ubá and Haden; and kaempherol only in variety Ubá. Biological assays showed that supplementation of commercial ration for wistar rats with lyophilized mango pulp at 10% resulted in increase of lipid peroxidation marker, evaluated by the thiobarbituric acid reactive substances (TBARS) test, both in the absence and the presence of oxidative stress induced by diclofenac, suggesting the occurrence of pro-oxidant effect. The biological assays with supplementation of AIN-G ration with lyophilized mango pulp at 3% showed reduction in lipid peroxidation, demonstrating the antioxidant effect, in the absence and the presence of oxidative stress induced by paracetamol. Total phenolics of agroindustry residues of variety Ubá were 48.95 mg gallic acid equivalent/kg DM (kilogram of dry matter) in skin and 68.45 mg gallic acid equivalent per kg DM in seed. Skin and seed contain mangiferin in the concentration of 270.5 mg kg-1 DM and 46.5 mg kg-1 DM, respectively. The skin contains quercetin at 785.3 mg kg-1 DM and kaempherol at 35.3 mg kg-1 DM. Hydroalcoholic extracts of residues showed antioxidant potential comparable to butylhydroxianisol (BHA) and gallic acid.

A agroindústria da manga é também um setor em crescimento nos mercados interno e externo, e a expectativa dos produtores é de ampliação do mercado consumidor. Para isso, a divulgação de informações relacionadas ao valor nutricional e funcional da manga pode agregar valor à fruta e aos seus produtos, resultando em aumento de consumo e de competitividade no mercado. A polpa de manga apresenta potencial para ser uma ótima fonte dietética de antioxidantes, por apresentar em sua composição carotenóides, ácido ascórbico e compostos fenólicos, os quais são moléculas com função antioxidante no meio biológico. Também os resíduos da agroindústria da manga, constituídos de casca e caroço, possuem compostos fenólicos que atribuem a eles a propriedade de serem uma fonte de antioxidantes, destinados a usos diversos. Entretanto, não foram identificados trabalhos na literatura enfocando o potencial antioxidante da polpa e de resíduos de mangas cultivadas no Brasil. Assim, foi proposto um estudo sobre o potencial antioxidante da polpa de quatro variedades de manga de valor comercial (Haden, Palmer, Tommy Atkins e Ubá), cultivadas em Minas Gerais, e do resíduo da agroindustrial da manga da variedade Ubá. O estudo foi desenvolvido com as mangas no estádio maduro, por meio da quantificação de constituintes antioxidantes (ácido ascórbico, carotenóides totais, β-caroteno, fenólicos totais), dos testes de atividade antioxidante in vitro e dos ensaios biológicos, utilizando-se ratos wistar. Utilizaram-se mangas no estádio maduro, sendo os frutos caracterizados em firmeza e a polpa em sólidos solúveis totais e cor. Verificou-se que os teores de ácido ascórbico da polpa das mangas foram de 15,69; 9,79; 10,54 e 77,71 miligramas por 100 gramas para as variedades Haden, Tommy Atkins, Palmer e Ubá, respectivamente. As mangas Haden e Tommy Atkins apresentaram teores de ácido ascórbico estatisticamente iguais; entre as demais variedades, houve diferença estatística no teor da vitamina. O teor de carotenóides totais variou de 1,91 a 2,63 microgramas por 100 gramas, e não houve diferença significante entre as variedades Tommy Atkins, Palmer e Ubá; a manga Haden apresentou teor de carotenóides totais estatisticamente menor do que o das demais. Os teores de β-caroteno diferiram estatisticamente entre as quatro variedades, sendo de 888,00; 608,39; 661,27 e 2.221,00 microgramas por 100 gramas para as variedades Haden, Tommy Atkins, Palmer e Ubá, respectivamente. Os teores de fenólicos totais diferiram entre as quatro variedades e foram de 62,10; 48,40; 128,20 e 208,70 miligramas de equivalentes de ácido gálico por 100 gramas para as mangas Haden, Tommy Atkins, Palmer e Ubá, respectivamente. Os teores de zinco variaram de 0,249 a 0,330 miligramas por 100 gramas de polpa e foram estatisticamente iguais entre as quatro variedades. Não houve diferença estatística entre os teores de ferro, que variaram de 0,121 a 0,298 miligramas por 100 gramas de polpa. Os teores de cobre variaram de 0,081 a 0,097 microgramas por 100 gramas, e Palmer e Ubá apresentaram teores estatisticamente iguais (0,083 e 0,081 microgramas por 100 gramas, respectivamente); Haden apresentou teor mais elevado (0,097 micrograma por 100 gramas); e Tommy Atkins, teor estatisticamente menor (0,077 micrograma por 100 gramas). Todas as quatro variedades apresentaram potencial antioxidante nos testes químicos, mas houve diferença significante entre as variedades, sendo a manga Ubá superior nos testes. Verificou-se diferença entre as variedades de mangas quanto ao teor de quercetina, mangiferina e kaempherol. A mangiferina foi detectada na polpa das variedades Ubá, Haden e Tommy Atkins; a quercetina, apenas nas variedades Ubá e Haden; e o Kaempherol, apenas na variedade Ubá. Nos ensaios biológicos, verificou-se que a suplementação da ração comercial de ratos wistar com polpa de manga liofilizada a 10% resultou em aumento do marcador de peroxidação de lipídio, avaliado pelo teste das substâncias reativas ao ácido tiobarbitúrico, tanto na ausência quanto na presença do estresse oxidativo induzido por diclofenaco, sugerindo a ocorrência do efeito prooxidante. Nos ensaios biológicos com a suplementação da ração AIN-G com polpa de manga liofilizada a 3%, observou-se redução da peroxidação de lipídios, evidenciando o efeito antioxidante, na presença e ausência do estresse oxidativo induzido por paracetamol. Os resíduos da agroindústria da manga Ubá apresentaram teor de fenólicos totais por quilo de matéria seca de 48,95 miligramas de equivalentes de ácido gálico na casca e de 68,45 miligramas de equivalentes de ácido gálico no caroço. A casca e a semente contêm mangiferina nas concentrações de 270,5 e 46,5 miligramas por quilo de material seco, respectivamente. A casca contém quercetina na quantidade de 785,3 miligramas por quilo de material seco e kaempherol na quantidade de 35,3 miligramas por quilo de material seco. Biological assays showed that supplementation of commercial ration for wistar rats with lyophilized mango pulp at 10% resulted in increase of lipid peroxidation marker, evaluated by the thiobarbituric acid reactive substances (TBARS) test, both in the absence and the presence of oxidative stress induced by diclofenac, suggesting the occurrence of pro-oxidant effect. The biological assays with supplementation of AIN-G ration with lyophilized mango pulp at 3% showed reduction in lipid peroxidation, demonstrating the antioxidant effect, in the absence and the presence of oxidative stress induced by paracetamol. Total phenolics of agroindustry residues of variety Ubá were 48.95 mg gallic acid equivalent/kg DM (kilogram of dry matter) in skin and 68.45 mg gallic acid equivalent per kg DM in seed. Skin and seed contain mangiferin in the concentration of 270.5 mg kg-1 DM and 46.5 mg kg-1 DM, respectively. The skin contains quercetin at 785.3 mg kg-1 DM and kaempherol at 35.3 mg kg-1 DM. Hydroalcoholic extracts of residues showed antioxidant potential comparable to butylhydroxianisol (BHA) and gallic acid.
