Resumo: | Cerebral ischemia is a reduction of blood flow in the brain, which causes hypoxia and consequent neuronal damage. It is the cause of the most common type of stroke, a disorder that can lead to death, or determine several sequels, mainly motor impairment of thoracic and pelvic members, in the contralateral hemisphere to the injured. Although physical activity is an preventive alternative and treatment for various cerebrovascular diseases, there is little information regarding the effects of exercise on skeletal muscle and motor performance of animals with cerebral ischemia. In this study, tests were performed for motor evaluation (foot fault test and parallel bars test), besides the histomorphometry of the biceps brachii, soleus and anterior tibial muscles of rats subjected to physical exercise on a treadmill before and after induction of cerebral ischemia by middle cerebral artery oclusion (MCAO). 36 Wistar rats were divided into six groups: CS: sedentary control animals (n = 6) who underwent a fake surgery (there were no MCAO); CEA: control animals exercised before the fake surgery (n = 6) ; CED: control animals trained after the fake surgery (n = 6); IS: sedentary ischemic animals (n = 6), IEA: exercised animals before induction of ischemia (n = 6) and IED: ischemic animals trained after induction of ischemia (n = 6). The exercise treadmill training consisted of six weeks, 30 minutes/day (five days/week) at a speed of 14 m/min for CEA and IEA groups, and 8m/min for CED and IED groups. Histomorphometry of skeletal muscles was performed, determining the frequency and area of muscle fibers type I and II as well as the density of muscle fibers with cytological alterations and component of interstitial of muscles. The sedentary-ischemic animals showed poorer performance on functional tests. Rats submitted to exercise after ischemia, showed significant recovery of motor performance. There was an increase in the frequency of type I fibers in the soleus muscle, both in sedentary-ischemic animals, as in the exercised animals before and after MCAO surgery. Type I fiber atrophy occurred in the biceps and anterior tibial muscles, and type I and II in the soleus of sedentary-ischemic animals, whereas in animals trained after ischemia, exercise attenuated muscle atrophy. Exercise promotes improvement in motor performance of ischemic animals, attenuates muscle atrophy caused by cerebral ischemia and can stimulate angiogenesis in skeletal muscle. |

É a causa do tipo mais frequente de acidente vascular encefálico, distúrbio que pode levar à morte, ou então determinar várias sequelas, principalmente prejuízo motor dos membros, torácico e pélvico, contralaterais ao hemisfério cerebral lesionado. Embora, a atividade física seja uma alternativa preventiva e terapêutica para diversas enfermidades cerebrovasculares, ainda existem poucas informações com relação aos efeitos do exercício sobre músculos esqueléticos e o desempenho motor de animais com isquemia cerebral. Neste trabalho, foram realizados testes de avaliação motora (passo em falso e barras paralelas), além da histomorfometria dos músculos bíceps braquial, sóleo e tibial anterior de ratos submetidos ao exercício físico em esteira, antes e após a indução da isquemia cerebral pela oclusão da artéria cerebral média (OACM). Foram utilizados 36 ratos Wistar distribuídos em seis grupos: CS: animais controle sedentários (n=6) que foram submetidos à falsa cirurgia (operação na qual não ocorreu a OACM); CEA: animais controle exercitados antes da falsa cirurgia (n=6); CED: animais controle exercitados após a falsa cirurgia (n=6); IS: animais isquemiados sedentários (n=6), IEA: animais exercitados antes da indução da isquemia (n=6) e IED: animais isquemiados exercitados após a indução da isquemia (n=6). O exercício físico consistiu no treinamento em esteira durante seis semanas, 30 minutos/dia (cinco dias/semana) a uma velocidade de 14 m/min para os grupos CEA e IEA, e de 8m/min para os grupos CED e IED. Realizou-se a histomorfometria dos músculos esqueléticos, determinando-se a frequência e a área de fibras musculares do tipo I e II, bem como a densidade de fibras musculares com alterações citológicas e de componentes do interstício muscular. Os animais isquemiados sedentários apresentaram pior desempenho nos testes funcionais. Ratos submetidos ao exercício, após a isquemia, apresentaram recuperação significativa do desempenho motor. Houve aumento na frequência de fibras I no músculo sóleo, tanto nos animais isquemiados sedentários, quanto nos animais exercitados antes e após a cirurgia de OACM. Ocorreu atrofia de fibras I no bíceps braquial e no tibial anterior, e do tipo I e II no sóleo dos animais isquemiados sedentários, sendo que nos animais treinados após a isquemia, o exercício atenuou a atrofia muscular.
