Introgressão de alelos de resistência aos patógenos da antracnose, mancha-angular e ferrugem em linhagens de feijão tipo carioca

Resumo: | Aiming to incorporate resistance to the pathogens anthracnose, angular spot, and rust in the bean lines BRSMG Talismã, VC 8 and VC 9, crosses were made between these lines and line Rudá-R (pyramided Rudá), donor of the genes Phg1, Co-4, Co-10 and Ur-ON, which confer resistance against Pseudocercospora griseola, Colletotrichum lindemuthianum and Uromyces appendiculatus. After the crosses were made, the backcross populations were obtained, and the RC1F1 plants were inoculated with the pathotype 65 of C. lindemuthianum. The lines shown to be resistant were genotyped with the markers SCARH13, SCARY20 and SCARF10, linked to the genes Phg1, Co-4 and to the genic bloc Co-10/Ur-ON, respectively. Based on the molecular data, 44 plants were selected and their seeds multiplied. The families originated from these plants were evaluated in three seasons (2007 dry and winter seasons and 2008 dry season) for grain yield, plant architecture and grain aspects. Based on these considerations and molecular data, 13 promising families were selected. From each of these families, 40 plants were inoculated with the mixture of the breeds 65 and 453 of C. lindemuthianum. The plants shown to be resistant were inoculated with the pathotype 31-17 of P. griseola. Ninety and five plants presented resistance to two pathogens and were genotyped with the markers SCARH13 (Phg1), SCARY20 (Co-4), SCARF10 (Co-10 /Ur-ON), SCARAS13 (Co-4) and SCARBA08 (Ur-ON). The resistant plants and carriers of the molecular marks of interest were selected. |

The resistant plants and carriers of the molecular marks of interest were selected.Visando incorporar resistência aos patógenos da antracnose, mancha-angular e ferrugem nas linhagens BRSMG Talismã, VC 8 e VC 9, foram realizados cruzamentos destas com a linhagem Rudá-R (Rudá piramidado), doadora dos genes Phg1, Co-4, Co-10 e Ur-ON, que conferem resistência à Pseudocercospora griseola, Colletotrichum lindemuthianum e Uromyces appendiculatus. Após a realização dos cruzamentos e obtenção das populações de retrocruzamento, as plantas RC1F1 foram inoculadas com o patótipo 65 de C. lindemuthianum. Aquelas que se mostraram resistentes foram genotipadas com os marcadores SCARH13, SCARY20 e SCARF10, ligados aos genes Phg1, Co-4 e ao bloco gênico Co-10/Ur-ON, respectivamente. Com base nos dados moleculares foram selecionadas 44 plantas, que tiveram suas sementes multiplicadas. As famílias provenientes destas plantas foram avaliadas em três safras (seca e inverno de 2007 e seca de 2008) quanto à produtividade de grãos, arquitetura de planta e aspecto dos grãos. Levando-se em conta estas avaliações e os dados moleculares, foram selecionadas 13 famílias que se mostraram promissoras. De cada uma dessas famílias foram inoculadas 40 plantas com a mistura das raças 65 e 453 de C. lindemuthianum. As plantas que se mostraram resistentes foram, em seguida, inoculadas com o patótipo 31-17 de P. griseola. Noventa e cinco plantas apresentaram resistência aos dois patógenos e foram genotipadas com os marcadores SCARH13 (Phg1), SCARY20 (Co-4), SCARF10 (Co-10 /Ur-ON), SCARAS13 (Co-4) e SCARBA08 (Ur-ON).
