Tratamento de feridas cutâneas experimentais em camundongos diabéticos com células-tronco mesenquimais associadas ou não ao plasma rico em plaquetas

Resumo: The effetiveness of treatment of experimental cutaneous wounds of diabetics mice (Mus musculus C57BL/6) with mesenchymal stem-cells (MSC), Platelet-rich Plasma (PRP) and MSC associated Io PRP was clinically evaluated. The diabetes mellitus (DM) was induced through the administration of streptozootocin diluted in buffered citrate of sodium. MSC were collected starting from the bone marrow of donors isogenic mice. healthy. expanded at laboratory and applied on the lesions of the appraised animals. PRP was obtained after processing of the total blood of donors isogenic mice, healthy, and applied on MSC in the bed lesional in the wound. Differences were obsewed (p <0,01) among the treated groups. for the variables area of the lesion (ARL), time of cicatrization (TEC) and collagen ñbers (FIC). The groups treated with isolated MSC and associated to PRP presented the smallest averages for the variables ARL and TEC when compared to the groups treated with PRP, physiologic solution and covering of the wounds wilh semi-permeable adherent membrane of poliuretane. Besides, the treatments with isolated or associated MSC and PRP they increased the percemage of collagens types I and III in the wound. The animals treated separately with PRP presented medium values for the variables ARL and TEC inferior lo the animals treated with physiologic solution and covering of the wounds with semir permeable adherent membrane of poliuretane. Similarly, the animals that just received scmi-permeable adhesive curative on the cutaneous wounds also presented medium values for the variables ARL and TEC smallest to the animals treated with physiologic Solution. However, differences were not observed (p>0,01) among these treatments for the increase of the percentage of collagens types I and III in the wound. The evaluation of the influence of the time for the complete cicatrization of the cutaneous wounds among the studied groups revealed a tendency more accentuated (p <0,01) for null values, which represent complete cicatrization of the lesion, among the animals of the groups treated with isolated MSC or associated to PRP, following by the groups just treated with PRP, semi-permeable adhcrent curative and group witness, respectively. These results allow to infer that the transplant of autogenous MSC is a more effective treatment for the cicatrization of cutaneous wounds of diabetic mice that the cutaneous application of PRP, daily asepsis of the cutaneous wounds with physiologic solution and covering of the lesions with semi-penneable adherent curative. However, the association between MSC and PRP does not accelerate the cicatrization of the cutaneous wounds of diabetic mice, when compared to the treatment with isolated MSC.

However, the association between MSC and PRP does not accelerate the cicatrization of the cutaneous wounds of diabetic mice, when compared to the treatment with isolated MSC.Avaliou-se clinicamente a eficácia do tratamento de feridas cutâneas experimentais de camundongos (Mus musculus CS7BL/6) diabéticos com células-tronco mesenquimais (CTM), plasma autólogo rico em plaquetas (PRP) e CTM associadas ao PRP. O quadro clínico de diabetes foi induzido por meio da administração de estreptozootocina diluída em tampão citrato de sódio. As CTM foram coletadas a partir da medula óssea de camundongos isogênicos doadores, saudáveis, expandidas em laboratório e aplicadas sobre as lesões dos animais avaliados. O PRP foi obtido após processamento do sangue total de camundongos isogênicos doadores, saudáveis, e aplicado juntamente com as CTM no leito lesional. Observaram-se diferenças (p<0,01) entre os grupos tratados, para as variáveis área da lesão (ARL), tempo de cicatrização (TEC) e fibras colágenas (FIC). Os grupos tratados com CTM isoladas e associadas ao PRP apresentaram as menores médias para as variáveis ARL e TEC quando comparado aos grupos tratados com PRP, solução fisiológica e cobertura das feridas com membrana aderente semi-permeável de poliuretano. Além disso, os tratamentos com CTM e PRP associados ou isolados aumentaram o percentual de colágenos tipos I e III no leito lesional. Os animais tratados isoladamente com PRP apresentaram valores médios para as variáveis ARL e TEC inferiores aos animais tratados com solução fisiológica e cobertura das feridas com membrana aderente semi-permeável de poliuretano. Analogamente, os animais que receberam apenas curativo adesivo semi- permeável sobre as feridas cutâneas também apresentaram valores médios para as variáveis ARL e TEC inferiores aos animais tratados com solução fisiológica. Não foram observadas diferenças (p>0,01) entre estes tratamentos para o aumento do percentual de colágenos tipos I e III no leito lesional. A avaliação da influência do tempo para a cicatrização completa das feridas cutâneas entre os grupos estudados revelou uma tendência mais acentuada (p<0,01) para valores nulos, os quais representam cicatrização completa da lesão, entre os animais dos grupos tratados com CTM isoladas ou associadas ao PRP, seguido dos grupos tratados apenas com PRP, curativo aderente semi-permeável e grupo testemunha, respectivamente. Estes resultados denotam que o transplante de CTM autólogas é um tratamento mais eficaz para a cicatrização de feridas cutâneas de camundongos diabéticos que a aplicação cutânea de PRP, assepsia diária das feridas cutâneas com solução fisiológica e cobertura das lesões com curativo aderente semi-permeável.
