Efeito do zinco isolado ou associado à vitamina E sobre testiculos de ratos Wistar adultos

Resumo: Antioxidants are molecules that can slow or prevent the oxidation of other substances. Effects have been described associations of different micronutrients that leverage in certain tissues or organs, leading to health benefits. Zinc and vitamin E (α-tocopherol) is considered an important antioxidant in cell membranes, and both have benefits in reproductive isolation, seen in studies of semen. The precise actions of zinc only or α-tocopherol as antioxidants on parameters of fertility are still unclear and they have not been evaluated when associated in the testis. The objective of this study was to evaluate the effect of zinc alone or associated with α-tocopherol (α-Toc) on the characteristics of components stereological testicular adult Wistar rats. We used 56 Wistar rats of reproductive age, divided into seven groups of eight animals. Six groups received Zn in drinking water. Three groups received 5, 10 and 20 mg Zn, while three other groups received the same dosage of Zn associated with 15 mg of α-Toc by gavage in three weekly doses. A control group of eight animals received only water. The diet was ad libitum during the 56 days of treatment. The first three groups and the control received 0.9% NaCl solution by gavage to standardize the procedures among the treatments. Morphometric analysis of the tubular and intertubular compartment was carried out using Image Pro Plus software. The averages were compared using the SNK test (Student Newman Keuls) with 5% tolerance. In seminiferous tubule, the treatment with 10 mg Zn increased in spermatocytes preleptotene / leptotene, round spermatids and the total number of germ cells. The association of Zn with 20 mg of α-Toc increased mitotic index. In the intertubular, mice treated with 5 mg of Zn showed an increase in the number of Leydig cells and macrophages and mice treated com10mg Zn associated with α-Toc had increased lymphatic space, the number of blood vessels and intertubular space. It is concluded that Zn acted alone, preferably in the cells, both in the tubule, and in the intertubular. The synergistic action of two antioxidants (Zn + Toc) was shown by the increase in intertubular space, lymphatic and blood vessels and also with increased mitotic index.

Já foram descritos efeitos de associações de diferentes micronutrientes que se potencializam em determinados tecidos ou órgãos, gerando benefícios à saúde. O zinco assim como a vitamina E (α-tocoferol) é considerado um antioxidante importante nas membranas celulares e ambos isoladamente apresentam benefícios na reprodução, vistos em estudos com sêmen. As ações precisas apenas do zinco ou do α-tocoferol, como antioxidantes sobre parâmetros da fertilidade, ainda não são claras e ambos ainda não foram avaliados quando associados no testículo. O objetivo do trabalho foi avaliar o efeito do zinco isolado ou associado à α-tocoferol (α-Toc) sobre as características morfométricas de componentes testiculares de ratos Wistar adultos. Foram utilizados 56 ratos Wistar em idade reprodutiva, divididos em sete grupos de oito animais. Seis grupos receberam Zn, na água de beber. Três grupos receberam 5, 10 e 20 mg de Zn, enquanto outros três grupos receberam as mesmas dosagens de Zn associadas a 15 mg de α-Toc por gavagem, em três doses semanais. Um grupo controle de 8 animais recebeu somente água. O regime alimentar foi ad libitum durante os 56 dias de tratamento. Os três primeiros grupos e o controle receberam solução de NaCl 0,9% por gavagem, para homogeneizar os procedimentos entre os tratamentos. Análises morfométricas do compartimento tubular e intertubular foram realizadas utilizando o software Image Pro Plus. Foram comparadas as médias utilizando-se o teste SNK (Student Newman Keuls) com 5% de tolerância. No túbulo, o tratamento com 10mg de Zn aumentou os espermatócitos em preleptóteno/leptóteno, espermátides arredondadas e o número total de células germinativas. A associação de 20mg de Zn com α-Toc aumentou o índice mitótico. No intertúbulo, os ratos tratados com 5mg de Zn apresentaram aumento no número de células de Leydig e macrófagos e os ratos tratados com10mg de Zn associado a α-Toc tiveram aumento do espaço linfático, do número de vasos sanguíneos e do espaço intertubular. Conclui-se que o Zn agiu isolado, preferencialmente nas células, tanto no túbulo, quanto no intertúbulo.
