Análise da hipótese da vicariância entre Dalbergia nigra Allem. ex Benth. e Dalbergia miscolobium Benth. (Fabaceae) com base em DNA cloroplastídico

Resumo: | The genus Dalbergia L. f. (Fabaceae: Papilionoideae) includes approximately 100 pantropical species. In Brazil there are 40 Dalbergia species distributed in different Brazilian ecossistems. D. nigra and D. miscolobium which occur in the Mata Atlântica and Cerrado regions, respectively, are considered to be vicariant. Vicariance between these two species has been investigated by phytosociological and chymosystematic studies, however, new approaches need to be used for these studies as this hypothesis has not been confirmed and is not fully accepted by the scientific community. In this work the chloroplast trnL intron sequences of D. nigra and D. miscolobium have been analyzed as a tool to distinguish these two species and possibly aid the discussion of vicariance between them. Leaf DNA samples was extracted, amplified and sequenced with specific primers for the trnL-c and trnL-d regions of the trnL gene present in the chloroplast DNA. The sequences obtained were aligned generating a matrix with 530 characters. Seventy-five variable sites were obtained, 37 of them were informative for the parsimony analysis. As a whole the maximum parsimony nd the neighbor-joining analyses were consistent, giving rise to trees with similar topologies, separating the different taxa into clades. The phylogenetic relationships between the species studied based on molecular data support those based on morphological data presented in the literature, however, they do not confirm the hypothesis of vicariance between D. nigra and D. miscolobium. |

(Fabaceae: Papilionoideae), possui cerca de 100 espécies de distribuição pantropical. No Brasil, há 40 espécies de Dalbergia distribuídas em áreas representativas dos diferentes ecossistemas brasileiros. D. nigra e D. miscolobium são espécies consideradas vicariantes, que ocorrem na Mata Atlântica e no Cerrado, respectivamente. Estas espécies figuram como objetos de estudo sobre vicariância em trabalhos anteriores de fitossociologia e quimiossistemática, porém, estes estudos necessitam de outras abordagens, visto ser a teoria de vicariância ainda polêmica entre pesquisadores. Neste trabalho, objetivou-se analisar as seqüências do íntron trnL em D. nigra e D. miscolobium, visando investigar a validade dessa região genômica para distinguir essas espécies e discutir possível vicariância entre elas. Amostras de DNA de plantas das espécies de Dalbergia foram extraídas e amplificadas com primers específicos trnL-c e trnL-d (cpDNA). As seqüências de DNA alinhadas produziram uma matriz com 530 caracteres. Excluindo os autopomórficos e os invariáveis, foram obtidos 75 sítios variáveis dos quais 37 foram informativos para análise de parcimônia. De modo geral, as análises de máxima parcimônia e neighbor-joining foram consistentes, resultando em árvores de topologias semelhantes entre si, com a separação dos taxa em clados.
