Componentes químicos e atividade biológica do Veneno de Pachycondyla striata F. Smith (Formicidae: Ponerinae)

Resumo: The venom produced by ants has been reported as a rich source of bioactive molecules with possible pharmacological importance. The reservoir of the venom gland of Pachycondyla striata was chemically analyzed with two-dimensional electrophoresis, HPLC and GC, followed by mass spectrometry analyses of the type Maldi-TOF-TOF and ESI-TOF-TOF and "de novo" sequencing assisted by software. The venom was composed primarily for proteins, peptides and hydrocarbons. Thirty-eight proteins/peptides and 48 non-protein compounds were found in the venom of P. striata. Whithin the identified proteins there are classic proteins from venom (phospholipase A, hyaluronidase and aminopeptidase N), allergenic proteins (allergens of venom) and peptides with biological activity (buforin, dinoponeratoxin Da-1837, U10-ctenitoxin Pn1a and sarafotoxin). Mainly long chain hydrocarbons (C19 - C36) were found among non-protein compounds. The antimicrobial activity of the venom was tested from venom purified by reversed-phase and ion exchange HPLC. Five fractions derived from the fractionation by reverse phase showed high antimicrobial activity against gram- positive and gram negative bacteria, but only analgesine 2, with reported antimicrobial activity, was identified in one of the fractions. To assess the neurobiological and anticonvulsant activity of the venom, the crude and denatured venom of P. striata in different concentrations (25, 100 and 250 mg / mL) was injected intracerebroventricularly in the mice and the behavior of the mice were evaluated. Significant differences were found in the immobility and exploration behaviors of the mice treated with crude venom concentration of 250 mg/mL. The venom of P. striata was ineffective as an anticonvulsant agent.

O reservatório da glândula de veneno de Pachycondyla striata foi analisado quimicamente usando técnicas de eletroforese bidimensional, HPLC e GC, seguido de análises de espectrometria de massas do tipo Maldi-TOF-TOF e ESI-TOF-TOF e técnica de sequenciamento de novo assistida por software. O veneno de P. striata foi composto principalmente por proteínas, peptídeos e hidrocarbonetos. Trinta e oito proteínas/peptídeos e 48 compostos não-proteicos, foram encontrados compondo o veneno de P. striata. Entre as proteínas identificadas estão proteínas clássicas de veneno (fosfolipase A, hialuronidase e aminopeptidase N), proteínas alergênicas (diferentes alérgenos de veneno) e peptídeos com atividade biológica comprovada (buforina, dinoponeratoxina Da- 1837, U10- ctenitoxina Pn1a e sarafotoxina). Entre os compostos não-proteicos, estão principalmente os hidrocarbonetos de cadeia longa (C19-C36). A atividade antimicrobiana do veneno foi testada e para isso o veneno foi purificado em HPLC por fase-reversa e por troca iônica. Cinco frações oriundas do fracionamento por fase reversa apresentaram elevada atividade antimicrobiana contra bactérias gram-positivas e gram-negativas, mas apenas a proteína analgesina 2, com atividade antimicrobiana comprovada, foi identificada em uma das frações. Para avaliar a atividade neurobiológica e anticonvulsivante do veneno, o veneno bruto e desnaturado de P. striata em diferentes concentrações (25, 100 e 250 mg/mL) foi injetado intracerebroventricular em camundongos e o comportamento dos camundongos avaliado. Diferenças significativas foram observadas nos comportamentos de imobilidade e exploração dos camundongos tratados com veneno bruto na concentração de 250 mg/mL.
