Micropropagação em biorreatores de imersão temporária e enraizamento de miniestacas e microestacas de clones híbridos de Eucalyptus globulus

Resumo: | This study aimed to evaluate the micropropagation through temporary immersion bioreactors RITA ® multiplication and elongation in vitro, as well as mini-cuttings and microcutting the rooting of clones Eucalyptus urophylla x E. globulus and Eucalyptus grandis x E. globulus. In the multiplication phase and stretching evaluated the cultivation liquid system (bioreactor RITA ® ) and semisolid (plastic bottles and test tubes), and the ratios between the different sources of nitrogen (nitrate / ammonium) on growth and productivity explants. The influence of propagation techniques, ( micro and mini-cuttings), different types of cuttings and substrates (with and without apical and middle leaf reduction) in the rooting of clones hybrid Eucalyptus globulus clones. It was found that the bioreactor of temporary immersion RITA ® and plastic pots showed the best results in the growth of explants. There was no difference between the reasons for nitrate nitrogen (NO3-) and ammonium (NH4 +) (1:1, 2:1 and 3:1) used in the multiplication phase and elongation, except for clone C16 in the phase of multiplication obtained greater growth and productivity in the ratio 2:1 (NO3-): (NH4 +). Overall cultures had a high degree of hyper-hydro, which is a disorder limiting factor in this study conditions for micropropagation in bioreactor of temporary immersion RITA ®. There was no significant superiority of microcuttings regarding mini-cuttings and was no effect of different substrates for only a few features. In overall, the of apical cuttings (without reduction and reduced leaf) was superiority in relation the intermediate. |

In overall, the of apical cuttings (without reduction and reduced leaf) was superiority in relation the intermediate.O presente trabalho teve como objetivos avaliar a micropropagação através do uso de biorreatores de imersão temporária RITA® nas fases de multiplicação e alongamento in vitro, bem como a miniestaquia e a microestaquia no enraizamento de clones Eucalyptus urophylla x E. globulus e Eucalyptus grandis x E. globulus. Na fase de multiplicação e alongamento avaliou-se o cultivo em sistema líquido (biorreator RITA®) e semissólido (potes plásticos e tubos de ensaio), além das diferentes razões entre as fontes de nitrogênio (nitrato/amônio) no crescimento e produtividade dos explantes. Foram verificados também a influência das técnicas de propagação (miniestaquia e microestaquia), diferentes substratos e tipos de estacas (apical com e sem redução foliar e intermediária) no enraizamento de quatro clones de Eucalyptus globulus. Constatou-se que o biorreator de imersão temporária RITA® e os potes plásticos apresentaram os melhores resultados no crescimento dos explantes. Não houve diferença entre as razões de nitrogênio nitrato (NO3 -) e amônio (NH4 +) (1:1, 2:1 e 3:1) utilizadas na fase de multiplicação e alongamento, exceto para o clone C16, que, na fase de multiplicação, obteve maior crescimento e produtividade na razão 2:1(NO3 - ):(NH4 +). De forma geral, as culturas apresentaram alto grau de hiper-hidricidade, sendo esta desordem um fator limitante nas condições deste estudo para a micropropagação em biorreator de imersão temporária RITA®. Não foi observada superioridade expressiva das microestacas em relação às miniestacas e houve efeito dos diferentes substratos apenas para algumas características.
