Avaliação da atividade biológica e obtenção de lipossomos contendo extrato de flores de Senna macranthera

Resumo: Plants and natural products of several different origins have been utilized for different goals. Among the most utilized medical plants, just a few have efficiency proved and tested. Among the plant types used in the popular medicine and found in the Brazilian pharmacopeia, the genus Senna is of great importance, with different activities that range from laxative to antibacterial activity. Within this genus, the species Senna macranthera (sicklepod) a tree frequently found in the woods but very used as ornamental tree in urban forestry. The flowering season are from April to June, when it becomes covered with large bright-yellow flowers The present study aimed to evaluate the antimicrobial, antioxidant and antitumor activity from the crude extracts hexane, dichloromethane ethyl acetate and ethanol obtained of the flower S. macranthera, the incorporation of the active crude extract in liposomes, and the isolation and structural identification of anthraquinones. The antibacterial activity against strains isolated from animals with bovine mastitis manifestation was positive for polar crude extracts ethyl acetate and ethanol. The crude extract ethyl acetate showed the lowest minimum inhibitory concentration for S. aureus and S. agalactiae strains. The crude extracts ethyl acetate and ethanol showed significant antioxidant activity when compared to standard compound BHT a 5 % probability with antioxidant activity values above 88%. The antitumor test, in turn, did not show good activity due to the necessity of using high concentrations of the crude extract to cause damage to cytotoxic cells. Through the technique of dynamic light scattering, we verified the formation of liposomes, suggesting the incorporation of the crude extract ethyl acetate because of a small variation in diameter (452 nm without the extract and 543 nm with it) and a decrease in light intensity with extract in a value equal to 60% of the value without the extract. The phytochemical screening of the crude extracts suggested the presence of aglucones and anthraquinones glycosides, in the polar crude extracts. The isolation and identification of chrysophanol in crude extract ethyl acetate corroborated with the phytochemical screening for the presence of anthraquinones compounds.

Entre as plantas medicinais mais utilizadas pela população, poucas têm ação comprovada. Dentre os gêneros de planta empregada na medicina popular e encontrada na farmacopéia brasileira, o gênero Senna tem grande importância, com diferentes atividades que vão desde o uso como laxante ao de atividade antibacteriana. Dentro deste gênero, a espécie Senna macranthera (fedegoso) é ornamental quando em flor e considerada própria para utilização em arborização urbana. O presente estudo teve como objetivo a avaliação das atividades antimicrobiana, antioxidante e antitumoral dos extratos obtidos da flor de Senna macranthera, a incorporação do extrato ativo em lipossomos, e o isolamento e identificação estrutural do mesmo. A atividade antibacteriana de isolados de mastite bovina foi positiva para os extratos polares, com formação de halo de inibição, sendo que o extrato de acetato de etila apresentou a menor concentração inibitória mínima para as cepas de S. aureus e S. agalactiae. Os extratos de acetato de etila e de etanol apresentaram atividade antioxidante significativa quando comparados ao padrão sintético BHT a 5% de probabilidade com valores de ação antioxidante acima de 88%. O teste antitumoral, por sua vez, não apresentou boa atividade devido à necessidade de se utilizar grandes concentrações do extrato para causar danos às células citotóxicas. Através da técnica de espalhamento dinâmico de luz verificou-se a formação de lipossomos, sugerindo a incorporação do extrato de acetato de etila devido uma pequena variação do diâmetro (452 nm sem o extrato e 543 nm com) e uma diminuição na intensidade de luz com extrato para um valor igual a 60% do valor sem extrato. A prospecção fitoquímica dos extratos polares sugeriu a presença de geninas e heterosídeos antraquinônicos, nos extratos mais polares.
