Análise comparativa do genoma de dois isolados de Cowpea aphid-borne mosaic virus (CABMV) obtidos de diferentes hospedeiros

Resumo: In Brazil and in other countries where passionfruit is grown, the viral disease known as passionfruit woodiness is a limiting factor for production. For many years this disease was associated with a single potyvirus, Passion fruit woodiness virus (PWV). However, studies conducted over the last 20 years demonstrated that two additional potyviruses, Cowpea aphidborne mosaic virus (CABMV) and East Asian Passiflora virus (EAPV) can also cause passionfruit woodiness. Plants infected by either one of these three viruses display mosaic in the leaves and woodiness of the pericarp of the fruit. In Brazil, molecular analysis has shown that several potyvirus isolates previously classified as PWV based on biological and serological properties are actually CABMV isolates. These isolates are capable of infecting legume crops such as common bean (Phaseolus vulgaris) and cowpea (Vigna unguiculata). To this date, only one CABMV isolate (CABMV- Zimbabwe, obtained from cowpea) has been completely sequenced. For some Brazilian isolates, the capsid protein (CP) coding region and the 3 -untranslated region have been sequenced. Sequence analysis did not indicate any features that could be correlated with biological differences among the isolates. The objective of this work was to determined the complete genomic sequence of two CABMV isolates from Brazil, one obtained from passionfruit with woodiness symptoms (isolate MGAvr, which also infects bean and cowpea), and one obtained from peanut (isolate BR1, which also infects bean and cowpea, but does not infect passionfruit). Both isolates were maintained in cowpea plants of the cultivar Pitiúba. Viral RNA was purified from concentrated virions and used as a template for cDNA synthesis, which was then used as a template for long range PCR using a high fidelity DNA polymerase and different combinations of degenerate and specific primers. The nucleotide sequences of isolates BR1 and MG-Avr are 95% identical and display 87 and 86% identity with CABMV-Z, respectively. Both isolates have the typical potyvirus genome features, including the potential to encode a polyprotein which can generate 8 to 10 putative viral proteins by autoproteolysis. Among these putative viral proteins, P3 and VPg are the most conserved between the two isolates, with 99% amino acid sequence identity, and the CP is the least conserved, with 85% identity. Phylogenetic analysis indicates that the two Brazilian isolates group with CABMV-Z in the legume-infecting species cluster, and in the Bean common mosaic virus (BCMV) subgroup. Mutagenesis studies and phenotypic analysis could identify genomic regions or amino acids involved in the biological differences between the two isolates.

Durante muitos anos esta virose esteve associada a uma única espécie de potyvirus, o Passion fruit woodiness virus (PWV). No entanto, estudos conduzidos ao longo dos últimos 20 anos demonstraram que duas outras espécies de potyvírus, o Cowpea aphid-borne mosaic virus (CABMV) e o East Asian Passiflora virus (EAPV), também podem causar a mesma doença. As plantas infectadas apresentam mosaico e endurecimento do pericarpo dos frutos. No Brasil, foi demonstrado recentemente, através de caracterização molecular, que isolados de potyvírus causando endurecimento dos frutos anteriormente classificados como PWV com base em características biológicas e sorologia são na verdade isolados de CABMV. Os diversos isolados analisados foram capazes de infectar espécies de leguminosas como o feijoeiro comum (Phaseolus vulgaris) e o caupi (Vigna unguiculata). Até o momento, somente um isolado de CABMV (CABMV-Zimbábue, obtido de caupi) teve seu genoma completamente sequenciado. Para alguns isolados brasileiros, a seqüência correspondente à região codificadora da proteína capsidial e a região 3 não traduzida foram determinadas. A análise destas sequências não foi capaz de fornecer informações capazes de explicar as diferenças biológicas observadas entre os isolados. O objetivo deste trabalho foi obter a sequência de nucleotídeos completa de dois isolados de CABMV do Brasil, um deles provenientes de plantas de maracujá-amarelo apresentando sintomas de endurecimento dos frutos (isolado MG-Avr, que também infecta feijão e caupi), e outro proveniente de amendoim (isolado BR1, também infecta feijão e caupi porém não infecta maracujá). Os isolados virais foram mantidos e multiplicados em plantas de caupi cv. Pitiúba. O RNA viral foi purificado a partir de preparações virais concentradas e utilizado como molde para a síntese de cDNAs, que por sua vez foram utilizados como molde para reações de PCR longo utilizando-se uma DNA polimerase com atividade de correção de erro e diferentes combinações de oligonucleotídeos degenerados e específicos. As sequências de nucleotídeos dos isolados BR1 e MG-Avr são 95% idênticas e apresentam 87 e 86% de identidade com o CABMV-Z, respectivamente. Ambos os isolados apresentam todas as características típicas de potyvírus, incluindo o potencial de codificar uma poliproteína que sofre autoproteólise gerando de 8 a 10 proteínas virais. Dessas proteínas, P3 e VPg são as mais conservadas entre os dois isolados, com 99% de identidade para a seqüência de aminoácidos, e a CP é a menos conservada, com 85% de identidade. A análise filogenética das seqüências completas de nucleotídeos indicou que os dois isolados brasileiros se agruparam juntamente com o isolado Z, no grupo de espécies de potyvírus que infectam leguminosas, no subgrupo do Bean common mosaic virus (BCMV).
